Verticillium wilt disease is one of the most destructive biotic stresses faced by cotton plants. Here, we performed a genome-wide association study (GWAS) in 215 Chinese Gossypium arboreum accessions inoculated as seedlings with Verticillium dahliae to identify candidate loci involved in wilt resistance. We identified 309 loci that had a significant association with Verticillium wilt resistance and - log(P) values >5.0; the highest signal appeared on Ca3 in a 74 kb haplotype block. Five genes were also located within this haplotype block. One of these genes, CG05, was positioned close to the m…

ABSTRACT Chronological events of the intercellular interaction between Verticillium lecanii and the postharvest pathogen Penicillium digitatum were investigated by transmission electron microscopy and gold cytochemistry. Growth inhibition of P. oligandrum as a response to V. lecanii attack correlated with striking host changes including retraction of the plasma membrane and cytoplasm disorganization. Such changes were associated with the deposition on the inner host cell surface of a chitin- and cellulose-enriched material which appeared to be laid down as a structural defense reaction. The ac…

Fungal and oomycete phytopathogens account for 10%-20% of annual global crop losses, and the escalating resistance to synthetic fungicides has intensified demand for plant-derived alternatives. The essential oil (EO) of Clinopodium nepeta (L.) Kuntze, obtained by hydrodistillation from aerial parts harvested in northeastern Morocco, was profiled by gas chromatography-mass spectrometry (GC-MS) and evaluated for antifungal and anti-oomycete activity against Verticillium dahliae, Phytophthora infestans, and Macrophomina phaseolina using broth microdilution minimum inhibitory concentration (MIC) a…

This study investigated the occurrence and field-level co-occurrence of cyst nematodes, Verticillium longisporum, and Pectobacterium spp. in highland kimchi cabbage fields in South Korea during 2023-2024. Three major pathogen groups and associated diseases were identified at the species level: cyst nematodes (CN; Heterodera trifolii and H. schachtii), Verticillium wilt caused by V. longisporum (VW), and bacterial soft rot caused by Pectobacterium carotovorum and P. brasiliense (BSR). Among 97 surveyed fields, individual detection frequencies for CN, VW, and BSR were 31%, 21%, and 54%, respecti…

Verticillium dahliae is a soilborne fungus that causes vascular wilt diseases on numerous plant species worldwide. The production of darkly melanized microsclerotia is crucial in the disease cycle of V. dahliae, as these structures allow for long-term survival in soil. Previously, transcriptomic and genomic analysis identified a cluster of genes in V. dahliae that encodes some dihydroxynaphthalene (DHN) melanin biosynthetic pathway homologues found in related fungi. In this study, we explored the roles of cluster-specific transcription factor VdCmr1, as well as two other genes within the clust…

Cotton S-adenosylmethionine decarboxylase-, rather than spermine synthase-, mediated spermine biosynthesis is required for salicylic acid- and leucine-correlated signaling in the defense response to Verticillium dahliae. Spermine (Spm) signaling is correlated with plant resistance to the fungal pathogen Verticillium dahliae. We identified genes for key rate-limiting enzymes in the biosynthesis of Spm, namely S-adenosylmethionine decarboxylase (GhSAMDC) and Spm synthase (GhSPMS). These were found by screening suppression subtractive hybridization and cDNA libraries of cotton (Gossypium) species…

Verticillium wilt causes massive annual losses of cotton yield, but the mechanism of cotton resistance to Verticillium dahliae is complex and poorly understood. In this study, a comparative proteomic analysis was performed in resistant cotton (Gossypium barbadense cv7124) on infection with V. dahliae. A total of 188 differentially expressed proteins were identified by mass spectrometry (MALDI-TOF/TOF) analysis and could be classified into 17 biological processes based on Gene Ontology annotation. Most of these proteins were implicated in stimulus response, cellular processes and metabolic proc…

ABSTRACT The potential of the mycoparasite, Verticillium lecanii, at protecting citrus fruits against green mold was explored at the cellular level. Treatmentthe fruit with V. lecanii or chitosan prior to inoculation with the causal agent of green mold, Penicillium digitatum, markedly reduced disease development compared with that of nontreated control citrus fruits in which symptoms were visible by 3 days after inoculation with the pathogen. Scanning electron microscope investigations of citrus samples, collected 5 days after inoculation with the pathogen, revealed striking differences in the…

KEY MESSAGE: Overexpression of a cotton defense-related gene GbSTK in Arabidopsis resulted in enhancing pathogen infection and oxidative str … cotton. Here, we focused on one such member, GbSTK. RT-PCR indicated that it is induced not only by Verticillium dahliae Kleb., but also by signaling molecules. Subcellular localization showed that GbSTK is present in the cell membrane, cytoplasm, and nucleus. Overexpression of GbSTK in Arabidopsis resulted into the enhanced resistance to V. dahliae. Moreover, …

The limited effectiveness of current strategies to control Verticillium wilt of olive (VWO) prompts the need for innovative approaches. This study explores the basal metabolome of 43 olive cultivars with varying resistance levels to Verticillium dahliae, offering alternative insights for olive crossbreeding programmes. The use of an innovative UHPLC-ESI-TimsTOF MS/MS platform enabled the annotation of more than 70 compounds across different olive organs (root, stem, and leaf) and the creation of a preliminary compilation of TIMSCCSN2 experimental data for more reliable metabolite annotation. M…

In vitro trials investigating the effects of albendazole and triclabendazole anthelmintics on the growth profiles of the egg-parasitic fungi Paecilomyces lilacinus and Verticillium chlamydosporium were undertaken. In addition, in vivo trials were conducted in goats fed on millet grain cultures of each fungus and administered albendazole and triclabendazole anthelmintics. In vitro growth revealed V. chlamydosporium to be more sensitive to albendazole compared to P. lilacinus. In contrast, triclabendazole had the least inhibitory effect on in vitro growth of both P. lilacinus and V. chlamydospor…

Verticillium wilt in cotton caused by Verticillium dahliae is one of the most serious plant diseases worldwide. Because no known fungicides or cotton cultivars provide sufficient protection against this pathogen, V. dahliae causes major crop yield losses. Here, an isolated cotton endophytic bacterium, designated Bacillus amyloliquefaciens 41B-1, exhibited greater than 50% biocontrol efficacy against V. dahliae in cotton plants under greenhouse conditions. Through high-performance liquid chromatography and mass analysis of the filtrate, we found that the antifungal compounds present in the stra…

Plant immunity is essential for survival against pathogen invasion. However, enhanced immune activation can restrict arbuscular mycorrhizal (AM) fungal colonization. Therefore, plants must finely balance immunity and symbiosis to optimize fitness, but the key regulators underlying this trade-off remain unclear. Here, we identify a novel mitogen-activated protein kinase, GhNTF3, as a central regulator of the AM symbiosis-immunity balance. Knockdown of GhNTF3 promotes AM symbiosis but decreases Verticillium wilt resistance in cotton. GhNTF3 overexpression suppresses AM symbiosis. GhNTF3 interact…

The number of raspberry plants dying from a sudden outbreak of gray mold, verticillium wilt, anthracnosis, and phytophthora infection has increased in recent times, leading to crop failure. The plants suffer tissue collapse and black roots, symptoms similar to a Botrytis-Verticillium-Colletotrichum-Phytophthora disease complex. A sizeable number of fungal isolates were acquired from the root and rhizosphere samples of wild raspberries from different locations. Subsequent in vitro tests revealed that a core consortium of 11 isolates of selected Trichoderma spp. was the most essential element fo…

The escalating severity of antifungal resistance necessitates novel crop protection strategies. RNAi is promising but faces challenges including environmental instability of dsRNA and poor cellular delivery efficiency. In this study, we engineered the cotton rhizobacterium Pantoea agglomerans YC4 to deliver dsRNA into Verticillium dahliae. Small RNA-seq revealed that bacterial outer membrane vesicles (OMVs) selectively enrich specific endogenous sRNAs compared with intracellular compartments, with a congruent sRNA enrichment pattern observed in fungal cells, thus confirming OMVs as the primary…
