Continuous monitoring with microfabricated capillary electrophoresis chip devices.

We report on the direct coupling of hydrodynamically flowing stream to a microchip capillary electrophoresis (CE) for continuous assays of liquid samples. The new interface relies on mounting the sample tubing onto a sharp inlet tip and allows rapid, convenient and reproducible electrokinetic loading from a continuously flowing stream directly into the narrow separation microchannel. The sharp inlet interface is characterized by its efficiency, stability and simplicity. The effect of the sample flow rate, applied voltages and other relevant variables, is described. It was found that the peak i

Analysis of methylene blue in human urine by capillary electrophoresis.

A capillary electrophoresis method for the determination of the dye methylene blue (tetramethylthionine, MB) in human urine depending on liquid/liquid-extraction and diode array detection has been developed, validated, and applied to samples of healthy individuals, who had been dosed with methylene blue within clinical studies. After extraction with dichloromethane and sodium hexanesulfonate, sample extracts were measured on an extended light path capillary. The dye was detected simultaneously at 292 and 592 nm using methylene violet 3 RAX as internal standard. The limit of quantification was

Capillary zone electrophoresis for the estimation of transdermal iontophoretic mobility.

The objective of the study was to investigate the relationship between transdermal iontophoretic flux--specifically, the electromigratory component--and electrophoretic mobility as determined by capillary zone electrophoresis (CZE). First, the steady-state iontophoretic transport rates of a series of dipeptides across porcine skin were determined in vitro. Co-iontophoresis of acetaminophen was used to quantify the respective contributions of electroosmosis (EO) and electromigration (EM). Second, the electrophoretic mobilities of the dipeptides and three other cationic drugs (lidocaine, propran

Microchip capillary electrophoresis with solid-state electrochemiluminescence detector.

We report microchip capillary electrophoresis (CE) coupling to a solid-state electrochemiluminescence (ECL) detector. The solid-state ECL detector was fabricated by immobilizing tris(2,2'-bipyridyl)ruthenium(II) (TBR) into an Eastman AQ55D-silica-carbon nanotube composite thin film on an indium tin oxide (ITO) electrode. After being made by a photolithographic method, the surface of the ITO electrode was coated with a thin composite film through a micromolding in capillary (MIMIC) technique using a poly(dimethylsiloxane) (PDMS) microchannel with the same pattern as an ITO electrode. Then the T

Continuous-flow capillary assay for measuring bacterial chemotaxis.

Bacterial chemotaxis may have a significant impact on the structure and function of bacterial communities. Quantification of chemotactic motion is necessary to identify chemoeffectors and to determine the bacterial transport parameters used in predictive models of chemotaxis. When the chemotactic bacteria consume the chemoeffector, the chemoeffector gradient to which the bacteria respond may be significantly perturbed by the consumption. Therefore, consumption of the chemoeffector can confound chemotaxis measurements if it is not accounted for. Current methods of quantifying chemotaxis use bac

A simple and low-cost electrochemiluminescence detector for capillary electrophoresis.

A simple and cost-effective electrochemiluminescence (ECL) detector for capillary electrophoresis (CE) has been developed. The detector was constructed by vertically gluing a 0.5 mL plastic sample vial onto a piece of 1.5 cm x 1 cm x 0.6 mm indium/tin oxide (ITO)-coated glass plate. End-column ECL detection was performed in a wall-jet configuration. Potential control of the ITO electrode was provided using a direct current (DC) battery. Tris(2,2'-bipyridyl)ruthenium(III) (Ru(bpy)3(3+))-based ECL reaction was used for sensitive detection of four trialkylamines (trimethylamine, triethylamine, tr

Capillary electrophoresis in diagnosis and monitoring of adenosine deaminase deficiency.

BACKGROUND: The diagnosis and monitoring of severe combined immunodeficiency disease (SCID) attributable to adenosine deaminase (ADA) deficiency requires measurements of ADA, purine nucleoside phosphorylase (PNP), and S-adenosyl-L-homocysteine-hydrolase (SAHH) activity and of deoxyadenosine metabolites. We developed capillary electrophoresis (CE) methods for the detection of key diagnostic metabolites and evaluation of enzyme activities. METHODS: Deoxyadenosine metabolites were separated in 30 mmol/L sodium borate-10 mmol/L sodium dodecyl sulfate (pH 9.80) at 25 degrees C on a 60-cm uncoated

The influence of HES on the filtration properties of capillary membrane plasmaseparation.

Plasmaseparation is a treatment under discussion for critically ill patients, especially in sepsis and multiorgan failure. These patients receive a variety of different fluid substitutes, including hydroxyethylstarch (HES). HES is known to influence rheological properties, but nothing is known about the possible interactions between HES and the plamaseparation procedure. We used an in vitro plasmaseparation circuit with heparinized porcine blood. Before priming the system, 2 liters of blood were supplemented by adding 100 ml of either NaCl 0.9% or HES (n=6 in each group). We monitored the tran

Mobilization of small DNAPL pools formed by capillary entrapment.

We performed an experimental study to quantify the critical conditions for the mobilization of small pools of dense nonaqueous phase liquids (DNAPLs) that may form at capillary-heterogeneity boundaries. A series of experiments were conducted in columns packed with uniform sands arranged to create capillary heterogeneities. DNAPL pools readily formed in these packings and were more easily mobilized than trapped DNAPL ganglia. A model was developed to describe the critical conditions for DNAPL pool mobilization. Pool mobilization was expected when a dimensionless pool trapping number N(T)p> 1 wh

Analysis of doxycycline by capillary electrophoresis. Method development and validation.

An optimized capillary electrophoresis method for the analysis of doxycycline is described. The influence of methanol as organic modifier, buffer pH, buffer concentration, capillary length, column temperature, Triton X-100 and methyl-beta-cyclodextrin was systematically investigated. A central composite design was performed in order to optimize the method. The optimal separation conditions were: capillary, uncoated fused-silica [40 cm (32 cm effective length) x 50 microm I.D.]; background electrolyte, a solution of 145 mM sodium carbonate and 1 mM EDTA brought to pH 10.3-methanol (89:11, v/v);

[Separation and determination of nycodenz and its impurities by capillary electrophoresis].

Separation and determination of nycodenz (a new radiography reagent) and its impurities by capillary electrophoresis was investigated. Since the analytes were polyols, Na2B4O7-H3BO3 was chosen as the buffer system. The analytes were negatively charged by complexing with borate. Then cetyltrimethylammonium bromide was used to reverse electroosmotic mobility in order to shorten the analysis time. 2-Propanol was added to optimize the separation further. Acceptable results of linearity, precision of corrected peak area and migration time were obtained. The RSD(n = 6) of corrected peak area and mig

Pulmonary capillary blood volume in hyperpnea-induced bronchospasm.

Reactive hyperemia of the bronchial circulation has been postulated to contribute to the airway narrowing that occurs following exercise or hyperpnea in subjects with asthma with hyperpnea-induced bronchospasm (HIB). Changes in lung parenchymal mechanics also occur in HIB, including increases in peripheral airway resistance. Since the peripheral airways and lung parenchyma are supplied by the pulmonary circulation, and changes in the pulmonary circulation could alter airway resistance or tissue mechanics, we hypothesized that pulmonary capillary blood flow would increase in association with HI
