Huntingtin is required for ER-to-Golgi transport and for secretory vesicle fusion at the plasma membrane.

Huntingtin is a large membrane-associated scaffolding protein that associates with endocytic and exocytic vesicles and modulates their trafficking along cytoskeletal tracks. Although the progression of Huntington's disease is linked to toxic accumulation of mutant huntingtin protein, loss of wild-type huntingtin function might also contribute to neuronal cell death, but its precise function is not well understood. Therefore, we investigated the molecular role of huntingtin in exocytosis and observed that huntingtin knockdown in HeLa cells causes a delay in endoplasmic reticulum (ER)-to-Golgi t

SH3P7/mAbp1 deficiency leads to tissue and behavioral abnormalities and impaired vesicle transport.

The intracellular adaptor protein SH3P7 is the mammalian ortholog of yeast actin-binding protein 1 and thus alternatively named as mAbp1 (or HIP55). Structural properties, biochemical analysis of its interaction partners and siRNA studies implicated mAbp1 as an accessory protein in clathrin-mediated endocytosis (CME). Here, we describe the generation and characterization of mice deficient for SH3P7/mAbp1 owing to targeted gene disruption in embryonic stem cells. Mutant animals are viable and fertile without obvious deficits during the first weeks of life. Abnormal structure and function of org

Characterization of the Saccharomyces cerevisiae cytosine transporter using energizable plasma membrane vesicles.

The purine-cytosine permease is a carrier localized in the plasma membrane of the yeast Saccharomyces cerevisiae. The energetics of cytosine transport catalyzed by this permease has been studied in an artificial system obtained by fusion between proteoliposomes containing beef heart cytochrome c oxidase and plasma membrane-enriched fractions of a S. cerevisiae strain overexpressing the permease. Upon addition of an energy donor, a proton-motive force (inside alkaline and negative) is created in this system and promotes cytosine accumulation. By using different phospholipids, it is shown that c

Salicylate inhibits the renal transport of inorganic sulfate in rat membrane vesicle preparations.

Salicylic acid (SA) administration produces a pronounced enhancement of the renal clearance of inorganic sulfate in rats. The purpose of the present investigation was to determine if SA inhibits the renal transport of inorganic sulfate in rat kidney cortex brush border membrane (BBM) and basolateral membrane (BLM) vesicle preparations. Sodium-dependent cotransport of inorganic sulfate was examined in BBM, and the values for KM and Vmax, determined using nonlinear regression analysis, were 0.52 +/- 0.41 mM and 2.84 +/- 1.26 nmol/mg protein/10 sec, respectively (N = 7). Bicarbonate-dependent sul

Huntingtin differentially regulates the axonal transport of a sub-set of Rab-containing vesicles in vivo.

Loss of huntingtin (HTT), the Huntington's disease (HD) protein, was previously shown to cause axonal transport defects. Within axons, HTT can associate with kinesin-1 and dynein motors either directly or via accessory proteins for bi-directional movement. However, the composition of the vesicle-motor complex that contains HTT during axonal transport is unknown. Here we analyze the in vivo movement of 16 Rab GTPases within Drosophila larval axons and show that HTT differentially influences the movement of a particular sub-set of these Rab-containing vesicles. While reduction of HTT perturbed t

Outer membrane vesicles mediate transport of biologically active Vibrio cholerae cytolysin (VCC) from V. cholerae strains.

BACKGROUND: Outer membrane vesicles (OMVs) released from Gram-negative bacteria can serve as vehicles for the translocation of virulence factors. Vibrio cholerae produce OMVs but their putative role in translocation of effectors involved in pathogenesis has not been well elucidated. The V. cholerae cytolysin (VCC), is a pore-forming toxin that lyses target eukaryotic cells by forming transmembrane oligomeric β-barrel channels. It is considered a potent toxin that contributes to V. cholerae pathogenesis. The mechanisms involved in the secretion and delivery of the VCC have not been extensively

Effects of temperature, pH elevators, and energy production inhibitors on horseradish peroxidase transport through endocytic vesicles.

We investigated the effects of reduced temperature, the pH elevators NH4Cl, monensin, and HEPES (N-2-hydroxy-ethylpiperazine-N'-2-ethanesulfonic acid) buffer, as well as the metabolic poisons NaF/KCN on transport of the fluid phase pinocytic marker, horseradish peroxidase (HRP), to lysosomes in Chinese hamster ovary (CHO) cells. In cell fractionation experiments, these agents appeared to block HRP transit at specific point(s) from "early" to "late" (i.e., low to high density) prelysosomal vesicles and lysosomes. Reduced temperature (17 degrees C) most strongly inhibited HRP transport from low

Lipid peroxidation and active calcium transport in inside-out vesicles of red blood cells from preeclamptic women.

We previously reported that in preeclampsia Ca-ATPase activity diminishes about 50% in red blood cells, myometrium and syncitiotrophoblast plasma membranes. In this work, we measured the active Ca++ uptake by inside-out vesicles of human red blood cells from preeclamptic and normotensive pregnant women. Active calcium uptake by the vesicles was diminished by 49+/-3% in the preeclamptic women as compared to the gestational controls ( 8.06 +/- 0.11 nmol Ca++/mg protein min, gestational controls; 4.08 +/- 0.1 nmol Ca++/mg protein min, preeclamptics). This lowered calcium uptake correlates well wi

Reversal of vinblastine transport by chlorpromazine in membrane vesicles from multidrug-resistant human CCRF-CEM leukaemia cells.

The mechanism of action of 2-chlorpromazine (2-chloro-10-(3-dimethylaminopropyl)-phenothiazine) as a reversal agent for P-glycoprotein-mediated multidrug resistance was investigated using inside out-orientated membrane vesicles prepared from vinblastine-resistant human CCRF-CEM leukaemia cells (VBL1000). 2-Chlorpromazine (10 microM) completely inhibited ATP-dependent P-glycoprotein-mediated vinblastine accumulation in the vesicles. Whereas in the absence of added ligands VBL transport was described by a hyperbolic function of vinblastine concentration, in the presence of 2-chlorpromazine vinbl

Integrative Proteomic and Phosphoproteomic Analysis Reveals Altered Vesicle Transport in Systemic Lupus Erythematosus.

BACKGROUND: Vesicle transport genes (VTGs) are involved in the pathogenesis and progression of systemic lupus erythematosus (SLE). A comprehensive multi-omics analysis is crucial to elucidate their molecular alterations and identify potential biomarkers and therapeutic targets. However, studies investigating global alterations of VTGs in SLE remain limited. In this study, we aimed to investigate the relationship between VTGs alterations and SLE progression. METHODS: We integrated proteomic and phosphoproteomic data from 130 SLE patients and 90 healthy controls (HC). This was combined with tra

Cell polarity: cell type-specific regulators, common pathways, and polarized vesicle transport.

Cell polarity, the asymmetric organization of cellular components, is evolutionarily conserved from unicellular and multicellular organisms and is crucial for many biological processes. Polarity is required to maintain cell and tissue integrity by regulating cell division, migration, orientation, cell-cell interactions, and morphogenesis. Impaired polarity leads to dysregulation of cellular functions and is associated with disease. Understanding how polarity is established, maintained, and regulated is thus critical to improving our knowledge of pathologies and devising novel therapies. Here,

Human mesenchymal stem/stromal cell-derived extracellular vesicle transport in meniscus fibrocartilage.

Extracellular vesicles (EVs) derived from endometrial-derived mesenchymal stem/stromal cells (eMSC) play a crucial role in tissue repair due to their immunomodulatory and reparative properties. Given these properties, eMSC EVs may offer potential benefits for meniscal repair. The meniscus, being partly vascularized, relies on diffusivity for solute trafficking. This study focuses on EVs transport properties characterization within fibrocartilage that remains unknown. Specifically, EVs were isolated from Crude and CD146+ eMSC populations. Green fluorescence-labeled EVs transport properties were

Two distinct mechanisms for bilirubin glucuronide transport by rat bile canalicular membrane vesicles. Demonstration of defective ATP-dependent transport in rats (TR-) with inherited conjugated hyperbilirubinemia.

Bilirubin is conjugated with glucuronic acid in hepatocytes and subsequently secreted in bile. The major conjugate is bilirubin diglucuronide. Using sealed vesicles which are primarily derived from the canalicular (CMV) and sinusoidal (SMV) membrane vesicle domains of the plasma membrane of hepatocytes, we demonstrated that bilirubin glucuronides are transported by CMV by both ATP- and membrane potential-dependent transport systems. In CMV from normal rats, these processes are additive. In CMV from TR- rats, which have an autosomal recessively inherited defect in biliary secretion of nonbile a

Machine Learning Identifies TRAPPC13/COPS5 as Biomarkers and Vesicle Transport Subtypes in Parkinson's Disease.

BACKGROUND: Parkinson's disease (PD) is a neurodegenerative disorder characterized by neuron loss and abnormal protein trafficking. Dysregulation of vesicle-mediated transport contributes to pathogenesis, but its diagnostic value and immune associations are unclear. METHODS: Transcriptomic data from GEO datasets (GSE20141, GSE20163, GSE7621) were analyzed. Differentially expressed vesicle-mediated transport-related genes were identified. Machine learning algorithms (least absolute shrinkage and selection operator, random forest, extreme gradient boosting) were integrated to select robust diag

The Prognostic Significance of Vesicle-Mediated Transport-Related Genes as Risk Markers in Gastric Cancer.

Vesicle-mediated transport plays a role in intercellular communication in tumor microenvironment, affecting tumor development and prognosis. This study attempted to establish risk markers for gastric cancer (GC) based on vesicle-mediated transport-related genes, providing a new perspective for evaluating immunotherapeutic response and prognosis in GC patients. We used TCGA-STAD data set for training set and performed differential analysis and Cox regression analysis to establish a vesicle-mediated transport-related GC prognostic risk model to predict survival rate of GC patients. GC prognostic

A zinc transporter drives glioblastoma progression via extracellular vesicles-reprogrammed microglial plasticity.

Glioblastoma (GBM) is the most aggressive form of brain cancer, with limited therapeutic options. While microglia contribute to GBM progression, the mechanisms by which they foster a protumorigenic immune environment remain poorly understood. We identify the zinc transporter Zrt- And Irt-Like Protein 4 (ZIP4) as a pivotal regulator of the GBM immune landscape. In orthotopic mouse models, ZIP4 drives tumor growth and behavioral changes. Mechanistically, ZIP4 modulates microglial plasticity through tumor-derived extracellular vesicles carrying triggering receptor expressed on myeloid cells-1 (TR

Presenilin controls kinesin-1 and dynein function during APP-vesicle transport in vivo.

Neurons and other cells require intracellular transport of essential components for viability and function. Previous work has shown that while net amyloid precursor protein (APP) transport is generally anterograde, individual vesicles containing APP move bi-directionally. This discrepancy highlights our poor understanding of the in vivo regulation of APP-vesicle transport. Here, we show that reduction of presenilin (PS) or suppression of gamma-secretase activity substantially increases anterograde and retrograde velocities for APP vesicles. Strikingly, PS deficiency has no effect on an unrelat
