Supplementation with lutein and zeaxanthin increases macular pigment optical density and cognitive performance in healthy teenagers in a randomized, double-blind, placebo-controlled trial.

Studies in adults have demonstrated that lutein and zeaxanthin (LZ) supplementation may improve eye and cognitive function, although limited studies have been conducted in teenagers. In this two-arm, 6-month, parallel-group, randomized, double-blind, placebo-controlled trial, supplementation with a placebo (sunflower oil) or lutein (10 mg) and zeaxanthin (2 mg) was examined for its effects on macular pigment optical density (MPOD), cognitive function, and sleep in teenagers aged 13 to 18 years with high screen use and consuming a diet low in fruits and vegetables. Eighty-two teenagers were ass

Lutein and zeaxanthin isomers may attenuate photo-oxidative retinal damage via modulation of G protein-coupled receptors and growth factors in rats.

BACKGROUND: Retina photoreceptor cells are specially adapted for functioning over comprehensive ambient light conditions. Lutein and Zeaxanthin isomers (L/Zi) can protect photoreceptor cells against excessive light degeneration. Efficacy of L/Zi has been assessed on some G protein-coupled receptors (GPCRs), transcription and neurotrophic factors in the retina of rats exposed to incremental intense light emitting diode (LED) illumination conditions. METHODS: Forty-two male rats (age: 8 weeks) were randomly assigned to six treatment groups, 7 rats each. The rats with a 3x2 factorial design were

Determination of Lutein and Zeaxanthin Esters and Their Geometric Isomers in Carotenoid Ester Concentrates Used as Ingredients in Nutritional Supplements: Validation of a Combined Spectrophotometric-HPLC Method.

A combined spectrophotometric-LC method is described for the determination of total lutein and zeaxanthin ester content in carotenoid ester concentrates, including their main geometrical isomers. The concept of composite-specific absorbance is introduced for this purpose. The method is applicable to carotenoid ester concentrates used as ingredients in oil suspensions and dosage forms. The sample is dissolved in a hexane-2-propanol mixture (95 + 5, v/v) for spectrophotometric measurement at a maximum absorption of ~445 nm. Subsequently, in parallel, a sample is saponified and chromatographed on

Nonlethal Levels of Zeaxanthin Inhibit Cell Migration, Invasion, and Secretion of MMP-2 via NF-κB Pathway in Cultured Human Uveal Melanoma Cells.

Zeaxanthin at nonlethal dosages (3-10 μM) significantly inhibited the cell migration of cultured uveal melanoma cells (C918 cell line) as determined by wound healing assay and Boyden chamber assay. Matrigel invasion assay showed that cell invasion of uveal melanoma cells could be significantly inhibited by zeaxanthin. Secretion of MMP-2 by melanoma cells was significantly inhibited by zeaxanthin in a dose-dependent manner as measured by ELISA kit. Zeaxanthin also significantly inhibited the NF-κB levels in nuclear extracts of the UM cells, which is the upstream of the MMP-2 secretion. These re

The effects of lutein, zeaxanthin and anthocyanins supplementation on visual function and vision protection in healthy school-aged children: a single-center prospective randomized controlled study.

Dietary carotenoids (lutein and zeaxanthin) and anthocyanins show retinal protective potential, yet their clinical efficacy in healthy children's developing visual systems remains unclear. We aimed to investigate whether lutein, zeaxanthin, and anthocyanin (LA) supplementation enhances visual function and vision protection in healthy school-aged children (6-10 years). Sixty-four participants were randomized to control or treatment (received LA supplementation daily) groups. Macular pigment optical density (MPOD), central foveal thickness (CFT), minimum foveal thickness (MFT), axial length (AL)

Interaction between Infection of Porphyromonas gingivalis, A Keystone Microbe of Oral Microbiome, and Serum Levels of Lutein/Zeaxanthin Is Associated with Risk for Age-related Macular Degeneration.

UNLABELLED: Porphyromonas gingivalis (P. gingivalis) functions as a catalyst bacterium in the development of periodontitis, and the serum antibody level against P. gingivalis is considered a surrogate marker for the activity level of periodontopathic microbiome. The chronic systemic inflammation induced by P. gingivalis elevates the risk of various systemic and neurodegenerative disorders, including atherosclerosis, diabetes, and Alzheimer's disease. Although the connection between human microbiome and age-related macular degeneration (AMD) remains relatively unexplored, it is noteworthy that

Aflatoxin B1-induced toxicity, oxido-inflammatory damage, and apoptosis in male rat reproductive circuitry were abrogated by co-treating with the xanthophyll-lutein and zeaxanthin.

Reproductive dysfunction is a recognised adverse effect of exposure to aflatoxin B1 (AFB1) in humans and animals. Despite the widely acknowledged health risks, exposure to AFB1 remains unavoidable. Conversely, lutein (LUT) and zeaxanthin (ZEA) are plants' potent antioxidants and anti-inflammatory agents, exhibiting promising potential for modulating inflammatory and apoptotic signalling pathways. This study aimed to investigate the effects of co-treatment with LUT/ZEA on reproductive function in rats intoxicated with AFB1. The study utilised male Wistar rats (n = 20 and n = 4 per cohort). Long-term Outcomes of Adding Lutein/Zeaxanthin and ω-3 Fatty Acids to the AREDS Supplements on Age-Related Macular Degeneration Progression: AREDS2 Report 28.

IMPORTANCE: After the Age-Related Eye Disease Study 2 (AREDS2) study, the beta carotene component was replaced by lutein/zeaxanthin for the development of the revised AREDS supplement. However, it is unknown if the increased risk of lung cancer observed in those assigned beta carotene persists beyond the conclusion of the AREDS2 trial and if there is a benefit of adding lutein/zeaxanthin to the original AREDS supplement that can be observed with long-term follow-up. OBJECTIVE: To assess 10-year risk of developing lung cancer and late age-related macular degeneration (AMD). DESIGN, SETTING, A

Macular pigment carotenoids in the retina and occipital cortex are related in humans.

OBJECTIVES: Lutein and zeaxanthin are dietary carotenoids that preferentially accumulate in the macular region of the retina. Together with meso-zeaxanthin, a conversion product of lutein in the macula, they form the macular pigment. Lutein is also the predominant carotenoid in human brain tissue and lutein status is associated with cognitive function in adults. The study objective was to evaluate the relationship between retinal and brain lutein and zeaxanthin in humans. METHODS: Donated brain tissue (occipital cortex and hippocampus) and matched retina were obtained from the National Diseas

Genetic deletion of Bco2 and Isx establishes a golden mouse model for carotenoid research.

OBJECTIVE: Low plasma levels of carotenoids are associated with mortality and chronic disease states. Genetic studies in animals revealed that the tissue accumulation of these dietary pigments is associated with the genes encoding β-carotene oxygenase 2 (BCO2) and the scavenger receptor class B type 1 (SR-B1). Here we examined in mice how BCO2 and SR-B1 affect the metabolism of the model carotenoid zeaxanthin that serves as a macular pigment in the human retina. METHODS: We used mice with a lacZ reporter gene knock-in to determine Bco2 expression patterns in the small intestine. By genetic di

A novel gastroprotective effect of zeaxanthin against stress-induced gastritis in male rats targeting the expression of HIF-1α, TFF-1 and MMP-9 through PI3K/Akt/JNK signaling pathway.

Stress-induced gastritis is a common problem in the intensive care unit. Zeaxanthin (ZE), a non-provitamin A carotenoid has been known to exert antioxidant and anti-inflammatory effects. In this study, we examined the effect of ZE on water avoidance stress (WAS)-induced gastritis in rats. 24 Sprague' Dawley male rats were divided into four groups; control, ZE, WAS and WAS+ZE. In the stressed rats, treatment with ZE effectively downregulated the gastric levels of total oxidant status (TOS), myeloperoxidase (MPO) and malondialdehyde (MDA), with significant upregulation of the antioxidant enzymes

Nitroxide free radicals protect macular carotenoids against chemical destruction (bleaching) during lipid peroxidation.

Macular xanthophylls (MXs) lutein and zeaxanthin are dietary carotenoids that are selectively concentrated in the human eye retina, where they are thought to protect against age-related macular degeneration (AMD) by multiple mechanisms, including filtration of phototoxic blue light and quenching of singlet oxygen and triplet states of photosensitizers. These physical protective mechanisms require that MXs be in their intact structure. Here, we investigated the protection of the intact structure of zeaxanthin incorporated into model membranes subjected to oxidative modification by water- and/or

Egg intake improves carotenoid status by increasing plasma HDL cholesterol in adults with metabolic syndrome.

Metabolic syndrome (MetS) is associated with reductions in plasma lutein and zeaxanthin, along with altered composition of their lipoprotein transporters which may affect disease risk. Egg yolk intake has been demonstrated to increase plasma lutein and zeaxanthin in other populations. The objective of this study was to investigate the effects of whole egg feeding on plasma and lipoprotein carotenoids in participants with MetS. Participants consumed 3 whole eggs per day (EGG, n = 20) or the equivalent amount of yolk-free egg substitute (SUB, n = 17), as part of a carbohydrate-restricted diet (C
