Chemical, Biological, and Ecological Evidence for Aerobic Deoxynivalenol Detoxification in Agronomic Soil-Derived Bacterial Communities.

Deoxynivalenol (DON) is a prevalent trichothecene mycotoxin in cereals that poses food and feed safety risks while causing important economic losses. Microbial biotransformation offers a selective, mild strategy for DON detoxification. Here, we screened aerobic soil-derived bacterial communities from diverse agricultural environments, using DON as the sole carbon source for this mycotoxin depletion. More than half of the tested enrichment samples showed a reduced DON signal, as observed by HPLC-UV. To assess the biological relevance, culture extracts were tested for cytotoxicity in HepG2 cells…

Genetic algorithm-guided sample design enables efficient machine learning-driven optimization of tauroursodeoxycholic acid production in engineered Escherichia coli.

Data-driven machine learning (ML) approaches offer powerful tools for modeling and optimizing complex fermentation processes, yet their application is often limited by the need for large experimental datasets, which is difficult to meet in low-throughput microbial fermentation systems. Here, we present a practical genetic algorithm (GA)-assisted adaptive sampling strategy to support sample-efficient ML-assisted optimization of microbial biotransformation processes. In this strategy, GA is employed upstream to iteratively select informative experimental conditions and generate a compact dataset…

Investigation of active site amino acid influence on carbon and chlorine isotope fractionation during reductive dechlorination.

Reductive dehalogenases (RDases) are corrinoid-dependent enzymes that reductively dehalogenate organohalides in respiratory processes. By comparing isotope effects in biotically catalyzed reactions to reference experiments with abiotic corrinoid catalysts, compound-specific isotope analysis (CSIA) has been shown to yield valuable insights into enzyme mechanisms and kinetics, including RDases. Here, we report isotopic fractionation (ε) during biotransformation of chloroform (CF) for carbon (εC = -1.52 ± 0.34‰) and chlorine (εCl = -1.84 ± 0.19‰), corresponding to a ΛC/Cl value of 1.13 ± 0.35. Differences in the metabolism and disposition of ursodeoxycholic acid and of its taurine-conjugated species in patients with primary biliary cirrhosis.

The clinical effectiveness of ursodeoxycholate in the treatment of liver disease may be limited by its poor absorption and extensive biotransformation. Because in vitro and in vivo studies suggest that the more hydrophilic bile acid tauroursodeoxycholate has greater beneficial effects than ursodeoxycholate, we have compared for the first time the absorption, metabolism, and clinical responses to these bile acids in patients with primary biliary cirrhosis (PBC). Twelve female patients with PBC were sequentially administered tauroursodeoxycholate and ursodeoxycholate (750 mg/d for 2 months) in a…

Absorption, elimination and metabolism of trichloroethylene: a quantitative comparison between rats and mice.

The absorption, elimination and metabolism of 14C-trichloroethylene (Tri) was studied in adult female Wistar rats and NMRI mice after administration of 200, 20 and 2 mg/kg Tri. Dose-dependent biotransformation of Tri to metabolites was observed in both species. Induction of hepatic mono-oxygenases by phenobarbital or polychlorinated biphenyls resulted in a higher rate of biotransformation after a single oral dose of 200 mg/kg 14C-Tri to rats. An increase in radioactivity covalently bound to liver and kidney macromolecules of induced rats as compared to control rats parallels the toxic effects …

Relationship between chemical structure of antibiotics and pharmacokinetics.

Modifications to the chemical structure of antibiotics can modify nearly all pharmacokinetic parameters: digestive absorption, half-life, protein binding, tissue distribution, metabolic biotransformation, biliary and renal elimination. Examples of increased oral bioavailability are given for beta-lactams, macrolides, tetracyclines, fosfomycin, quinolones and acyclovir. The structural modifications responsible for prolonged or shortened half-life, increased or decreased biliary excretion, increased or decreased renal clearance for the different families of antibiotics are reviewed.

Metabolism of cathinone to d-norpseudoephedrine in humans.

Cathinone, a potent psychostimulant isolated from young shoots of Catha edulis was given to four human volunteers. Examination of urine collected from the volunteers at predetermined intervals showed the presence of unchanged cathinone, d-norpseudoephedrine, and two unidentified basic substances. The observed biotransformation of cathinone to the less potent psychostimulant, d-norpseudoephedrine involves reduction of a ketone group to alcohol, a common metabolic pathway in humans.

Antioxidant Oxidation Leads to Greater Accumulation of Novel Organophosphate Esters (OPEs) than Traditional OPEs in soil and Groundwater.

Government agencies have implemented stringent regulations on directly used organophosphate esters (OPEs), such as triphenyl phosphate (TPhP). However, it remains unclear to what extent NOPEs derived from high production volume Organophosphite antioxidants (OPAs) contribute to contamination and environmental risks in industrial-park soil and groundwater. Here, we integrated field investigations of soil and groundwater from a representative industrial park with soil incubation experiments, in vitro rat liver S9 biotransformation assays, and hazard assessment modeling. Spontaneous formation of highly concentrated phenylacetylcarbinol via in situ phase separation in deep eutectic solvents using Candida magnoliae whole cells.

Deep eutectic solvents (DESs) are promising green media for biocatalysis, yet their application in pyruvate decarboxylase (PDC)-mediated phenylacetylcarbinol (PAC) biotransformation with product separation remains limited. This study screened nine DESs using frozen-thawed whole cells (FT-WHC) of Candida magnoliae (C. mag.), followed by evaluation of water content and DES to phosphate buffer ratios. Glycerol-based DES resulted in significantly higher (p ≤ 0.05) volumetric PDC activity and PAC productivity than other DESs. Choline chloride: glycerol (ChCl: Gly (1:2) 20% (v/v) H2O) achieved the h…

Microbial Modifications of Androstane and Androstene Steroids by Penicillium vinaceum.

The biotransformation of steroid compounds is a promising, environmentally friendly route to new pharmaceuticals and hormones. One of the reaction types common in the metabolic fate of steroids is Baeyer-Villiger oxidation, which in the case of cyclic ketones, such as steroids, leads to lactones. Fungal enzymes catalyzing this reaction, Baeyer-Villiger monooxygenases (BVMOs), have been shown to possess broad substrate scope, selectivity, and catalytic performance competitive to chemical oxidation, being far more environmentally green. This study covers the biotransformation of a series of andr…

The Respective Roles of CYP3A4 and CYP2D6 in the Metabolism of Pimozide to Established and Novel Metabolites.

Pimozide is a dopamine receptor antagonist indicated for the treatment of Tourette syndrome. Prior in vitro studies characterized N-dealkylation of pimozide to 1,3-dihydro-1-(4-piperidinyl)-2H-benzimidazol-2-one (DHPBI) via CYP3A4 and, to a lesser extent, CYP1A2 as the only notable routes of pimozide biotransformation. However, drug-drug interactions between pimozide and CYP2D6 inhibitors and CYP2D6 genotype-dependent effects have since been observed. To reconcile these incongruities between the prior in vitro and in vivo studies, we characterized two novel pimozide metabolites: 5-hydroxypimoz…

Ritualistic use of the holly Ilex guayusa by Amazonian Jívaro Indians.

In Amazonian Peru and Ecuador leaf decoctions of the rainforest holly Ilex guayusa with high caffeine concentrations are used as a morning stimulant. After daily ingestion, ritualistic vomiting by male Achuar Indians, better known as Jívaros, reduces excessive caffeine intake, so that blood levels of caffeine and biotransformed dimethylxanthines do not cause undesirable CNS and other effects. Emesis is learned and apparently not due to emetic compounds.

Bidirectional interactions between curcumin and gut microbiota in transgenic mice with Alzheimer's disease.

Alzheimer's disease (AD) is a neurodegenerative disease with increasing prevalence worldwide, while there are no effective drugs at present. Curcumin, a natural polyphenolic substance isolated from turmeric, is a promising natural compound to combat AD, but its pharmacology remains to be fully understood for its poor in vivo bioavalibility. Inspired by the recently reported associations between gut microbiota and AD development, the present study investigated the interactions of curcumin with gut microbiota of APP/PS1 double transgenic mice from two directions: (i) curcumin influences gut micr
