Activity-dependent Notch signalling in the hypothalamic-neurohypophysial system of adult mouse brains.

Notch signalling has a key role in cell fate specification in developing brains; however, recent studies have shown that Notch signalling also participates in the regulation of synaptic plasticity in adult brains. In the present study, we examined the expression of Notch3 and Delta-like ligand 4 (DLL4) in the hypothalamic-neurohypophysial system (HNS) of the adult mouse. The expression of DLL4 was higher in the supraoptic nucleus (SON) and paraventricular nucleus (PVN) compared to adjacent hypothalamic regions. Double-labelling immunohistochemistry using vesicular GABA transporter and glutamat

Interaction between familial amyotrophic lateral sclerosis (ALS)-linked SOD1 mutants and the dynein complex.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder characterized by progressive motor neuron death. More than 90 mutations … r the disease progression. Motor neurons with long axons are particularly susceptible to defects in axonal transport. Our results demonstrate a direct "gain-of-interaction" between mutant SOD1 and dynein, which may provide insights into the mechanism by which mutant SOD1 could contribute to a defect in retrograde axonal transport or other dynein functions. The abe …

An autoradiographic study of midbrain-diencephalic projections to the inferior olivary nucleus in the opossum (Didelphis virginiana).

Techniques of intra-axonal transport were utlizied to elucidate the organization of diencephalic and midbrain projections to the inferior olivary nucleus of the Virginia opossum. Retrograde transport of horseradish peroxidase injected into the olive suggests that terminals within it arise from the subparafascicular nucleus of the caudal thalamus, the nucleus of Darkschewitsch, the fields of Forel, the interstitial nucleus of Cajal, the periaqueductal grey, the caudal pretectal nucleus, the tegmentum dorsomedial to the red nucleus, the red nucleus (minimal), the nucleus linearis, as well as the…

Loss of Miro1-directed mitochondrial movement results in a novel murine model for neuron disease.

Defective mitochondrial distribution in neurons is proposed to cause ATP depletion and calcium-buffering deficiencies that compromise cell function. However, it is unclear whether aberrant mitochondrial motility and distribution alone are sufficient to cause neurological disease. Calcium-binding mitochondrial Rho (Miro) GTPases attach mitochondria to motor proteins for anterograde and retrograde transport in neurons. Using two new KO mouse models, we demonstrate that Miro1 is essential for development of cranial motor nuclei required for respiratory control and maintenance of upper motor neuro

Ganglioside changes in the regenerating goldfish optic system: comparison with glycoproteins and phospholipids.

Axonally transported radioactivity in sialoglycoconjugates, labeled by intraocular injection of [3H]N-acetylmannosamine, increased significantly during regeneration of goldfish optic axons at 30 degrees C. Ganglioside radioactivity showed the largest increase--approximately eightfold--in the optic nerve tract at 8 days after optic nerve crush while sialoglycoprotein radioactivity increased fourfold under the same conditions. As regeneration proceeded the magnitude of the increase in the nerve tract diminished for both glycoconjugates. In the optic tectum, however, transported radioactivities r

Neuroprotective effect of Sigma-2 modulator CT2074 in a mouse model of ocular hypertension.

Ocular neurodegenerative diseases, particularly glaucoma, represent a significant global cause of blindness, with current therapies inadequately addressing the degeneration of the retina and optic nerve. Recent research has identified the sigma-2 receptors as a potential druggable target to offer neuroprotection in managing ocular neurodegenerative disorders. This study investigates the neuroprotective potential of CT2074, a sigma-2 receptor modulator, in a mouse model of primary open-angle glaucoma. Male mice were subjected to unilateral magnetic bead-induced elevation of intraocular pressure

Increase in dephosphorylation of the heavy neurofilament subunit in the monkey chronic glaucoma model.

PURPOSE: To investigate the phosphorylation of the heavy neurofilament subunit (NF-H), which could be deeply involved in axonal transport of retinal ganglion cells (RGCs), in an experimental glaucoma model of chronic elevation of intraocular pressure (IOP) in monkeys. METHODS: One eye in adult monkeys was randomly selected for laser treatment, and IOP was maintained between 30 and 40 mm Hg throughout the experiment. The eyeballs with the optic nerve and optic chiasm were enucleated as one tissue and were subject to immunocytochemical observation, using two NF-H-specific antibodies, NF-200 and…

Role of HAUS7 as a DOCK3 binding partner in facilitating axon regeneration.

The molecular mechanisms involved in reconstructing the eye-to-brain connection and functional recovery following optic nerve damage remain unclear. This study revealed that HAUS augmin-like complex subunit 7 (HAUS7) is a molecule that binds to dedicator of cytokinesis 3 (DOCK3), a regulator of neurotrophic factor signaling and axon regeneration. We observed a distribution pattern of HAUS7 expression, suggesting that neuronal HAUS7 is transported from the cell body to the growth cone under the control of DOCK3. In addition, phosphorylation of DOCK3 at Y562 by tropomyosin receptor kinase B sign

Acute temperature sensitivity in optic nerve axons explained by an electrogenic membrane potential.

Classical work in squid axon reports resting membrane potential is independent of temperature, but our findings suggest that this is not the case for axons in mammalian optic nerve. Refractory period duration changes over 10 times between 37 °C and room temperature, and afterpotential polarity is also acutely temperature sensitive, inconsistent with changes in temperature impacting nerve function only through altered rates of ion channel gating kinetics. Our evidence suggests that the membrane potential is enhanced by warming, an effect reduced by exposure to ouabain. The temperature dependenc

Assessment of the rate of spinal motor axon regeneration by choline acetyltransferase immunohistochemistry following sciatic nerve crush injury in mice.

OBJECT: The purpose of this study was to examine whether choline acetyltransferase (ChAT) staining can be used for assessing the rate of motor neuron regeneration at an early phase of axon outgrowth. METHODS: The authors developed a new sciatic nerve crush model in adult mice. In this model, in addition to performing a sciatic nerve crush injury, the authors excised the ipsilateral lumbar L3-6 dorsal root ganglion (DRG), which resulted in degeneration of the sensory fibers entering into the sciatic nerve. Crushed nerve sections obtained at Day 3 or Day 7 postinjury were analyzed by means of i

Relationship of axonal voltage-gated sodium channel 1.8 (NaV1.8) mRNA accumulation to sciatic nerve injury-induced painful neuropathy in rats.

Painful peripheral neuropathy is a significant clinical problem; however, its pathological mechanism and effective treatments remain elusive. Increased peripheral expression of tetrodotoxin-resistant voltage-gated sodium channel 1.8 (NaV1.8) has been shown to associate with chronic pain symptoms in humans and experimental animals. Sciatic nerve entrapment (SNE) injury was used to develop neuropathic pain symptoms in rats, resulting in increased NaV1.8 mRNA in the injured nerve but not in dorsal root ganglia (DRG). To study the role of NaV1.8 mRNA in the pathogenesis of SNE-induced painful neur

Activity-induced synaptic capture and exocytosis of the neuronal serine protease neurotrypsin.

Extracellular proteolysis plays an essential role in synaptic remodeling that is indispensable for cognitive function. The extracellular serine protease neurotrypsin was implicated in cognitive function, because humans lacking a functional form of neurotrypsin suffer from severe mental retardation. By immunoelectron microscopy, neurotrypsin has been localized to presynaptic terminals, suggesting a local proteolytic function after its synaptic release. Here, we studied axonal trafficking and synaptic exocytosis of neurotrypsin by live imaging of hippocampal neurons expressing neurotrypsin fused

Imaging the transport dynamics of single alphaherpesvirus particles in intact peripheral nervous system explants from infected mice.

ABSTRACT Alphaherpesvirus particles travel long distances in the axons of neurons using host microtubule molecular motors. The transport dynamics of individual virions in neurons have been assessed in cultured neurons, but imaging studies of single particles in tissue from infected mice have not been reported. We developed a protocol to image explanted, infected peripheral nervous system (PNS) ganglia and associated innervated tissue from mice infected with pseudorabies virus (PRV). This ex vivo preparation allowed us to visualize and track individual virions over time as they moved from the s

Perturbed myelination process of premyelinating oligodendrocyte in Niemann-Pick type C mouse.

Niemann-Pick disease type C (NPC) is an autosomal recessive neurovisceral lipid storage disease caused by a loss of NPCI function, which results in perturbation of intracellular cholesterol transport. In BALB/c npc(nih) mice, the murine ortholog of NPCI gene is mutated. In NPC mouse, hypomyelination is conspicuous in the cerebral white matter and corpus callosum in addition to neuronal storage. However, the pathogenesis on hypomyelination is not well elucidated. We hypothesized that the hypomyelination in NPC mice resulted from either defective differentiation of oligodendrocyte lineage cells

IDPN neuropathy in the cat: coexistence of proximal and distal axonal swellings.

Administration of beta,beta'-iminodipropionitrile (IDPN) to rodents has previously been shown to produce neurofilament-filled axonal swellings in the proximal regions of motor and sensory nerve fibers. Because of the distinctive distribution of these swellings, IDPN has been classed as a proximal axonopathy and thereby distinguished from other disorders in which similar axonal swellings occur in the distal parts of the axon (distal axonopathies). This report describes the pathology in the peripheral nerves of cats which received intermittent injections of IDPN and calls attention to two previo

Collateral sprouting of sensory axons in the glabrous skin of the hindpaw after chronic sciatic nerve lesion in adult and neonatal rats: a morphological study.

The anterograde transport of wheat germ agglutinin-horseradish peroxidase conjugate was used to study the normal distribution of sensory nerve axons in the plantar skin of the rat hindlimb and at various times after chronic sciatic nerve injury in adult and neonatal rats. In adults, thin saphenous nerve axons were found in a small area laterally to the normal saphenous nerve territory 2-24 weeks after sciatic nerve lesion. In neonatal rats, at 6 and 10 weeks after sciatic nerve injury thin saphenous nerve axons were found almost or all over the sole of the foot, respectively, and in all 5 toes
