Principles of Endocrine Regulation: Reconciling Tensions Between Robustness in Performance and Adaptation to Change.

Endocrine regulation in the hypothalamic-pituitary-thyroid (HPT) axis is orchestrated by physiological circuits which integrate multiple internal and external influences. Essentially, it provides either of the two responses to overt biological challenges: to defend the homeostatic range of a target hormone or adapt it to changing environmental conditions. Under certain conditions, such flexibility may exceed the capability of a simple feedback control loop, rather requiring more intricate networks of communication between the system's components. A new minimal mathematical model, in the form o

Integrating and synthesizing adversity and resilience knowledge and action: The ICARE model.

This article proposes a model for understanding the effects of adverse childhood experiences (ACEs) as dynamic and interrelated biobehavioral adaptations to early life stress that have predictable consequences on development and health. Drawing upon research from multiple theoretical and methodological approaches, the intergenerational and cumulative adverse and resilient experiences (ICARE) model posits that the negative consequences of ACEs result from biological and behavioral adaptations to adversity that alter cognitive, social, and emotional development. These adaptations often have nega

Comprehensive targeting of resistance to inhibition of RTK signaling pathways by using glucocorticoids.

Inhibition of RTK pathways in cancer triggers an adaptive response that promotes therapeutic resistance. Because the adaptive response is multifaceted, the optimal approach to blunting it remains undetermined. TNF upregulation is a biologically significant response to EGFR inhibition in NSCLC. Here, we compared a specific TNF inhibitor (etanercept) to thalidomide and prednisone, two drugs that block TNF and also other inflammatory pathways. Prednisone is significantly more effective in suppressing EGFR inhibition-induced inflammatory signals. Remarkably, prednisone induces a shutdown of bypass

Redox activation of ATG5 licenses autophagy upon nutrient restriction.

Dietary restriction (DR) protects against metabolic disease, extends lifespan, and is associated with remodeling of tissue reactive oxygen species (ROS). ROS control biological adaptation through reversible oxidation of protein cysteines, yet the targets of DR-initiated redox signaling are unknown. Here we generate OxiDR, a tissue-resolved atlas of the cysteine redox proteome that quantifies oxidation state under DR. Rather than oxidizing the proteome broadly, DR selectively targets a high-amplitude set of cysteines in a tissue-specific manner, allowing systematic classification of biological

Photoelectrochemical determination of nitrite as a biomarker of exercise-induced nitric oxide metabolism in athletes using a CdS/polyaniline sensor.

The precise detection of nitrite as a stable, end product and downstream metabolite of NO is critical for understanding exercise-induced physiological adaptation in athletes. However, typical electrochemical techniques usually have restricted sensitivity, photointerference, and matrix-dependent inaccuracy when detecting biological samples. In this work, we fabricated a PEC platform exploiting an ultrahigh-photocurrent CdS/polyaniline (CdS/PANI) heterostructured film to facilitate accurate and selective nitrite detection as a biologically important health parameter of exercise-induced NO metabo

Epigenomics and Ayurgenomics: Molecular Analysis of Ayurvedic Prakriti in Personalized Healthcare.

Personalized healthcare increasingly requires models that explain individual variation in disease risk, prevention, and treatment response. Ayurvedic Prakriti offers a constitution-based framework for classifying human variability through Vata, Pitta, and Kapha phenotypes, while Ayurgenomics attempts to connect these traditional classifications with molecular biology. Despite growing interest, the biological relationship between Prakriti, epigenomic regulation, and precision healthcare remains insufficiently established, particularly because available evidence is limited by small cohorts, hete

Polysaccharide-Based Injectable Bioadhesive Hydrogel for Local Arterial Occlusion.

Injectable hydrogels offer a promising material platform for local filling and sealing by enabling needle delivery, in situ network formation, and conformal adaptation to irregular biological spaces. In vascular occlusion applications, effective material localization requires cohesive post-injection network recovery and stable retention at the target site after blood flow restoration. Here, we developed a polysaccharide-based injectable bioadhesive hydrogel, termed CCOD-gel, composed of chitosan, catechol-modified chitosan, oxidized dextran, and 3,4-dihydroxybenzaldehyde through dynamic Schiff

A Narrative Review on Stress Granules and Ultraviolet Stress Granules.

Background: Stress granules (SGs) are membraneless structures formed through liquid-liquid phase separation in response to cellular stress. These dynamic assemblies play a pivotal role in cellular adaptation and recovery from external stressors by modulating protein synthesis and preserving mRNA integrity, thereby influencing various cellular biological processes. Their significance has been demonstrated in diverse phenomena, including tumorigenesis, drug resistance, cellular senescence, and neurodegenerative disorders. Objective: This narrative review focuses on SGs and ultraviolet-induced SG

Continuous neural control of a bionic limb restores biomimetic gait after amputation.

For centuries scientists and technologists have sought artificial leg replacements that fully capture the versatility of their intact biological counterparts. However, biological gait requires coordinated volitional and reflexive motor control by complex afferent and efferent neural interplay, making its neuroprosthetic emulation challenging after limb amputation. Here we hypothesize that continuous neural control of a bionic limb can restore biomimetic gait after below-knee amputation when residual muscle afferents are augmented. To test this hypothesis, we present a neuroprosthetic interface

HIF-1α signaling: Essential roles in tumorigenesis and implications in targeted therapies.

The hypoxic microenvironment is an essential characteristic of most malignant tumors. Notably, hypoxia-inducible factor-1 alpha (HIF-1α) is a key regulatory factor of cellular adaptation to hypoxia, and many critical pathways are correlated with the biological activity of organisms via HIF-1α. In the intra-tumoral hypoxic environment, HIF-1α is highly expressed and contributes to the malignant progression of tumors, which in turn results in a poor prognosis in patients. Recently, it has been indicated that HIF-1α involves in various critical processes of life events and tumor development via r

Human Skin Pigmentation: From a Biological Feature to a Social Determinant.

Skin pigmentation is the most variable human characteristic that can be observed and has been used throughout history to classify humans into distinct groups. Many factors influence skin colour, but the melanin pigment is considered the most important because its type and quantity can determine variations in pigmentation shades. The evolution of skin pigmentation started around 1.6-2 million years ago. As a result of migratory phenomena to places with less ultraviolet radiation (UVR) and other seasonal regimes, the selection of depigmented skin and different tanning capabilities occurred over

Characterization of H9N2 Avian Influenza Viruses Isolated from Poultry Products in a Mouse Model.

Low pathogenic H9N2 avian influenza viruses have spread in wild birds and poultry worldwide. Recently, the number of human cases of H9N2 virus infection has increased in China and other countries, heightening pandemic concerns. In Japan, H9N2 viruses are not yet enzootic; however, avian influenza viruses, including H5N1, H7N9, H5N6, and H9N2, have been repeatedly detected in raw poultry meat carried by international flight passengers from Asian countries to Japan. Although H9N2 virus-contaminated poultry products intercepted by the animal quarantine service at the Japan border have been charac

Evolution of A(H1N1) pdm09 influenza virus masking by glycosylation.

Introduction: As the pathogen that caused the first influenza virus pandemic in this century, the swine-origin A(H1N1) pdm09 influenza virus has caused continuous harm to human public health. The evolution of hemagglutinin protein glycosylation sites, including the increase in number and positional changes, is an important way for influenza viruses to escape host immune pressure. Based on the traditional influenza virus molecular monitoring, special attention should be paid to the influence of glycosylation evolution on the biological characteristics of virus antigenicity, transmission and pat

Enhancement of tolerance of Indian mustard (Brassica juncea) to mercury by carbon monoxide.

Carbon monoxide (CO) is a hazardous gaseous molecule, whose concentration in atmosphere is recently rising. CO also is an endogenous regulator of a variety of biological processes in animals and plants. However, whether CO regulates plant adaptation to Hg-contaminated environments is unknown. In this study, we investigated the effect of CO on biological responses of Indian mustard (Brassica juncea), a plant species frequently used for heavy metal accumulation, to mercury (Hg) toxicity. Exposure of B. juncea to Hg(II) triggered production of O(2)(-) and H(2)O(2) as well as peroxides. However, s

Molecular biology of neurohormone precursors in the eyestalk of Crustacea.

Our knowledge concerning the primary structures of crustacean neuropeptides has been broadened considerably during the last few years and has greatly contributed to the successful application of molecular biological techniques to crustacean neuroendocrine research. In this review, we compare and discuss the preprohormones of the Red Pigment Concentrating Hormone (RPCH), the Pigment-Dispersing Hormone (PDH) and the different members of the Crustacean Hyperglycemic Hormone, Molt-Inhibiting and Gonad-Inhibiting Hormone family (CHH/MIH/GIH peptide family), recently elucidated by cloning and sequen

Thyroid dysfunction: Another piece in the complex puzzle of Alzheimer's disease?

An expanding body of evidence suggests that thyroid dysfunction may be clinically relevant in Alzheimer's disease (AD), although the significance of this association remains unsettled. Rather than reflecting a single biological pathway, thyroid hormone alterations in AD may involve downstream systemic adaptations related to inflammation and frailty, central dysregulation of thyroid hormone bioavailability and signaling, and upstream vulnerability mechanisms. Clarifying how these pathways intersect with cognitive and functional decline may help refine the biological interpretation of clinical h

Systematic evaluation of deep learning design choices for image-based organ-on-chip quality assessment.

Label-free brightfield microscopy is widely used to monitor organ-on-chip (OOC) cultures, but manual quality assessment can be subjective, labor-intensive, and difficult to standardize across biological conditions. Using a public dataset of 3072 microscopy images from six cell lines, we systematically evaluated four questions relevant to automated OOC quality assessment: (1) how model architecture and image resolution affect performance, (2) whether predictive information is localized or distributed across the image, (3) whether biological metadata provides complementary information beyond the

Gene selection based on adaptive neighborhood-preserving multi-objective particle swarm optimization.

The analysis of high-dimensional microarray gene expression data presents critical challenges, including excessive dimensionality, increased computational burden, and sensitivity to random initialization. Traditional optimization algorithms often produce inconsistent and suboptimal results, while failing to preserve local data structures limiting both predictive accuracy and biological interpretability. To address these limitations, this study proposes an adaptive neighborhood-preserving multi-objective particle swarm optimization (ANPMOPSO) framework for gene selection. ANPMOPSO introduces fo

The feeding biomechanics and dietary ecology of Paranthropus boisei.

The African Plio-Pleistocene hominins known as australopiths evolved derived craniodental features frequently interpreted as adaptations for feeding on either hard, or compliant/tough foods. Among australopiths, Paranthropus boisei is the most robust form, exhibiting traits traditionally hypothesized to produce high bite forces efficiently and strengthen the face against feeding stresses. However, recent mechanical analyses imply that P. boisei may not have been an efficient producer of bite force and that robust morphology in primates is not necessarily strong. Here we use an engineering meth
