The chromosomes of Rodentolepis nana (Siebold, 1852) Spasskii, 1954 obtained from naturally infected mice conventionally maintained in a Braziuan laboratory animal house.

The karyotype of Rodentolepis nana obtained from mice in Rio de Janeiro, Brazil, was described. The diploid chromosome number obtained by the division of embryonic cells was 2n = 12. The first and the third pairs presented subterminal centromeres and the other pairs were all acrocentric. The studied species differed in chromosome morphology when compared to previous description by Mutafova and Gergova (1994) in Bulgaria, suggesting an intraspecific variation.

Roberts syndrome is caused by mutations in ESCO2, a human homolog of yeast ECO1 that is essential for the establishment of sister chromatid cohesion.

Roberts syndrome is an autosomal recessive disorder characterized by craniofacial anomalies, tetraphocomelia and loss of cohesion at heterochromatic regions of centromeres and the Y chromosome. We identified mutations in a new human gene, ESCO2, associated with Roberts syndrome in 15 kindreds. The ESCO2 protein product is a member of a conserved protein family that is required for the establishment of sister chromatid cohesion during S phase and has putative acetyltransferase activity.

Chromosome alterations in cancer development and apoptosis.

Normal somatic cells are programmed to die (or undergo apoptosis) whereas cancer cells program themselves to survive. Some of the cytogenetic alterations that might be involved in apoptosis and continuous cell proliferation of normal and cancer cells such as shortening of telomeres, formation of dicentric chromosomes, transfer of telomeric DNA to the homologous chromosomes, malfunction (inactivation) of the centromeres, endoreduplication of chromosomes and other structural and numerical chromosome abnormalities are discussed here.

Centric fission of chromosome 7 with 47,XX,del(7)(pter----cen::q21----qter)+cen fr karyotype in a mother and proximal 7q deletion in two malformed newborns.

In the present paper we report a characteristic pattern of external and internal malformations in two male siblings with proximal 7q interstitial deletion as the unbalanced product of a rearranged chromosome 7 in the mother with karyotype 47,XX,del(7)(pter----cen::q21----qter),+fr. The interstitial 7q deletion in the mother included centromeric fission, break at 7q21 and preservation of the proximal q arm fragment.

Combining multi-antigenic immunodot with indirect immunofluorescence on HEp-2 cells improves the diagnosis of systemic sclerosis.

Systemic sclerosis (SSc) is associated, in nearly all patients, with autoantibodies (Ab). Accordingly, and in order to identify major (anti-CEN A/B and anti-Topo I) but also minor Abs, the usefulness of combining indirect immunofluorescence (IIF) on HEp-2 cells with an 11 multi-antigenic SSc immunodot was explored. 1689 samples tested at the request of clinicians, were evaluated retrospectively. The positivity rate was 28.8% and the diagnosis of SSc was supported for 232 samples. Two groups of Abs were considered: group 1, Abs (anti-CENP A/B, anti-Topo I) present at elevated levels in SSc pati

Coexistence of anti-topoisomerase I and anticentromere antibodies in a patient with systemic sclerosis. Efficacy of treatment combining rituximab and nintedanib. A case report.

In the diagnostic of systemic sclerosis (SSc), both anti-centromere (ACA) and anti-topoisomerase I (ATA) antibodies are considered mutually exclusive, though their coexistence has been also reported in some patients. Notably, nintedanib has been approved for the treatment of interstitial lung disease associated to SSc. Herein, we present the clinical case of a 41-year-old woman with SSc who shows an immunological seroconversion (from ACA positivity to a coexistence of ACA and ATA antibodies) together with changes in her clinical phenotype. Besides, the patient responds positively to the treatm…

Complete monosomy mosaic of chromosome 21: case report and review of literature.

Complete monosomy mosaic of chromosome 21 is a rare disorder. The syndromic features are highly variable. This study describes a girl of Mexican origin with complete monosomy 21 in mosaicism with novel findings, including cortical atrophy, macrostomia, pectum excavatum and immune deficiencies. Parental karyotypes were normal. FISH analysis with probes from 21q22.1-q22.2 region and centromere of X DNA probe was performed on peripheral blood lymphocytes whereas 21q22.1-q22.2 and 21q, 4p, 4q subtelomeric DNA probes were tested in fibroblasts. We propose that the monosomy 21 mosaicism is the cause…

[Lampbrush chromosomes of the Japanese quail (Coturnix coturnix japonica): a new version of cytogenetic maps].

Avian oocyte chromosomes are transfomed into giant transcriptionally active lampbrush chromosomes (LBCs) at meiosis 1 diplotene. These chromosomes are a convenient tool for high-resulution cytogenetic analysis. Using differential staining with fluorochromes DAPI and CMA3, we have constructed detailed cytological maps for lampbrush macrochromosomes 1-5 and ZW of the Japanese quail Coturnix coturnix japonica. We also performed a comparative analysis ofmitotic chromosomes and LBCs corresponding to them. We estimated the decondensation coefficient during LBC formation and determined the centromere…

New insights into the pathogenesis of systemic sclerosis.

Systemic sclerosis (SSc) is a connective tissue disorder characterized by vascular abnormalities and excessive collagen synthesis. Extracellular matrix overproduction by fibroblasts results from abnormal interactions among endothelial cells, mononuclear cells (lymphocytes and monocytes) and fibroblasts, in a setting of vascular hyperreactivity and tissue hypoxia. Many autoantibodies have been identified in the sera of SSc patients; some of them are specific to the disease, such as anti-centromere antibodies in limited SSc, anti-topoisomerase 1 and anti-RNA polymerase I/III antibodies in diffus…

Dicentric Y chromosome in azoospermic males.

Two azoospermic, infertile men with a pseudodicentric Y chromosome are reported. The small isodicentric Y chromosomes were composed of duplicated short arm and proximal long arm Y, as proven by fluorescence in situ hybridisation using a Y centromere-specific DNA probe, pDP97, and a short arm probe pY-80. Both lacked germinal cells in the gonads. It was assumed that the azoospermia was caused by deletion or disruption of the azoospermic factor gene located at distal Yq11. Patient 2 measured 147 cm (-4.1 SD) in height and so it was assumed that he had also lost the "statural determinants" gene.

Inheritance of the Robertsonian translocation (1/21) in the Holstein-Friesian cattle. I. Chromosome analysis.

A new type of Robertsonian translocation (1/21) was found in Holstein-Friesian breed, and transmitted through the three generations, namely from the bull to 6 out of 11 daughters and 4 out of 7 grand-daughters/sons. Although the father of the bull was not available for cytogenetical studies, it was ascertained that 5 half-sibs of the bull had a normal karyotype. By C-banded karyotypes, the 9 translocation carriers within the three generations had a dicentric centromere in the translocated chromosome. It seems that no morphological effects have been noted to be associated with the translocation…

Structural chromosome abnormalities in Down syndrome: a study of two families.

Two families were ascertained through a proband with Down syndrome and a structural rearrangement involving two chromosomes 21. It is suggested that in one patient the chromosome is an isochromosome formed by misdivision of the centromere of a maternal telocentric chromosome 21 and that in the other a Robertsonian translocation involving chromosome 21 was inherited from the mother, who is a 46,XX/46,XX, -21,+t(21q21q) mosaic. The origin of the mosaicism is discussed and considered to be likely to be the result of breakage and reunion at the chromatid, rather than the chromosome, level.

Cytological identification of the chromosome carrying the IXth linkage group (including H-2) in the house mouse.

Mus poschiavinus x M. musculus hybrids, which had seven metacentric chromosomes derived from the poschiavinus complement, were repeatedly backcrossed to M. musculus and selected for the chromosome carrying the H-2 complex. A line called T1/Klj was established which had one metacentric chromosome. It was shown by linkage tests and by cytogenetic studies that one arm of this metacentric chromosome corresponds to the M. musculus acrocentric chromosome carrying linkage group IX, in which the H-2 complex is found. The distance from the H-2 locus to the centromere was tentatively estimated as 14 map…

Heteromorphic XY chromosomes with pericentromeric recombination suppression in Hippophae (Elaeagnaceae).

Among the relatively few flowering plants with strongly heteromorphic XY chromosomes are dioecious species of Hippophae (Elaeagnaceae). To u … (S2, 1.9-2.5 Ma), the latter with more species-specific inversions. The SLRs coincide with the pericentromere regions, which already in the ancestor of Hippophae were greatly enlarged by the insertion of another chromosome. Dosage compensation occurs primarily in floral tissues and mainly through downregulation of female X-specific genes, balancing expressi …

A working model for the formation of Robertsonian chromosomes.

Robertsonian chromosomes form by fusion of two chromosomes that have centromeres located near their ends, known as acrocentric or telocentric chromosomes. This fusion creates a new metacentric chromosome and is a major mechanism of karyotype evolution and speciation. Robertsonian chromosomes are common in nature and were first described in grasshoppers by the zoologist W. R. B. Robertson more than 100 years ago. They have since been observed in many species, including catfish, sheep, butterflies, bats, bovids, rodents and humans, and are the most common chromosomal change in mammals. Robertson…

Clinical relevance of specific and non-specific autoantibodies in systemic sclerosis without overlap features.

OBJECTIVES: The diagnosis of systemic sclerosis (SSc) relies on specific autoantibodies, but the clinical value of non-specific and other immune-mediated inflammatory disease (IMID)-associated antibodies in patients without overlap features is unclear. This study determines the frequency and clinical relevance of a broad autoantibody panel in this well-defined SSc cohort. METHODS: This is a cross-sectional study including 263 consecutive SSc patients without overlap syndromes. A panel was tested for anti-Scl70, anti-centromere, anti-RNApol-III, anti-PM-Scl, anti-dsDNA, anti-Sm, anti-Ro52, ant…

CENPF as an independent prognostic and metastasis biomarker corresponding to CD4+ memory T cells in cutaneous melanoma.

Owing to recent advances in immunotherapies, the overall survival of patients with skin cutaneous melanoma (SKCM) has increased; however, the 5-year survival rate of metastatic patients remains poor. Skin cutaneous melanoma-upregulated genes were screened via analysis of differentially expressed genes (GSE3189 and GSE46517), and metastasis-related oncogenes were identified via weighted gene coexpression network analysis of the GSE46517 dataset. As confirmed by the Tumor Immune Estimation Resource, we found highly expressed centromere protein F (CENPF) in SKCM and its metastases. Immunostaining…
