Treatment retention in a specialized alcohol programme after an episode of alcoholic hepatitis: Impact on alcohol relapse.

AIMS: Alcoholic hepatitis (AH) is a life-threatening complication of alcohol use disorder (AUD). Alcohol abstinence is the main predictor of the long-term prognosis of AH. It is unknown whether AUD treatment retention (TR) after an AH episode impacts alcohol relapse and mortality or what baseline factors influence TR. METHODS: Design: case-control study; Study population: hospitalized patients (1999-2012) with an episode of biopsy-proven AH were included (n = 120); Assessment: demographic and clinical data, the High-Risk Alcoholism Relapse (HRAR) scale, mortality and alcohol relapse were asse

Influence of alcohol use and family history of alcoholism on neural response to alcohol cues in college drinkers.

BACKGROUND: Heavy drinkers show altered functional magnetic resonance imaging (fMRI) response to alcohol cues. Little is known about alcohol cue reactivity among college age drinkers, who show the greatest rates of alcohol use disorders. Family history of alcoholism (family history positive [FHP]) is a risk factor for problematic drinking, but the impact on alcohol cue reactivity is unclear. We investigated the influence of heavy drinking and family history of alcoholism on alcohol cue-related fMRI response among college students. METHODS: Participants were 19 family history negative (FHN) li

Prenatal alcohol exposure, blood alcohol concentrations and alcohol elimination rates for the mother, fetus and newborn.

Fetal alcohol spectrum disorders (FASDs) are a common cause of intellectual impairment and birth defects. More recently, prenatal alcohol exposure (PAE) has been found to be a risk factor for fetal mortality, stillbirth and infant and child mortality. This has led to increased concern about detection and management of PAE. One to 2 h after maternal ingestion, fetal blood alcohol concentrations (BACs) reach levels nearly equivalent to maternal levels. Ethanol elimination by the fetus is impaired because of reduced metabolic capacity. Fetal exposure time is prolonged owing to the reuptake of amn

Neonatal alcohol exposure produces hyperactivity in high-alcohol-sensitive but not in low-alcohol-sensitive rats.

Children of women who consume high amounts of alcohol during their pregnancies vary greatly in physical and behavioral outcomes. Although many factors, such as dose and timing of exposure, undoubtedly contribute to this variation, one important determinant may be genetic differences in the response to alcohol. The present study examined activity levels in high alcohol sensitivity (HAS) and low alcohol sensitivity (LAS) rats following neonatal alcohol exposure. These lines were selectively bred for extremes in ethanol-induced "sleep times." The HAS and LAS offspring were exposed to alcohol via

Dual-Functional Reductant/Copper Catalytic System for Aerobic Oxidation of Alcohols and N-Heterocycles without an External Base.

This work developed a practical and highly efficient Cu(II) catalytic system for the aerobic oxidation of alcohols and the dehydrogenative aromatization of N-heterocycles. This approach overcomes the reliance of traditional Cu(II) catalysts on strong bases. We designed a new N,N,O-tridentate Cu(II) complex (Cat 2), based on the concept of simplifying the catalytic system, and synthesized a dual-functional reductant CoBr2(NMI)2, which combines both reducing and coordination ability. Under mild conditions, this system employs air or O2 as the oxidant and efficiently catalyzes the transformation …

Reaction-Coordinate Analysis of DMAP-Mediated Allylation of Allylic Alcohols: Coupled Proton Transfer and C-C Bond Formation.

Density functional theory calculations reveal that the DMAP-mediated allylation of cyclic Baylis-Hillman alcohols proceeds through a concerted substitution pathway in which proton transfer is directly coupled to bond reorganization, as an alternative to stepwise pathways involving discrete ionization. Although formation of an allylic cation has been invoked to rationalize this transformation, the energetic feasibility of ionization under neutral conditions has not been explicitly examined. Intrinsic reaction-coordinate (IRC) analysis provides direct insight into how proton transfer and bond re…

Hydrogen bonding implications on sweetness behavior of sugar alcohols in food model systems: A thermophysical and FTIR study.

Sugar alcohols play an important role in the pharmaceutical and food industry due to their dominant effect on blood glucose levels and are used as artificial sweeteners in dairy products. The present study was conducted to explore the hydrogen bonding effect and sweetness response on structure-based properties of sugar alcohols (lactitol) in aqueous dimethyl sulfoxide (DMSO) using volumetric, acoustic, and FTIR techniques to get better considerations about mechanisms of sweet taste chemoreception and give valuable information about lactitol stabilization in the presence of DMSO, and solute-sol…

Structure-Guided Engineering of Alcohol Dehydrogenase for the Stereodivergent Synthesis of Amino Alcohols via Desymmetrization of Cyclic Diketones.

Chiral cyclic vicinal amino alcohols constitute essential structural motifs in numerous pharmaceutical agents, yet their asymmetric synthesis from readily available symmetric precursors remains a formidable challenge due to difficulties in controlling stereochemistry and preventing over-reduction. Herein, we report a stereodivergent, one-pot dual-enzyme cascade that successfully overcomes these limitations to access all four stereoisomers of 2-aminocyclohexanol from 1,2-cyclohexanedione. Central to this strategy is the development of a "rudder" model-guided protein engineering approach, which …

Chemodivergent Syntheses of Pyrroles and 1-Pyrrolines from β-Amino Alcohols and Ketones Employing ADC and BH Strategies.

We report an iridium-catalyzed chemodivergent strategy for the selective syntheses of pyrroles and 1-pyrrolines from β-amino alcohols and ketones by fine-tuning the catalyst loading and base under acceptorless dehydrogenative coupling (ADC) and borrowing hydrogen (BH) pathways. Whereas previous transition-metal-catalyzed ADC and BH protocols favor aromatized pyrroles due to their thermodynamic stability, our method uniquely enables controlled access to 1-pyrroline by suppressing aromatization. Mechanistic studies indicate that imine formation, Ir-mediated dehydrogenation, intramolecular cyclod…

Dynamic Kinetic Asymmetric Allylation of Ketones: Stereodivergent Synthesis of E- or Z-Substituted α,β-Stereogenic Tertiary Alcohols.

Despite the widespread occurrence of enantiomerically enriched tertiary alcohols in bioactive molecules and pharmaceuticals, efficient methods for preparing optically pure α,β-stereogenic tertiary alcohols remain scarce. Here, we report a dynamic kinetic asymmetric allylation of readily available racemic ketones with 2-aza-1,4-dienes. Notably, through the judicious selection of chiral ligands for copper catalysts, both E- and Z-isomers can be accessed with high efficiency and stereocontrol. Density functional theory (DFT) calculations reveal that ligand-controlled face-selective attacks via di…

Epoxy ester-orthoester rearrangement as a key step in the method of indirect dihydroxylation of monoterpenoid homoallyl alcohols.

Polyhydroxy containing compounds, in particular those derived from monoterpenoids, have attracted attention as biologically active compounds, but a key limitation in their synthesis is the need for regio- and stereoselective hydroxylation. Here, the synthesis of two dihydroxy derivatives and potential metabolites of the antiparkinsonian agent Prottremine, which is a monoterpenoid diol with a para-menthene scaffold, was carried out using an epoxy ester-orthoester rearrangement as the key step. The isomerization of epoxyacetate derivative of Prottremine to corresponding orthoester proceeded ster

Engineering an AlkS-PalkB Transcription Factor-Based Biosensor With Improved Sensitivity to Isobutanol and Other Short-Chain Alcohols.

Isobutanol, a promising biofuel with higher energy content than ethanol, presents a sustainable alternative through biosynthesis. However, e … ction with high sensitivity, the engineered biosensor retains responsiveness to several short-chain alcohols. This biosensor provides a foundation for high-throughput screening of isobutanol and other short-chain alcohol-producing strains, though additional improvements in selectivity and operating range may be required for efficient implementation.

Chromium-catalyzed sustainable C-C and C-N bond formation: C-alkylation and Friedländer quinoline synthesis using alcohols.

The synthesis of a novel phosphine-based pincer chromium(II) complex CrCl2(PONNH) (Cr-1) is reported in this study. The complex exhibited promising catalytic performance in C-C and C-N bond formation using the borrowing hydrogen methodology. Cr-1 catalyzed the α-alkylation of ketones using primary alcohols as alkyl surrogates in the presence of catalytic amount of a base. Cr-1 was also found to catalyze the β-alkylation of secondary alcohols using primary alcohols. In addition, the dehydrogenative annulation of 2-aminobenzyl alcohols with ketones to form quinolines was achieved using Cr-1 as t…

External ligand-free nickel-catalyzed atom-economical coupling of isatins with allyl alcohols via a redox-relay strategy.

Herein, we present a redox-relay strategy that enables the nickel-catalyzed, atom-economical coupling of isatins and allyl alcohols. This reaction is notable for its lack of requirement for additional ligands or reducing agents, and minimal catalyst usage (only 1%), thus meeting the principles of green chemistry. Mechanistic studies have shown that allyl alcohols act as both reductants and precursors for α,β-unsaturated ketones in the reaction.

Cross-Multicarrousel Mechanism and Kinetics of Mn-PNP Catalyzed Diamide Formation from the Dehydrogenation of Alcohols and Diamines.

Herein, a density functional theory based mechanistic and kinetics study of experimentally reported pincer metal complex catalyzed homogenous dehydrogenation of methanol and diamine into diamide is explored. The reaction proceeds through dehydrogenation and hydrogenation reactions for the reversible interconversion between methanol and diamine. The mechanism proceeds via a step-by-step formation of aldehyde, amide, and diamide complexes. The generated formaldehyde reacts with ethylenediamine in the second cycle to make monoamide, which reacts with formaldehyde from the first cycle to produce d

Copper-catalyzed C(sp3)-H alkylation of fluorene with primary and secondary alcohols using a borrowing hydrogen method.

Despite the limited success of copper-catalyzed alkylations, (NNS)CuCl proved to be an effective catalyst for the sp3 C-H alkylation of fluorene with alcohols. Various primary alcohols and challenging secondary alcohols were successfully used. The practical applicability of the method was effectively tested with several post-functionalization reactions. This copper-catalyzed alkylation of fluorene involved a borrowing hydrogen mechanism.

Palladium-Catalyzed Synthesis of β-Alkynyl Ketones via Selective 1,3-Alkynyl Migration of α,α-Disubstituted Allylic Alcohols.

Herein, a palladium-catalyzed 1,3-alkynyl migration of allylic alcohol for the synthesis of β-alkynyl ketone was described. This intramolecular rearrangement reaction demonstrated an enhanced reactivity compared to the traditional intermolecular alkynylation by circumventing the dimerization of alkynes, exhibiting a specific selectivity toward β-alkynyl elimination. Moreover, this reaction featured wide substrate scope, good functional group tolerance, and 100% atom economy.
