The restriction of the heart morphogenetic field in Xenopus laevis.

We have examined the spatial restriction of heart-forming potency in Xenopus laevis embryos, using an assay system in which explants or explant recombinates are cultured in hanging drops and scored for the formation of a beating heart. At the end of neurulation at stage 20, the heart morphogenetic field, i.e., the area that is capable of heart formation when cultured in isolation, includes anterior ventral and ventrolateral mesoderm. This area of developmental potency does not extend into more posterior regions. Between postneurula stage 23 and the onset of heart morphogenesis at stage 28, the

The glomus cell of the carotid labyrinth of Xenopus laevis.

The ultrastructure of the glomus cells of the carotid labyrinth was investigated in the anuran, Xenopus laevis. These cells show many catecholamine containing granules. About 50 cells in groups of 3-5 are located near the sinusoids. Morphologically, their organelles are similar to those previously reported in Bufo vulgaris. Striking features are (1) intimate appostion of the glomus cell to smooth muscle (g-s connection), (2) gap junctions between adjacent glomus cells, (3) exocytotic figures. Based on these findings a possible function of the glomus cell is discussed.

The orientation of the visuotectal map in Xenopus: developmental aspects.

Rotations and translocations of the eye anlage were performed in Xenopus embryos of stages ranging from 21/22 to 30. Some of the operations involved grafting wild-type eye anlagen into albino host orbits. Operations were performed under a variety of operating media and conditions. In later larval life, or after metamorphosis, the visuotectal maps from the operated eyes were recorded electrophysiologically. Results fell into two classes. In the majority, the orientation of the visuotopic map corresponded to the orientation of the eye at the time of recording. In the minority the visuotopic maps

Reconstitution of genome remodeling using Xenopus egg extracts.

Changes in 3D genome organization drive critical cellular transitions including cell division and differentiation. Cytoplasmic extracts derived from eggs of the African clawed frog Xenopus laevis offer a powerful system for reconstituting genome remodeling events in vitro and dissecting the underlying biochemical mechanisms. Female X. laevis produce thousands of large eggs that can be readily lysed by centrifugation to yield egg cytoplasm-commonly referred to as egg extract. This extract contains the full complement of molecules required to support multiple rounds of chromatin assembly, nuclea

Working Memory Y-Maze Test for Xenopus Tadpoles.

Working memory refers to the multifaceted cognitive systems that are considered necessary for retaining information while performing complex tasks, such as reasoning and learning in humans. In vertebrates, working memory is evident as a conserved alternating searching strategy. We can use automated tracking tools to measure this search strategy in Xenopus tadpoles. The free-movement pattern Y-maze assay is validated for assessing spatial working memory and cognitive flexibility in Xenopus. Deviations from randomness in search strategies are quantified by comparing relative alternating and repe

Neighbor cells restrain furrowing during Xenopus epithelial cytokinesis.

Cytokinesis challenges epithelial tissue homeostasis by generating forces that pull on neighboring cells. Junction reinforcement at the furrow in Xenopus epithelia regulates the speed of furrowing, suggesting that cytokinesis is subject to resistive forces from epithelial neighbors. We show that contractility factors accumulate near the furrow in neighboring cells, and increasing neighbor cell stiffness slows furrowing. Optogenetically increasing contractility in one or both neighbor cells slows furrowing or induces cytokinetic failure. Uncoupling mechanotransduction between dividing cells and

Expression analysis of thg1l during Xenopus laevis development.

The tRNA-histidine guanylyltransferase 1-like (THG1L), also known as induced in high glucose-1 (IHG-1), encodes for an essential mitochondria-associated protein highly conserved throughout evolution, that catalyses the 3'-5' addition of a guanine to the 5'-end of tRNA-histidine (tRNAHis). Previous data indicated that THG1L plays a crucial role in the regulation of mitochondrial biogenesis and dynamics, in ATP production, and is critically involved in the modulation of apoptosis, cell-cycle progression and survival, as well as in cellular stress responses and redox homeostasis. Dysregulations o

Age-associated DNA methylation changes in Xenopus frogs.

Age-associated changes in DNA methylation have been characterized across various animals, but not yet in amphibians, which are of particular interest because they include widely studied model organisms. In this study, we present clear evidence that the aquatic vertebrate species Xenopus tropicalis displays patterns of age-associated changes in DNA methylation. We have generated whole-genome bisulfite sequencing (WGBS) profiles from skin samples of nine frogs representing young, mature, and old adults and characterized the gene- and chromosome-scale DNA methylation changes with age. Many of the

Modeling Bainbridge-Ropers Syndrome in Xenopus laevis Embryos.

The Additional sex combs-like (ASXL1-3) genes are linked to human neurodevelopmental disorders. The de novo truncating variants in ASXL1-3 proteins serve as the genetic basis for severe neurodevelopmental diseases such as Bohring-Opitz, Shashi-Pena, and Bainbridge-Ropers syndromes, respectively. The phenotypes of these syndromes are similar but not identical, and include dramatic craniofacial defects, microcephaly, developmental delay, and severe intellectual disability, with a loss of speech and language. Bainbridge-Ropers syndrome resulting from ASXL3 gene mutations also includes features of

Using Xenopus to analyze neurocristopathies like Kabuki syndrome.

Neurocristopathies are human congenital syndromes that arise from defects in neural crest (NC) development and are typically associated with malformations of the craniofacial skeleton. Genetic analyses have been very successful in identifying pathogenic mutations, however, model organisms are required to characterize how these mutations affect embryonic development thereby leading to complex clinical conditions. The African clawed frog Xenopus laevis provides a broad range of in vivo and in vitro tools allowing for a detailed characterization of NC development. Due to the conserved nature of c

Collagen-Embedded Tumor Transplantations in Xenopus laevis Tadpoles.

The Xenopus laevis tadpole provides a valuable model for studying tumorigenesis and tumor immunity by intravital real-time microscopy. Using well-characterized thymic lymphoid tumor lines (15/0 and ff-2) that are transplantable into their compatible hosts (LG-15 isogenic clones and the F inbred strain, respectively), a system of semisolid tumor engraftment has been designed. Because these lymphoid tumor cell lines are not adherent and grow in suspension, they are first immobilized in a matrix of type I rat tail collagen before transplantation as a semisolid tumor graft under the transparent do

Xmc mediates Xctr1-independent morphogenesis in Xenopus laevis.

In the frog, Xenopus laevis, fibroblast growth factor (FGF) signaling is required for both mesoderm formation and the morphogenetic movements that drive the elongation of the notochord, a dorsal mesodermal derivative; the coordination of these distinct roles is mediated by the Xenopus Ctr1 (Xctr1) protein: maternal Xctr1 is required for mesodermal differentiation, while the subsequent loss of Xctr1 promotes morphogenesis. The signaling cascade activated by FGF in the presence of Ctr1 has been well characterized; however, the Xctr1-independent, FGF-responsive network remains poorly defined. We

Organizer induction determines left-right asymmetry in Xenopus.

Vertebrates appear bilaterally symmetrical but have considerable left-right (LR) asymmetry in the anatomy and placement of internal organs such as the heart. Although a number of asymmetrically expressed genes are known to affect LR patterning, both the initial source of asymmetry and the mechanism that correctly orients the LR axis remain controversial. In this study, we show that the induction of dorsal organizing centers in the embryo can orient LR asymmetry. Ectopic organizing centers were induced by microinjection of mRNA encoding a variety of body axis duplicating proteins, including mem

Efficient hormone-inducible protein function in Xenopus laevis.

A major problem in analyzing gene function during Xenopus development has been the inability to induce gene expression in a temporally controlled manner. We have attempted to solve this problem with a system of hormone-activated protein function, using the myogenic gene MyoD as a paradigm. We show that microinjection of RNA for MyoD fused to the ligand-binding domain of either the estrogen or glucocorticoid receptor results in hormone-dependent activation of MyoD function, as assayed by ectopic induction of muscle-specific actin mRNA. Induction is tightly regulated in both isolated animal caps

High tyrosinase activity in albino Xenopus laevis oocytes.

Tyrosinase was measured in oocytes of the recently described albino mutant (apap) of Xenopus laevis. Although these oocytes show no pigmentation and the eggs are known to contain no melanosomes, tyrosinase - which is probably the only enzyme necessary for melanin synthesis from tyrosine - was increased more than twofold relative to the wild type. Tyrosinase recovered from albino and wild type oocytes showed the same Km with respect to tyrosine, and this was not altered by previous gonadotrophin stimulation in vivo. The tyrosinase assay, based on [14]tyrosine incorporation into acid-insoluble p

Control performance of Amphibian Metamorphosis Assays with Xenopus laevis.

The amphibian metamorphosis assay (AMA) is an in vivo screen to assess potential interactions of chemicals with the amphibian thyroid system. Tadpoles are exposed for 21-days, then assessed for development and growth after 7 days and at test termination. This paper presents data from studies performed to satisfy test orders from the US EPA's Endocrine Disruptor Screening Program. Data Evaluation Records were used to collate the control variability and performance of biological endpoints in AMAs conducted in different laboratories, then supplemented with recent studies. We examine the statistic

Endogenous Retroviruses Augment Amphibian (Xenopus laevis) Tadpole Antiviral Protection.

The global amphibian declines are compounded by infections with members of the Ranavirus genus such as Frog Virus 3 (FV3). Premetamorphic anuran amphibians are believed to be significantly more susceptible to FV3 while this pathogen targets the kidneys of both pre- and postmetamorphic animals. Paradoxically, FV3-challenged Xenopus laevis tadpoles exhibit lower kidney viral loads than adult frogs. Presently, we demonstrate that X. laevis tadpoles are intrinsically more resistant to FV3 kidney infections than cohort-matched metamorphic and postmetamorphic froglets and that this resistance appear

Xenopus in revealing developmental toxicity and modeling human diseases.

The Xenopus model offers many advantages for investigation of the molecular, cellular, and behavioral mechanisms underlying embryo development. Moreover, Xenopus oocytes and embryos have been extensively used to study developmental toxicity and human diseases in response to various environmental chemicals. This review first summarizes recent advances in using Xenopus as a vertebrate model to study distinct types of tissue/organ development following exposure to environmental toxicants, chemical reagents, and pharmaceutical drugs. Then, the successful use of Xenopus as a model for diseases, inc

Dicer inactivation stimulates limb regeneration ability in Xenopus laevis.

The ontogenetic decline of regeneration capacity in the anuran amphibian Xenopus makes it an excellent model for regeneration studies. However, the cause of the regeneration ability decline is not fully understood. MicroRNAs regulate animal development and have been indicated in various regeneration situations. However, little is known about the role of microRNAs during limb regeneration in Xenopus. This study investigates the effect of Dicer, an enzyme responsible for microRNA maturation, on limb development and regeneration in Xenopus. Dicer is expressed in the developing Xenopus limbs and i
