Cellular and Extracellular MicroRNA Dysregulation in LRRK2-Linked Parkinson's Disease.

Cell-free microRNAs in body fluids have emerged as promising biomarker candidates in neurodegenerative diseases. While several studies have identified dysregulated miRNAs in sporadic Parkinson's disease, it remains unclear whether distinguishable alterations of cell-free miRNAs occur in genetic forms of the disease, such as those associated with the LRRK2 G2019S mutation. In this proof-of-concept study, we used a human induced pluripotent stem cell-derived dopaminergic neuron model to investigate whether the LRRK2 G2019S mutation induces detectable changes in the intra- and extracellular miRNA

Novel application of diode engraving laser for preparing corneal allogenic intrastromal ring segments: a comparative study with femtosecond laser and manual trephine.

Corneal allogenic intrastromal ring segments (CAIRS) provide a biocompatible option for the treatment of keratoconus and ectatic disorders, however surgical outcomes rely heavily on pre-operative planning and precise donor tissue preparation. Femtosecond (FS) lasers ensure micron-level accuracy but are costly and often inaccessible, whereas manual trephines are more affordable but limited to fixed geometries and variable thickness. This study evaluated a low-cost automated handheld portable diode engraving laser as a novel alternative for CAIRS preparation. In an ex vivo feasibility study, hum

NeoThy™ immune-humanized mice can produce anti-drug antibodies to support immunogenicity assessment for biological drug products.

INTRODUCTION: Assessment of immunogenicity for new therapeutic protein products and biosimilars is an important aspect of drug development and the safety evaluation process. Conventional animal models typically do not reliably predict immunogenicity, as the human drug is seen as foreign in the animal model. To address this need, we conducted a proof-of-concept study with neonatal thymus (NeoThy™) immune-humanized mice using drug products known to have higher rates of immunogenicity in patients. NeoThy immune-humanized mice develop a fully engrafted human immune system that includes a human thy

Can scent-detection dogs detect the stress associated with trauma cue exposure in people with trauma histories? A proof-of-concept study.

INTRODUCTION: Post-traumatic stress disorder (PTSD) is an impairing mental health condition with high prevalence among military and general populations alike. PTSD service dogs are a complementary and alternative intervention needing scientific validation. We investigated whether dogs can detect putative stress-related volatile organic compounds (VOCs) in the breath of people with trauma histories (54% with PTSD) exposed to personalized trauma cues. METHODS: Breath samples were collected from 26 humans over 40 experimental sessions during a calm (control breath sample) and stressed state indu

Outpatient allografting in hematologic malignancies and nonmalignant disorders--applying lessons learned in the canine model to humans.

The novel allogeneic transplant approach outlined in this chapter proposes the dual concepts that currently used intensive cytoreductive conditioning programs can be successfully replaced by nonmyeloblative immunosuppression and that stem cell grafts create marrow space for engraftment through subclinical GVH reactions. Immunosuppression can be conceptually divided into two parts, one targeted exclusively towards host cells before transplantation, and the other aimed at both host and donor T lymphocytes after transplantation. The resultant effect is to either establish mutual graft-host tolera

Exercise-mobilized donor lymphocyte infusions enhanced with cytokine stimulation for the prevention and treatment of leukemic relapse after allogeneic hematopoietic cell transplantation.

Donor lymphocyte infusions (DLI) are a standard therapy following allogeneic hematopoietic cell transplantation (alloHCT) for preventing and treating leukemic relapse in high-risk patients, particularly those with myeloid malignancies such as acute myeloid leukemia (AML), chronic myeloid leukemia (CML), and myelodysplastic syndrome (MDS). However, the efficacy of DLI remains suboptimal and is accompanied by a significant risk of life-threatening graft-versus-host disease (GvHD), highlighting the urgent need for strategies that enhance graft-versus-leukemia (GvL) effects while mitigating GvHD.

Sociocultural perspective on organ and tissue donation among reservation-dwelling American Indian adults.

OBJECTIVE: To discover the sociocultural patterns that influence decisions about organ and tissue donation among American Indian (AI) adults. DESIGN: This qualitative ethnographic study used a social-ecological framework. A snowball sampling technique was used to recruit 21 Oglala Lakota Sioux participants (age >or= 19 years) living on the Pine Ridge Indian Reservation in South Dakota, USA. Face-to-face interviews were conducted using open-ended questions derived from the social-ecological perspective of Stokols (1992). Interviews were audiotaped and transcribed. Data were categorized into co

A Novel 3D-Printed Titanium Prosthesis with Osseointegrated Dental Implants for Reconstruction of Segmental Mandibular Defects.

INTRODUCTION: Segmental mandibular defects present complex reconstructive challenges and are associated with significant functional and aesthetic morbidity. While vascularized bone flaps remain the standard for reconstruction, they are associated with donor-site morbidity, prolonged operative times, and limitations in predictable dental rehabilitation. This proof-of-concept study evaluated the mechanical feasibility, in a synthetic bench-top model, of a patient-specific, 3D-printed titanium mandibular prosthesis incorporating conventional dental implants for both dental rehabilitation and biol

[Risk of perinatal complication and egg donation: Role of resorting to cross-border care?].

OBJECTIVES: In France, egg donation is covered by Social insurance among women<43 years old. Because of shortage of egg donor, women aged 43 years or more cannot resort to egg donation in French infertility centers, leading them to turn to the foreign centers having practices different and less regulated than in France. We are thus brought to take care of the pregnancy and of the delivery of these women. Our objective was to estimate if the perinatal risks are more important after egg donation abroad than in case of egg donation in a French center. MATERIAL AND METHODS: Retrospective study be

Older Swedish Adults with High Self-Perceived Health Show Optimal 25-Hydroxyvitamin D Levels Whereas Vitamin D Status Is Low in Patients with High Disease Burden.

Controversy pervades the definition of adequate and optimal vitamin D status. The Institutes of Medicine have recommended serum 25(OH)D levels above 50 nmol/L based upon evidence related to bone health, but some experts, including the Endocrine Society and International Osteoporosis Foundation, suggest a minimum serum 25(OH)D level of 75 nmol/L to reduce the risk of falls and fractures in older adults. In a cross-sectional study, we compared vitamin D status in people ≥75 years selected from four groups with a frailty phenotype, combined with a control group free from serious illness, and who

Gene expression adaptation of metastases to their host tissue.

Adaptation of metastatic cells to their host tissue determines the pathogenicity of cancer. Yet, it remains elusive to what extent the host environment drives gene expression programs in metastatic cells. We present a new concept to identify adaptive mechanisms that enable metastases to establish themselves in a novel tissue context. We generated quadruple-paired single-cell RNA-sequencing data from malignant and benign tissues from untreated donors with colorectal adenocarcinoma and liver metastasis. Utilizing a computational approach, we deduce tissue-adaptive expression patterns by identify

Top-Down Hierarchical Allograft Engineering.

BACKGROUND: More than 40 years of intensive research in tissue engineering have not produced clinically transplantable bioartificial organs. This limitation is intrinsic to the biological complexity of creating transplantable organs from the bottom-up using scaffolds seeded with stem cells. In contrast, allograft transplantation provides fully functional and architecturally organized organs at a level that no current bioengineering platform is even close to reproducing. OBSERVATIONS: This review defines the concept of allograft engineering: a top-down, allograft-centered paradigm that begins

Designing an Interoperable Intelligent Agent for the Blood Transfusion Chain in Geneva, Switzerland.

INTRODUCTION: Blood supply chain management is a cornerstone of modern medicine, yet it faces persistent structural challenges, including unpredictable supply and demand, perishability, and fragmentation of information systems. These limitations can hinder real-time inventory visibility and complicate extended blood compatibility searches, particularly for poly-transfused patients at risk of alloimmunization. METHODS: We designed a proof-of-concept combining (1) an interoperable health data exchange pipeline based on HL7 FHIR, SNOMED-CT, and LOINC standards, and (2) an intelligent matching ag

Transplantation of GABAergic Interneuron Progenitors Restores Cortical Circuit Function in an Alzheimer's Disease Mouse Model.

In addition to dementia, Alzheimer's patients suffer from sleep impairments and aberrations in sleep-dependent brain rhythms. Deficits in inhibitory GABAergic interneuron function disrupt one of those rhythms, slow oscillation in particular, and actively contribute to Alzheimer's progression. The degree to which transplantation of healthy donor interneuron progenitors restores slow oscillation rhythm in young APP/PS1 mice is tested. Medial ganglionic eminence (MGE) progenitors are harvested from mouse embryos and transplanted them into host APP/PS1 mutant cortices. 3D light-sheet and structure

Advancing lung transplantation through machine learning and artificial intelligence.

PURPOSE OF REVIEW: To explore the current applications of artificial intelligence and machine learning in lung transplantation, including outcome prediction, drug dosing, and the potential future uses and risks as the technology continues to evolve. RECENT FINDINGS: While the use of artificial intelligence (AI) and machine learning (ML) in lung transplantation is relatively new, several groups have developed models to predict short-term outcomes, such as primary graft dysfunction and time-to-extubation, as well as long-term outcomes related to survival and chronic lung allograft dysfunction.

Fascinating Transformation of SAM-Competitive Protein Methyltransferase Inhibitors from Nucleoside Analogues to Non-Nucleoside Analogues.

The abnormal expression of protein methyltransferase (PMT) has been linked with many diseases such as diabetes, neurological disorders, and cancer. S-Adenyl-l-methionine (SAM) is a universal methyl donor and gets converted to S-adenyl-l-homocysteine (SAH), an endogenous competitive inhibitor of SAM. Initially developed SAM/SAH mimetic nucleoside analogues were pan methyltransferase inhibitors. The gradual understanding achieved through ligand-receptor interaction paved the way for various rational approaches of drug design leading to potent and selective nucleoside inhibitors. The present pers

Sulfotransferase 4A1 activity facilitates sulfate-dependent cellular protection to oxidative stress.

Sulfotransferase 4A1 (SULT4A1) is an orphan member of the cytosolic SULT superfamily that contains enzymes that catalyze the sulfonation of hydrophobic drugs and hormones. SULT4A1 has been assessed through all classical SULT approaches yet no SULT activity has been reported. To ascertain SULT4A1 function and activity, we utilized Saccharomyces cerevisiae as a model system, which exhibits no endogenous SULT activity nor possesses SULT-related genes. We observed that ectopic SULT4A1 expression in yeast displays similar subcellular localization as reported in mouse neurons and observed that SULT4

Cytomegalovirus Therapy: Role of Letermovir in Prophylaxis and Treatment in Transplant Recipients.

Cytomegalovirus (CMV) is a common viral pathogen in the transplant population and is associated with significant morbidity and mortality. CMV prevention is paramount; however, selecting the best preventive strategy depends on many factors including donor-recipient CMV serostatus, transplant-specific risks, antiviral toxicities and cost. Novel CMV therapeutics such as letermovir (LTV) are desperately needed to optimize CMV management. Uniquely among CMV antiviral therapies, LTV inhibits the viral terminase complex in the CMV DNA synthesis pathway and disrupts viral genome packaging. Further, it

Depression patient-derived cortical neurons reveal potential biomarkers for antidepressant response.

Major depressive disorder is highly prevalent worldwide and has been affecting an increasing number of people each year. Current first line antidepressants show merely 37% remission, and physicians are forced to use a trial-and-error approach when choosing a single antidepressant out of dozens of available medications. We sought to identify a method of testing that would provide patient-specific information on whether a patient will respond to a medication using in vitro modeling. Patient-derived lymphoblastoid cell lines from the Sequenced Treatment Alternatives to Relieve Depression study we
