Hippocampal neural stem cells (NSCs) have attracted significant attention due to their essential role in maintaining cognitive functions, such as memory and spatial orientation through neurogenesis. Cognitive impairment is a common and debilitating complication of traumatic brain injury (TBI), yet its underlying mechanisms remain poorly understood and effective clinical interventions are lacking. In this study, we observed persistent cognitive deficits in a mouse model of TBI, a phenomenon that has been widely documented in previous studies, and importantly, we found that these impairments wer

Brain-stem auditory evoked potentials (BAEPs) were recorded in 12 dead subjects (mean age, 72.6 +/- 14.8 years), 30.6 +/- 19.5 hours (range 9-70) after abolished systemic circulation. Death was due to cardiac failure (n = 10), intracerebral hemorrhage (n = 1) and larynx cancer (n = 1). The presence and amplitude of the stimulus artifact were evaluated. The mean (+/- SD) amplitudes of the stimulus artifact was 0.03 +/- 0.02 microV on the left side and 0.01 +/- 0.02 microV on the right side. These findings in accordance with previous studies on comatose patients and brain dead subjects confirm t

Neuropsychological impairment and pathologically delayed Acoustic Brain stem Responses (ABR) have been found in patients examined after minor head injury. The relation of these alterations with the emergence of post-concussional symptoms is unknown. In this study 27 patients were examined with ABR within 48 hours of a clearly defined head injury and with a complete neuropsychological test battery one month after the trauma. They were checked for postconcussional symptoms such as headache, depression or dizziness one year later. ABR recordings were pathological in four patients. Neuropsychologi…

Bacterial meningitis is a serious infection of the central nervous system (CNS) that occurs after bacteria interact with and penetrate the blood-brain barrier (BBB). The BBB is comprised of highly specialized brain microvascular endothelial cells (BMECs) that function to separate the circulation from the CNS and act as a formidable barrier for toxins and pathogens. Certain bacteria, such as Streptococcus agalactiae (group B Streptococcus [GBS]), possess the ability to interact with and penetrate the BBB to cause meningitis. Modeling bacterial interaction with the BBB in vitro has been limited

In the present study, we investigated the feasibility of using human neural stem cells (NSCs) in the treatment of diffuse central nervous system (CNS) alterations in a murine model of mucopolysaccharidosis VII (MPS VII), a lysosomal storage disease caused by a genetic defect in the beta-glucuronidase gene. An immortalized NSC line derived from human fetal telencephalon was genetically engineered to overexpress beta-glucuronidase and transplanted into the cerebral ventricles of neonatal MPS VII mouse. Transplanted human NSCs were found to integrate and migrate in the host brain and to produce l

In the present study, brain-stem auditory evoked potentials (BAEPs) in 31 children in post-traumatic coma with diffuse brain injury were examined. The BAEPs were recorded in the first 72 h after hospital admission and the findings of 29 patients related to the level of neurological recovery at 6 months after head injury. On the basis of the results, children were divided into three groups: the first consisted of children with bilateral and normal BAEPs (n = 19); the second of those with asymmetrical BAEPs (n = 6); and the third of those in whom BAEPs has disappeared or in whom only responses o

PURPOSE: Reirradiation is increasingly used in children and adolescents/young adults (AYA) with recurrent primary central nervous system tumors. The Pediatric Normal Tissue Effects in the Clinic (PENTEC) reirradiation task force aimed to quantify risks of brain and brain stem necrosis after reirradiation.
METHODS AND MATERIALS: A systematic literature search using the PubMed and Cochrane databases for peer-reviewed articles from 1975 to 2021 identified 92 studies on reirradiation for recurrent tumors in children/AYA. Seventeen studies representing 449 patients who reported brain and brain ste…

Populations of neural stem cells (NSCs) reside in a number of defined niches in the adult central nervous system (CNS) where they continually give rise to mature cell types throughout life, including newly born neurons. In addition to the prototypical niches of the subventricular zone (SVZ) and subgranular zone (SGZ) of the hippocampal dentate gyrus, novel stem cell niches that are also neurogenic have recently been identified in multiple midline structures, including circumventricular organs (CVOs) of the brain. These resident NSCs serve as a homeostatic source of new neurons and glial cells

An agent-based model of therapeutic neural stem cell (NSC) migration is developed and used to predict the migration of NSCs in naïve mouse brain. The model utilizes generalized q-sampling imaging which resolves white matter fibers that cross in the brain and is shown to better account for variations in NSC migration patterns as compared to diffusion tensor imaging. In calibrating the model to experimental data, we show that the model is able to reproduce the spatial distribution of NSCs in the mouse brain. In addition, we show that the spatial distribution of NSCs in the mouse brain is sensiti

Traumatic brain injury poses substantial public health and socio-economic threat globally. Hearing loss is one of the most common sensory complications of TBI. Post-traumatic hearing handicap causes impairment in cognition and communication, which further reduce the quality of life of TBI patients. Treatment option for TBI and concurrent auditory complications are limited. Stem cell-derived exosome therapy has recently proven a versatile and potent treatment. After a meticulous dissection of relevant literature, we present this comprehensive, up-to-date, pathophysiology-specific and translatio

INTRODUCTION: Exosomes are nanoscale extracellular vesicles that widely participate in intercellular communication. An increasing number of studies have reported on the neuroprotective effects of stem cell-derived exosomes in brain diseases through various delivery methods. However, only a few reports are available on the delivery and uptake of stem cell-derived exosomes in the brains of mice of different ages.
METHODS: PKH-26-labelled mesenchymal stem cell-derived exosomes were collected, and their uptake was investigated in the brains of mice aged 2 weeks, 2 months, and >6 months, 24 hours

Neuropathic pain is characterized by hyperalgesia, allodynia or spontaneous pain arising from lesions or pathology in the somatosensory nervous system. Multiple mechanisms contribute to this pain following peripheral nerve and spinal cord injuries. Evidence shows that injury-induced changes in dendritic spine morphology in the dorsal horn may contribute to neuropathic pain presentation. Dendritic spines, critical postsynaptic structures for synaptic transmission, undergo remodelling from filopodia-like structures to mature, mushroom-shaped spines in nociceptive spinal cord regions after injury

Traumatic brain injury (TBI) is associated with life-threatening and permanent disabilities. Given the limited capacity of neurons to regenerate, effective treatments for TBI are lacking. Neural stem cells (NSCs) can differentiate into fully functioning neurons and thus hold promise for TBI treatment. Nonetheless, NSC differentiation and proliferation are slow and inefficient. Studies have shown that piezoelectric stimulation is capable of promoting the differentiation and proliferation of NSCs. Here, we describe barium titanate-reduced graphene oxide (BTO/rGO) hybrid piezoelectric nanosticker

Transplantation of neural stem cells (NSCs) holds promise for repairing traumatic brain injury (TBI) but their therapeutic performance is hindered due to the low efficient differentiation into neurons. Direct injection of differentiation modulators to the lesion site has limited improvement to neuronal differentiation as they tend to diffuse or be degraded. In the present study, we report a simple and versatile strategy to engineer the NSCs with a micropatch to improve their therapeutic performance in TBI treatment. The micropatches are fabricated through microcontact printing technique and ca
