The role of antibody indexes in clinical virology.

BACKGROUND: Serological techniques are an essential part of the diagnostic tools used in clinical virology. Among these techniques, antibody indexes are not novel, but do require specific expertise. Their niche has expanded substantially in recent years due to increasing evidence of their performance to diagnose viral infections. OBJECTIVES: This narrative review describes the background and clinical applications of antibody indexes. The first objective is to provide an overview of the theoretical background, insights for implementation, limitations and pitfalls. The second objective is to re

Virology, Epidemiology, Pathogenesis, and Control of COVID-19.

The outbreak of emerging severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) disease (COVID-19) in China has been brought to global attention and declared a pandemic by the World Health Organization (WHO) on March 11, 2020. Scientific advancements since the pandemic of severe acute respiratory syndrome (SARS) in 2002~2003 and Middle East respiratory syndrome (MERS) in 2012 have accelerated our understanding of the epidemiology and pathogenesis of SARS-CoV-2 and the development of therapeutics to treat viral infection. As no specific therapeutics and vaccines are available for disease

Comparative virology of HTLV-1 and HTLV-2.

Human T cell leukemia virus type 1 (HTLV-1) was the first discovered human retrovirus and the etiologic agent of adult T-cell leukemia and HTLV-1-associated myelopathy/tropical spastic paraparesis. Shortly after the discovery of HTLV-1, human T-cell leukemia virus type 2 (HTLV-2) was isolated from a patient with hairy cell leukemia. Despite possession of similar structural features to HTLV-1, HTLV-2 has not been definitively associated with lymphoproliferative disease. Since their discovery, studies have been performed with the goal of highlighting the differences between HTLV-1 and HTLV-2. A

Highlighting the 'blood-nerve barrier' in virology research.

The blood-nerve barrier (BNB) shields peripheral nerves from the blood in order to maintain the homeostasis of the nervous system. In the field of infectious diseases, little information is currently available concerning the BNB. Recently documented evidence in virology suggests that elevated permeability of the BNB by immune cells and the natural absence of the BNB in the olfactory mucosa play significant roles in neuroprotection as well as neuropathogenesis. Importantly, the BNB can behave more flexibly than previously thought. In the near future, drug delivery via manipulation of the BNB wi

1st Workshop of the Canadian Society for Virology.

The 1st Workshop of the Canadian Society for Virology (CSV2016) was a Special Workshop of the 35th Annual Meeting for the American Society for Virology, held on 18 June 2016 on the beautiful Virginia Tech campus in Blacksburg, Virginia. The workshop provided a forum for discussion of recent advances in the field, in an informal setting conducive to interaction with colleagues. CSV2016 featured two internationally-renowned Canadian keynote speakers who discussed translational virology research; American Society for Virology President Grant McFadden (then from University of Florida, now relocate

An update on HDV: virology, pathogenesis and treatment.

Hepatitis delta is an inflammatory liver disease caused by infection with HDV. HDV is a single-stranded circular RNA pathogen with a diameter of 36 nm. HDV is classified in the genus Deltavirus and is still awaiting a final taxonomic classification up to the family level. HDV shares similarities with satellite RNA and viroids including a small circular single-stranded RNA with secondary structure that replicates through the 'double rolling circle' mechanism. The HDV RNA genome is capable of self-cleavage through a ribozyme and encodes only one structural protein, the hepatitis delta antigen (H

Chapter 1 Overview of Health-Related Water Virology.

Viruses are a major cause of waterborne and water-related diseases. Extreme examples include the outbreak of hepatitis A and of viral gastroenteritis in Shanghai caused by shellfish harvested from a sewage-polluted estuary. Viruses predominantly associated with waterborne transmission are members of the group of enteric viruses that primarily infect cells of the gastrointestinal tract, and are excreted in the faeces of infected individuals. The viruses concerned are highly host specific, which implies that their presence in water environments is sound evidence of human faecal pollution. In som

Importance and impact of veterinary virology in Germany.

The causative agent of tobacco mosaic and of foot and mouth disease (FMD) were recognized in 1898 as "filterable" or "invisible"--and eventually termed "virus". Four years later the viral aetiology of yellow fever was established, and the new discipline took off. Thus animal virology started with a veterinary problem, and Germany's contribution during the following decades came mainly from the chairs of veterinary teaching and research establishments in Giessen, Munich and Hanover, the Riems Institute, and the Federal Research Institute for Animal Virus Diseases in Tübingen. From a superficial

Clinical virology of Lassa fever in hospitalized patients.

We measured levels of virus in sequential specimens from 137 patients with Lassa fever. The probability of fatal disease increased significantly with the level of viremia measured either on admission or during the course of illness. The odds ratio of death in patients with viremia greater than 10 TCID50/ml was 3.7 (90% confidence interval, 1.9-7.2). The same ratio in patients with viremia greater than 10 TCID50/ml and with levels of aspartate aminotransferase greater than or equal to 150 IU/liter was 21.5 (95% confidence interval, 5.2-99.0). Virus was found in throat cultures from 39% of virem

Aleutian disease in mink: virology, immunology and pathogenesis.

Aleutian disease is an immunological disease of mink caused by a persistent virus infection. Recently Aleutian disease viral antigen has been extracted from tissue of mink early in the course of infection and the Aleutian disease virus has been isolated. The virus particles were similar in morphology and size to certain parvovirus having a 23 nm diameter, spherical shape and icosahedral capsid. Recent developments in serological techniques, i.e. immunofluorescence, complement fixation, countercurrent electrophoresis and immunodiffusion, have enabled rapid progress toward understanding the natu

[Microbiological diagnosis of ocular surface infections aside from virology].

The ocular surface is a structure exposed to multiple stresses that can directly or indirectly lead to infectious contamination and compromise visual function. This significant risk justifies multidisciplinary care, often in an emergency context, bringing together ophthalmologists and microbiologists to optimize and guide microbiological testing on small volume samples and ensure the most appropriate treatment. Herein, we present the various steps leading to microbiological diagnosis, from pre-analytical requirements, sampling methods, transport conditions, and then to the analytical steps of

Virology under the Microscope-a Call for Rational Discourse.

Viruses have brought humanity many challenges: respiratory infection, cancer, neurological impairment and immunosuppression to name a few. Virology research over the last 60+ years has responded to reduce this disease burden with vaccines and antivirals. Despite this long history, the COVID-19 pandemic has brought unprecedented attention to the field of virology. Some of this attention is focused on concern about the safe conduct of research with human pathogens. A small but vocal group of individuals has seized upon these concerns - conflating legitimate questions about safely conducting viru
