Nanoparticle delivery enhances the immunomodulatory efficacy of dietary methionine in broiler chickens.

Methionine, an essential sulfur-containing amino acid, is fundamental for humoral immunity through its roles in immunoglobulin synthesis and as a precursor to the intracellular antioxidant glutathione. Enhancing the bioavailability of such key nutrients is a significant goal in nutritional immunology. This study investigated whether nanotechnology could enhance the immunomodulatory effects of methionine by comparing the efficacy and relative bioavailability (RBV) of conventional DL-methionine (DL-Met) with a novel nanoparticle-conjugated methionine (nano-Met) in an avian model. The humoral imm

Infection Dynamics and Host Biomarker Identification for Spotty Liver Disease in Chickens.

Spotty liver disease (SLD), primarily caused by Campylobacter hepaticus and (less frequently) by Campylobacter bilis, significantly impacts commercial layer hens by causing liver lesions, reducing egg production, and increasing mortality, meaning it can result in serious economic losses for farmers. This study explored the relationship between infection, liver dysfunction, and reproduction, aiming to identify host genetic markers for tracking SLD progression. Hens were orally inoculated with the C. hepaticus strain NSW44L and monitored over a seven-day period. Pathogen colonisation was quantif

Protocol for virome characterization in low-volume respiratory samples from broiler chickens.

The poultry industry is a major global source of animal protein but remains vulnerable to immunosuppressive viral infections that compromise bird health and productivity. This study evaluated five viral purification methods for metagenomic analysis of respiratory samples from broiler chickens in Santa Catarina, Brazil. Tracheal swabs from ten flocks (one per farm) were pooled, and 50 µL of a herpes simplex virus type 2 (HSV-2) and murine norovirus (MNV-1) mix was added as an internal positive control. The sample was centrifuged (2000 × g for 30 min), filtered (0.45 μm), and subjected to five p…

Serological and molecular detection of Toxoplasma gondii in chickens in southern Syria.

Toxoplasma gondii is a foodborne protozoan parasite that infects all warm-blooded animals and humans worldwide. Poultry become infected with T. gondii oocysts by ground feeding, and humans contract toxoplasmosis by the consumption of raw or undercooked infected meat. In Syria, no research has been documented regarding T. gondii infection in chickens; therefore, this study was carried out to determine the infection rate in chickens using serological and molecular assays. A total of 59 birds (50 females and 9 males) including free range (FR) and caged chickens were purchased from the southern go…

Mitochondrial dysfunction is a hallmark of woody breast myopathy in broiler chickens.

The woody breast (WB) myopathy poses significant economic and welfare concerns to the poultry industry, however, there is no effective strategy to mitigate this pathology due to its unknown etiology. After showing previously that hypoxia is a key factor in WB progression, we used here various techniques demonstrating dysregulated mitochondria (morphology, biogenesis, tethering, function, and bioenergetics) in WB-affected muscles and in hypoxic myoblasts compared to healthy tissues and normoxic cells, respectively. The increased levels of calcium (Ca2+) in both WB-affected tissues and hypoxic m

Novel recombinant avian infectious bronchitis viruses from chickens in Korea, 2019-2021.

Infectious bronchitis virus (IBV) has evolved through various mutation mechanisms, including antigenic drift and recombination. Four genotypic lineages of IBVs including GI-15, GI-16, GI-19, and GVI-1 have been reported in Korea. In this study, we isolated two IBVs from chicken farms, designated IBV/Korea/289/2019 (K289/19) and IBV/Korea/163/2021 (K163/21), which are two distinct natural recombinant viruses most likely produced by genetic reassortment between the S1 gene of K40/09 strain (GI-19 lineage) and IBV/Korea/48/2020 (GI-15 lineage) in co-infected commercial chickens. Comparative seque…

A copro-antigen ELISA for the detection of ascarid infections in chickens.

A reliable method of diagnosing the most prevalent helminth infections in chickens is vital for developing effective control strategies. Ascaridia galli and Heterakisgallinarum are phylogenetically close nematode species that can elicit the development of cross-reactive antibodies in chickens. Therefore, an enzyme-linked immunoassay (ELISA) based on Ascaridia galli antigens in faeces of chickens to detect and quantify infections with both A. galli and H. gallinarum was developed. The ELISA utilised polyclonal antibodies that were obtained from rabbits immunised with soluble antigens isolated f…
