Hyperdiversity and functional potential of the coral mycobiome: characterising host-specific and environmentally driven niches across the South China Sea reefs using long-read 28S rRNA metabarcoding.

Coral-associated fungi are an integral yet understudied component of the coral holobiont. To elucidate their diversity, drivers, and potenti … clustered, representing 16 phyla, 61 classes, 84 orders, 117 families, 156 genera, and 187 species. Ascomycota dominated the fungal communities, followed by Basidiomycota and Chytridiomycota. Phylogenetic analyses revealed a deeply divergent fungal lineage distinct from known phyla. Pending further molecular and morphological validation, this lineage is prov …

Phaeohyphomycosis in a Solid Organ Transplant Recipient: Case Report of a New Pathogenic Species of Kirschsteiniothelia.

Phaeohyphomycoses are rare infections caused by melanin-pigmented, saprophytic fungi commonly found in soil and on decaying plant matter. Infections typically affect immunocompromised individuals and present a wide clinical spectrum. Here, we report the case of a 63-year-old immunocompromised patient, with a history of combined heart-lung transplantation, who consulted in 2022 due to the appearance of a subcutaneous lesion on the thigh. A complete surgical debridement was performed and histopathological examination concluded the presence of a deep fungal infection. Three years later, relapse o

Bio-guided isolation of trichodermin from a native Trichoderma brevicompactum strain with potent antifungal activity against phytopathogenic fungi.

In agriculture, the presence of soil-borne phytopathogenic fungi represents a limiting factor that causes significant crop losses. In this context, Trichoderma brevicompactum represents a viable alternative to chemical fungicides, acting as an effective biological control agent for managing fungal diseases. The objective of this work was to characterize the antifungal activity of a native isolate of T. brevicompactum FCQ18, obtained from soil associated with tomato cultivation, against Macrophomina phaseolina, Rhizoctonia solani, and Sclerotinia sclerotiorum. Direct confrontation assays showed

Tracing the origins of crmA megasynthase through lichen genomes.

The products of isocyanide synthase (ICS) biosynthetic gene clusters (BGCs) have been implicated in microbial interactions, pathogenesis, and metal homeostasis. While several ICS BGCs have been described as mediating metal-associated ecologies, the evolutionary history of these clusters is unexplored among Lecanoromycetes, a clade comprised predominantly of symbiotic, lichen-forming fungi (LFF), which are known to thrive in both metal-contaminated and scarce environments. Analyzing nearly 4,000 fungal genomes, including 90 Lecanoromycetes, we identified a significant 3-fold enrichment of ICS-e

Machine learning identifies novel signatures of antifungal drug resistance in Saccharomycotina yeasts.

Antifungal drug resistance is a major challenge in fungal infection management. Numerous genomic changes are known to contribute to acquired drug resistance in clinical isolates of specific pathogens, but whether they broadly explain natural resistance across entire lineages is unknown. We leveraged genomic, ecological, and phenotypic trait data from naturally sampled strains from nearly all known species in subphylum Saccharomycotina to examine the evolution of resistance to eight antifungal drugs. The phylogenetic distribution of drug resistance varied by drug; fluconazole resistance was wid

The Biological Roles of Class II Hydrophobins at the Cell Wall of Filamentous Ascomycetes.

One of the most elusive aspects of fungal cell wall biology has been dilucidating the specific functions of most cell wall proteins. Class II hydrophobins are small amphipathic proteins produced exclusively by filamentous ascomycetes. A comparative synthesis of their biological functions across species has been lacking for this specific group of proteins. This review examines the available genetic, cell biological, and biochemical evidence on Class II hydrophobins in filamentous ascomycetes, with emphasis on Sordariomycetes, organized by both gene copy number and functional category. Gene dele

Carbonic anhydrases contribute to mitochondrial function, conidial development, and pathogenicity of Magnaporthe oryzae.

Carbonic anhydrases (CAs) are metalloenzymes widely present in biological systems, capable of catalyzing the reversible hydration of carbon dioxide (CO₂) to form bicarbonate (HCO3⁻) and protons (H+). This biochemical reaction plays a crucial role in various physiological processes. Previous studies have shown that MoCA1, a carbonic anhydrase in Magnaporthe oryzae, is localized in the mitochondria and is involved in conidial development and pathogenicity. However, the functions of other carbonic anhydrases in M. oryzae, such as MoCA2, MoCA4, and MoCA6, remain largely unexplored. Through subcell

Use of Basic Substances and Other Natural Compounds as Alternatives to Synthetic Fungicides for the Control of Grapevine Downy and Powdery Mildews.

Consumer demand, retailer involvement, policies to reduce pesticide application, and the growth of organic agriculture are steering research toward safer alternatives like basic substances: compounds used as food, food ingredients, or in medicine with a potential secondary application in plant protection. Downy mildew (DM) and powdery mildew (PM) are major grapevine diseases, and many fungicide applications are required for their management. In this 2-year field trial, chitosan, whey, sweet orange essential oil (EO), Equisetum arvense, Urtica spp., and Ampelomyces quisqualis were tested agains

Spatial Distribution of Astins in Aster tataricus and Their Production by Cyanodermella asteris.

Aster tataricus is a plant used in Traditional Chinese Medicine for the treatment of cough, phlegm, and asthma. Phytochemical studies of A. tataricus have resulted in the isolation of 23 peptides, among which the astins are recognized for their potential application as anticancer drugs. However, it was found that some of the astins, namely, astins C, F and G, are in fact produced by an endophytic fungus, Cyanodermella asteris, isolated from the inflorescences of the plant, while the remainder were suggested to be transformation products of these astins by A. tataricus. Using mass spectrometry
