A comparison of the Yale-Brown Obsessive Compulsive Scale for "heavy drinking" with a single item craving measure: construct validity and clinical utility.

The measurement of alcohol "craving" began with single-item scales. Multifactorial scales developed with the intention to capture more fully the phenomenon of craving. This study examines the construct validity of a multifactorial scale, the Yale-Brown Obsessive Compulsive Scale for heavy drinking (Y-BOCS-hd). The study compares its clinical utility with a single item visual-analogue craving scale. The study includes 212 alcohol dependent subjects (127 males, 75 females) undertaking an outpatient treatment program between 1999-2001. Subjects completed the Y-BOCS-hd and a single item visual-ana…

Comparing the effectiveness of virtual reality vs 2D display-based cue reactivity paradigms to induce nicotine-craving: a behavioral and psychophysiological study.

Craving is a key symptom of nicotine addiction, driving the urge to seek cigarettes. It is often strongly influenced by social and environmental cues associated with the typical use of the substance. In both clinical and laboratory setting, craving can be experimentally triggered using cue reactivity paradigms, where nicotine-related cues are presented to evoke, and then assess, the desire to smoke. In the last few years, virtual reality (VR) has begun to attract a lot of interest in recreating realistic simulations to investigate craving through cue-reactivity exposure. However, a direct comp…

Effect of dietary stimulation on acute changes in GLP-1, alcohol responses, craving, and attentional bias: a randomized, placebo-controlled laboratory study.

RATIONALE: Preclinical and clinical findings support the potential for GLP-1 receptor activation to reduce alcohol consumption and reward. Human experimental studies are needed to clarify whether acute changes in endogenous GLP-1 influence alcohol craving and responses to alcohol. Dietary stimulation may present a method to study the effects of endogenous GLP-1 on responses to alcohol. OBJECTIVES: Assess the effects of a dietary manipulation on acute changes in GLP-1 and laboratory responses to alcohol. METHODS: Healthy young adult heavy drinkers (N = 40) were recruited to participate in two…

Profiling gene alterations in striatonigral neurons associated with incubation of methamphetamine craving by cholera toxin subunit B-based fluorescence-activated cell sorting.

INTRODUCTION: In both rats and humans, methamphetamine (Meth) seeking progressively increases during abstinence, a behavioral phenomenon termed "incubation of Meth craving". We previously demonstrated a critical role of dorsal striatum (DS) in this incubation in rats. However, circuit-specific molecular mechanisms in DS underlying this incubation are largely unknown. Here we combined a newly developed fluorescence-activated sorting (FACS) protocol with fluorescence-conjugated cholera toxin subunit B-647 (CTb-647, a retrograde tracer) to examine gene alterations in the direct-pathway (striatoni…

Alterations in glucose and insulin resistance following stress and alcohol cues predict alcohol craving in those with AUD and obesity: A preliminary study.

BACKGROUND: Alcohol use disorders (AUD) and obesity (OB) are highly prevalent and often comorbid. Recent evidence suggests potential mechanistic overlap that may impact treatments such as Glucagon-like Peptide 1 receptor agonists (GLP1-RAs) for AUD. While alcohol intake can affect blood glucose control, and chronic alcohol use increases diabetes risk, the interplay between obesity, metabolic function, and provoked alcohol craving in AUD has not been previously assessed. Thus, we asked whether AUD and AUD + OB showed distinct patterns of fasting glucose, insulin, and insulin resistance (HOMA) r…

Effect of stress on the relationship between relief-seeking and cue-induced alcohol craving and anxiety: A virtual reality cue exposure study.

OBJECTIVE: Stress is well-known to increase alcohol craving and alcohol cue-induced anxiety in individuals with alcohol use disorder (AUD). While previous research suggests that individuals who drink for relief experience strong alcohol cravings and associated anxiety, the interaction between stress and relief-seeking motives in predicting craving and anxiety responses to alcohol cues is not well understood. Using a virtual reality cue exposure paradigm, this study investigated the moderating role of stress on the relationship between relief-seeking motives and cue-induced alcohol craving and …

A role for the right dorsolateral prefrontal cortex in enhancing regulation of both craving and negative emotions in internet gaming disorder: A randomized trial.

Reward-seeking and relief from negative emotions are two central motivational drives underlying addictions. Impaired executive control over craving and negative emotions contributes to compulsive addictive behaviors. Neuroimaging evidence has implicated the prefrontal cortex (PFC) in regulating craving or emotions. This study aims at examining whether anodal transcranial direct current stimulation (tDCS) over a specific region of the PFC would enhance both regulation processes. Thirty-three men with internet gaming disorder received active (1.5 mA for 20 minutes) and sham tDCS over the right d…
