Usefulness of Left Ventricular Assist Device in the Recovery of Severe Amphetamine-Associated Dilated Cardiomyopathy.

Exposure to synthetic drugs such as amphetamines may lead to significant consequences on the cardiovascular system. The prognosis of young adults with amphetamine-induced cardiomyopathy remains poor. We present 2 young patients (aged <40 years) who developed severe dilated cardiomyopathy after chronic amphetamine use. Thorough psychological evaluations demonstrated favorable features with patients being reliable and committed to their health problems. A left ventricular assist device (HeartMate II; Abbott, Chicago, IL) was implanted in the patients shortly after admission to optimize hemodynam…

Gender Effects in the Efficacy of Racemic Amphetamine Sulfate in Children with Attention-Deficit/Hyperactivity Disorder.

INTRODUCTION: A laboratory classroom study in children aged 6-12 years with attention-deficit/hyperactivity disorder (ADHD) found that racemic amphetamine sulfate (RA-AMPH) significantly improved performance versus placebo from 45 min through 10 h post-dose (NCT01986062). A secondary analysis assessed gender as a potential moderator of response to treatment comparing the ADHD Rating Scale-IV (ADHD-RS-IV) and Swanson, Kotkin, Agler, M-Flynn and Pelham (SKAMP) rating scales. METHODS: After 8 weeks of open-label RA-AMPH dose optimization using improvement in ADHD-RS-IV symptoms as a guide, 97 su…

Effect of chronic variable stress on sensitization to amphetamine in high and low sucrose-consuming rats.

BACKGROUND: Individual vulnerability to stress manifests in the interaction of innate properties and environment. There is a growing interes … ucrose consumption to chronic variable stress and the stress effect on behavioural sensitization to amphetamine. METHODS: Male Wistar rats were subjected to a chronic stress regimen and subsequent repeated treatment with amphetamine (1 mg/kg, intraperitoneally). Fifty-kHz ultrasonic vocalizations, locomotor activity and stereotypies were measured. RESULTS: …

Amphetamine Dose-Dependently Decreases and Increases Binge Intake of Fat and Sucrose Independent of Sex.

OBJECTIVE: Amphetamine was formerly used as a treatment to combat obesity, but amphetamine's use as an appetite suppressant was discontinued because of its significant abuse potential. Most of the rewarding and reinforcing effects of amphetamine differ by sex, with females showing higher levels of drug intake and amphetamine-induced motivation, relapse, and locomotion, but it is unknown whether amphetamine's effects on feeding also differ by sex. Furthermore, previous research on the anorectic effects of amphetamine has been focused primarily on its effects on baseline homeostatic feeding, but…

Effects of amphetamine exposure during adolescence on behavior and prelimbic cortex neuron activity in adulthood.

Repeated exposure to psychostimulants during adolescence produces long-lasting changes in behavior that may be mediated by disrupted development of the mesocorticolimbic dopamine system. Here, we tested this hypothesis by assessing the effects of amphetamine (AMPH) and dopamine receptor-selective drugs on behavior and neuron activity in the prelimbic region of the medial prefrontal cortex (PFC). Adolescent male, Sprague-Dawley rats were given saline or 3 mg/kg AMPH between postnatal day (P) 27 and P45. In Experiment 1, locomotor behavior was assessed during adulthood following challenges with …

Cocaine‑ and amphetamine‑regulated transcript (CART) is associated with dopamine and is protective against ischemic stroke.

Cocaine and amphetamine‑regulated transcript (CART) is a neuropeptide that can protect brains against ischemic injury. The aim of the present study was to investigate the effects of the CART within ischemic stroke and it possible mechanism. The expression levels of dopamine (DA) and CART in ischemic brain tissues of mice were measured following middle cerebral artery occlusion (MCAO). After receiving the treatment of DA and CART, the infarct volume of brain was measured in mice with MCAO. In addition, the function and potential mechanism of CART in ischemic stroke were further investigated. DA…

Dopamine and acetylcholine release sites are enriched in the striatum a … cholinergic modulation of projection neurons is blocked in brain slices taken from mice exposed to amphetamine and engaged in amphetamine-induced stereotypy, and lacking responsiveness to salient cues. Our findings support a model whereby activity in striosomes is normally under strong regulation by cholinergic interneurons, favoring behavioral flexibility, …

Sodium butyrate reverses the inhibition of Krebs cycle enzymes induced by amphetamine in the rat brain.

There is increasing interest in the possibility that mitochondrial impairment may play an important role in bipolar disorder (BD). The Krebs cycle is the central point of oxidative metabolism, providing carbon for biosynthesis and reducing agents for generation of ATP. Recently, studies have suggested that histone deacetylase (HDAC) inhibitors may have antimanic effects. The present study aims to investigate the effects of sodium butyrate (SB), a HDAC inhibitor, on Krebs cycle enzymes activity in the brain of rats subjected to an animal model of mania induced by D-amphetamine (D-AMPH). Wistar …

Clozapine pre-treatment has a protracted hypolocomotor effect on the induction and expression of amphetamine sensitization.

Amphetamine locomotor sensitization is an animal model for the study of addiction and schizophrenia. The antipsychotic clozapine blocks the hyperlocomotion induced by an acute injection of amphetamine, but its effect on locomotor sensitization after repeated amphetamine administration remains unknown. In the present study we investigate the effect of repeated administration of clozapine on the induction and expression of amphetamine locomotor sensitization. We propose that repeated administration of clozapine blocks the induction and expression of amphetamine sensitization. Male Sprague-Dawley…

Differential effects of amphetamine and haloperidol on temporal reproduction: dopaminergic regulation of attention and clock speed.

Healthy volunteers were tested on 7-s and 17-s peak-interval timing procedures following d-amphetamine (20mg-oral), haloperidol (2mg-oral), and placebo treatments in order to assess the dopaminergic regulation of temporal processing. Individual differences were observed in the drug effects such that two different patterns of timing behavior emerged. In the first pattern, d-amphetamine produced proportional leftward shifts of the timing functions while haloperidol produced proportional rightward shifts. This symmetrical pattern of results suggests that clock speed is regulated by the effective …

A comparison of amphetamine- and methamphetamine-induced locomotor activity in rats: evidence for qualitative differences in behavior.

RATIONALE: Methamphetamine (METH) is typically characterized as a more potent psychostimulant than amphetamine (AMPH), but few studies have directly compared the effects of these drugs at low, behaviorally activating doses that tend not to produce focused stereotypy. OBJECTIVES: The objective of the study was to compare the effects of AMPH or METH treatment on locomotor activity in an open-field arena, focusing on their ability to produce conditioned locomotor activity, sensitization, and cross-sensitization. MATERIALS AND METHODS: Adult male rats were given AMPH or METH (0.5 or 1.0 mg/kg) f…

Tolerance to amphetamine hypophagia: a real-time depiction of learning to suppress stereotyped movements in the rat.

To analyze how tolerance develops to amphetamine-induced hypophagia, the authors recorded real-time licking responses in rats given chronic injections of the drug and access to milk for 30 min. Initially, licking was greatly reduced and occurred only late in the session. The acquisition of tolerance was characterized by a decrease in the latency to initiate licking, a gradual increase in the number of licks, and a reorganization of the temporal licking pattern such that licks were distributed throughout the session, interspersed with pauses. On post-tolerance dose-response tests, licking was d…

Behavioral sensitization to amphetamine in rats: changes in the rhythm of head movements during focused stereotypies.

RATIONALE: The indirect-acting dopamine agonist amphetamine induces in rats, motor stereotypies that have served as the primary behavioral end points in studies related to schizophrenia, drug abuse, and L-dopa-induced dyskinesias. Despite the experimental importance of this rodent response to this drug, little is known about the rhythmic character of the behavior and how such rhythms change (or sensitize) in response to repeated treatment with amphetamine. OBJECTIVE: To address this lack of quantitative information, the rhythms of focused stereotypies (characterized by a lack of locomotion an…

Cocaine and amphetamine attenuate the discriminative stimulus effects of naltrexone in opioid-dependent rhesus monkeys.

This study tested the hypothesis that stimulants (indirect dopamine agonists) attenuate the discriminative stimulus of naltrexone in monkeys chronically treated with L-alpha-acetylmethadol (LAAM). Four rhesus monkeys (Macaca mulatta) received LAAM (1.0 mg/kg s.c.) twice daily and discriminated a withdrawal-precipitating dose of naltrexone (0.0178 mg/kg s.c.) from saline. Selective involvement of cytochrome P450 2D subfamily in in vivo 4-hydroxylation of amphetamine in rat.

The cytochrome P450 (P450) 2D subfamily catalyzes ring hydroxylation of amphetamines. We tested the hypothesis that P450 2D is selectively involved in amphetamine 4-hydroxylation. Urinary elimination of 4-hydroxyamphetamine and amphetamine was determined in male Sprague-Dawley rats pretreated with P450 inducers and inhibitors. The urinary 24-h metabolic ratio (amphetamine/4-hydroxyamphetamine) was not affected by the inducers 3-methylcholanthrene, isosafrole, phenobarbital, ethanol, pregnenolone-alpha-carbonitrile, and clofibrate. Isosafrole did, however, increase amphetamine elimination along…
