Cylicospirura wishkai sp. nov. (Spiruroidea: Spirocercidae), a new nematode species in the neotropical felid jaguarundi, Herpailurus yagouaroundi (Carnivora: Felidae).

Cylicospirura is a genus of spirocercid nematodes infecting felids around the globe. Eleven species have been characterised, with C. felineus and C. subaequalis being reported in jaguarundis, Herpailurus yagouaroundi, from Brazil. Herein, we described spirocercid reddish worms collected from three jaguarundis from Costa Rica by using hierarchical clustering and dendrogram plots of morphological characters reported from our specimens and compared to other Cylicospirura spp., histopathological observations of associated lesions, and phylogenetic analyses of both mitochondrial and ribosomal loci.

An ocular cysticercosis case: Caused by Asian genotype of Taenia solium.

An ocular cysticercosis case of a 42-year-old male, who presented with anterior uveitis is being reported. Microscopical examination of the cyst revealed presence of only one hooklet suggestive of T. solium cysticercus. Mitochondrial DNA analysis confirmed it to be T. solium cysticercus of Asian genotype. This is the first report on molecular typing of cysticercus isolate from ocular cysticercosis patient in India. The study suggests that the molecular analysis of cox1 gene may be a useful diagnostic tool in cases where microscopic examination is not confirmatory.

Genetic variations among Echinococcus granulosus isolates in Egypt using RAPD-PCR.

Cystic echinococcosis (CE), caused by hydatid cysts, is a widespread and hazardous disease in humans and animals worldwide. The aim of the current study was to investigate the genetic variations among Echinococcus granulosus cyst strains isolated from sheep, camel, pig, and donkey using RAPD-PCR analysis. Seven primers of arbitrary sequences were used in the PCR reactions. The screened primers gave total patterns ranging from 27 to 39 reproducible bands for each isolate. Each population isolate gave its specific pattern. Although distinct polymorphic patterns were obtained among the four isola

Molecular genetic profiles among individual Clonorchis sinensis adults collected from cats in two geographic regions of China revealed by RAPD and MGE-PCR methods.

Clonorchis sinensis causes the important food-borne zoonosis, clonorchiasis, which is endemic in East Asia, especially in China mainly in Guangdong, Guangxi and Heilongjiang provinces and Korea. Although comparisons on isoenzymes and some molecular profiles of C. sinensis collected from different parts of China and Korea have been studied, few works on the genetic variation among the individuals from different regions of China has been reported. In the present study, individual adults of C. sinensis were collected from cats in two geographic locations (Guangdong province in the South and Heilo

First molecularly supported detection of Oochoristica sp. (Cestoda: Anoplocephalidae) in a Yucatán spiny-tailed iguana (Cachryx defensor) from Italy: A case report.

During a parasitological investigation conducted in Italy on domestic reptiles, a motile proglottid was macroscopically observed in a fecal sample from a Yucatán spiny-tailed iguana (Cachryx defensor). Subsequent analysis using the quali-quantitative flotation technique (Mini-FLOTAC®) revealed the presence of cestode eggs. DNA was extracted from the sample and amplified using conventional PCR. Sequence comparison with those available in GenBank showed the highest similarity (98%) with Oochoristica hemidactyli, supporting identification at the genus level (Oochoristica sp.). To our knowledge, t

Molecular analysis supports the morphological identification of the amphistome Pseudodiscus collinsi (Paramphistomoidea: Platyhelminthes) in wild elephants of Sri Lanka.

Post-mortem examinations of elephants in Sri Lanka's Yala National Park morphologically identified amphistome flukes as Pseudodiscus collinsi, a species previously found in Indian elephants. These Sri Lankan worms had 100% ITS-2 sequence identity with that of the Indian P. collinsi Wayanad specimen (GenBank: PQ046280). This species is currently placed in the paramphistomoid family Gastrodiscidae. We compared three markers, ITS-2, partial 28S rDNA (D1-D3 domain), and partial mitochondrial cox1 sequences to previously published data and constructed three maximum likelihood (ML) phylogenies. Description of a new Neotropical species of Mathevotaenia (Cestoda: Anoplocephalidae), a parasite of the Virginia opossum Didelphis virginiana.

The cestodes of the genus Mathevotaenia Akhumyan, 1946, are parasites of mammals, although some species also occur in reptiles. The genus comprises 61 species distributed worldwide, 25 of which occur in the American continent. During a collection of mammals in Yucatán, Mexico, six specimens of a cestode belonging to Mathevotaenia were found in the Virginia opossum Didelphis virginiana Kerr. The aim of this study is to morphologically describe these specimens, which represent a new species, and to evaluate their phylogenetic position within the Anoplocephalidae, based on cox1, 28S rDNA and ITS

New records of Trichinella patagoniensis from Argentina.

Trichinella spiralis was long considered the sole Trichinella species in Argentina. However, since 2004, various Trichinella species, including the encapsulated Trichinella patagoniensis and Trichinella britovi, as well as the unencapsulated Trichinella pseudospiralis, have been detected in the country. The present study aimed to identify Trichinella ML at the species level from cougars naturally infected from Argentina. To this end, muscle tissue samples from one cougar each from Córdoba, Neuquén, and Santa Cruz Provinces were individually analysed using the artificial digestion technique. DN

GENETIC ANALYSIS OF ANCIENT CLONORCHIS SINENSIS EGGS ATTAINED FROM GORYEONG MUMMY OF JOSEON DYNASTY PERIOD.

Although Clonorchis sinensis is a parasite that still infects many people in East Asia, its genetics remain largely unknown. We conducted ancient DNA analysis of C. sinensis eggs obtained from a Joseon period mummy newly discovered in South Korea. Clonorchis sinensis DNA was amplified for internal transcribed spacer 1, cytochrome c oxidase subunit 1, and NADH dehydrogenase subunit 2 and 5 genes. The results of BLAST/NCBI showed that the consensus sequences were 98.24 to 100% identical to the modern and ancient C. sinensis sequences reported from Korea, China, Japan, and other Asian countries.

Discovery of filarial nematode DNA in Amblyomma americanum in Northern Virginia.

Ticks collected in 2011 were screened for the presence of filarial nematode genetic material, and positive samples were sequenced for analysis. Monanema-like filarial nematode DNA was recently discovered in Amblyomma americanum in northern Virginia, marking the first time genetic material from this parasite has been discovered in ticks in the state. Phylogenetic analysis revealed that this material was directly related to a previously discovered filarial nematode in A. americanum populations in Maryland as well as recently identified parasites in Ixodes scapularis from southern Connecticut. Schistosoma kisumuensis n. sp. (Digenea: Schistosomatidae) from murid rodents in the Lake Victoria Basin, Kenya and its phylogenetic position within the S. haematobium species group.

Schistosoma kisumuensis n. sp. is described based on 6 adult males and 2 adult females collected from the circulatory system of 3 murid rodent species, Pelomys isseli, Mastomys natalensis, and Dasymys incomtus. Specimens were collected from a single location, Nyabera Swamp, in Kisumu, Kenya in the Lake Victoria Basin. This new species is morphologically similar to members of the S. haematobium group, currently represented by 8 species parasitizing artiodactyls and primates, including humans. Schistosoma kisumuensis differs from these species by producing relatively small Schistosoma intercalat

Discovery of Paragonimus proliferus in Northern Vietnam and their molecular phylogenetic status among genus Paragonimus.

During an epidemiological survey for Paragonimus and paragonimosis in northern Vietnam, we found extremely large excysted metacercariae (2.50 +/- 0.14 mm in length and 0.72 +/- 0.08 mm in width; mean +/- standard deviation of 20 samples) in mountainous crabs, Potamiscus mieni. Adult worms were successfully obtained by intraperitoneal injection with those large excysted metacercariae in a cat. Morphological and morphometric data of those large excysted metacercariae and the adult worms derived from them are identical to those of Paragonimus proliferus found in Yunnan province, China. However, w

Taenia solium DNA is present in the cerebrospinal fluid of neurocysticercosis patients and can be used for diagnosis.

Neurocysticercosis is the most frequent parasitic infection of the CNS and the main cause of acquired epilepsy worldwide. Seizures are the most common symptoms of the disease, together with headache, involuntary movements, psychosis and a global mental deterioration. Absolute diagnostic criteria include the identification of cysticerci, with scolex, in the brain by MRI imaging. We demonstrate here, for the first time, that T. solium DNA is present in the cerebrospinal fluid of patients. The PCR amplification of the parasite DNA in the CSF enabled the correct identification of 29/30 cases (96.7

Molecular identification and larval morphological description of Contracaecum pelagicum (Nematoda: Anisakidae) from the anchovy Engraulis anchoita (Engraulidae) and fish-eating birds from the Argentine North Patagonian Sea.

Anisakids use invertebrates as paratenic and/or intermediate hosts as a basic feature of larval transmission. The third-stage larva usually develops in invertebrates which are prey items of finfish paratenic hosts. Contracaecum larvae molt twice inside the egg and hatch as free third-stage larvae ensheathed in the second-stage larval cuticle. Copepods act as paratenic or obligatory hosts, usually ingesting these free L3 larvae, and fish act as intermediate/paratenic or metaparatenic hosts preying on infected copepods. Fish-eating birds acquire L3 larvae by ingesting infected fish where they de

Mitochondrial and nuclear ribosomal DNA evidence supports the existence of a new Trichuris species in the endangered françois' leaf-monkey.

The whipworm of humans, Trichuris trichiura, is responsible for a neglected tropical disease (NTD) of major importance in tropical and subtropical countries of the world. Whipworms also infect animal hosts, including pigs, dogs and non-human primates, cause clinical disease (trichuriasis) similar to that of humans. Although Trichuris species are usually considered to be host specific, it is not clear whether non-human primates are infected with T. trichiura or other species. In the present study, we sequenced the complete mitochondrial (mt) genome as well as the first and second internal trans
