Modulation of cardiopulmonary depressor reflex in nucleus ambiguus by electroacupuncture: roles of opioids and γ-aminobutyric acid.

Stimulation of cardiopulmonary receptors with phenylbiguanide (PBG) elicits depressor cardiovascular reflex responses, including decreases in blood pressure and heart rate mediated in part by the brain stem parasympathetic cardiac neurons in the nucleus ambiguus (NAmb). The present study examined NAmb neurotransmitter mechanisms underlying the influence of electroacupuncture (EA) on the PBG-induced hypotension and bradycardia. We hypothesized that somatic stimulation during EA modulates PBG responses through opioid and γ-aminobutyric acid (GABA) modulation in the NAmb. Anesthetized and ventila…

A review of the potential effect of electroacupuncture and moxibustion on cell repair and survival: the role of heat shock proteins.

In recent years, a considerable amount of research has been focused on the underlying mechanisms of electroacupuncture and moxibustion assisted tissue repair. Intracellular protein denaturation is a significant pathological step of acute conditions such as stroke, myocardial infarction and acute pancreatitis. Protein aggregation can be observed after the protein denaturation step in chronic diseases of the central nervous system like Alzheimer's and Parkinson's disease, and also in other chronic system diseases including cataract formation. Heat shock proteins (HSPs) are fundamental for intrac…

Disinhibitory involvement of the anterior cingulate cortex in the descending antinociceptive effect induced by electroacupuncture stimulation in rats.

The present study was conducted to clarify the role of the anterior cingulate cortex (ACCX) in acupuncture analgesia. Experiments were performed on 35 female Wistar albino rats weighing about 300 g. Single unit recordings were made from ACCX neurons with a tungsten microelectrode. Descending ACCX neurons were identified by antidromic activation from electrical shocks applied to the ventral part of the ipsilateral PAG through a concentric needle electrode. Cathodal electroacupuncture stimulation of Ho-Ku (0.1 ms in duration, 45 Hz) for 15 min was done by inserting stainless steel needles bilate…

Electroacupuncture-induced analgesia in a rat model of ankle sprain pain is mediated by spinal alpha-adrenoceptors.

In a previous study, we showed that electroacupuncture (EA) applied to the SI-6 point on the contralateral forelimb produces long-lasting and powerful analgesia in pain caused by ankle sprain in a rat model. To investigate the underlying mechanism of EA analgesia, the present study tested the effects of various antagonists on known endogenous analgesic systems in this model. Ankle sprain was induced in anesthetized rats by overextending their right ankle with repeated forceful plantar flexion and inversion of the foot. When rats developed pain behaviors (a reduction in weight-bearing of the af…

Effect of electroacupuncture tolerance by different frequencies on the opioid inhibition to cardiovascular activity in the spinal cord of rats.

Continuous 2-15 Hz or 2 Hz electroacupuncture (EA) of 6 h applied to rats resulted in electroacupuncture tolerance (ET). The effect of 2-15 Hz ET on the recovery of mean arterial blood pressure (MAP) and heart rate (HR) of rats after hemorrhagic shock, and the effect of intrathecal (i.t.) injection of [D-ala2, D-leu5]enkephalin (DADLE) on MAP and HR of 2 Hz ET rats were observed. The results were as follows: 1) there was no dramatic difference between the 2-15 Hz ET group and the control group in the recovery of MAP and HR from hemorrhagic shock; 2) DADLE (25 micrograms) caused almost the same…

Astrocytic NOX2 contributes to ROS-associated mitochondrial dysfunction in cerebral ischemia-reperfusion injury: modulation by electroacupuncture.

BACKGROUND: Cerebral ischemia-reperfusion injury (CIRI) causes severe neuronal damage, in which oxidative stress and mitochondrial dysfunction are major pathological events. Astrocytes are important regulators of redox homeostasis in the injured brain, yet whether astrocytic NADPH oxidase 2 (NOX2) contributes to mitochondrial impairment after ischemia remains poorly understood. Electroacupuncture (EA) has been reported to protect against ischemic brain injury, but the cellular mechanisms involved remain unclear. METHODS: In this study, a mouse middle cerebral artery occlusion/reperfusion (MCA…

Electroacupuncture Attenuates Cerebral Ischemia-Reperfusion Injury via Vagus Nerve-α7 Nicotinic Acetylcholine Receptor-Dependent Microglial Polarization.

BACKGROUND: Neuroinflammation is a central pathological process in cerebral ischemia/reperfusion injury. Electroacupuncture has demonstrated neuroprotective potential in ischemic stroke; however, whether electroacupuncture engages a defined central cholinergic anti-inflammatory reflex to regulate microglial phenotype remains unknown. METHODS: A rat model of middle cerebral artery occlusion/reperfusion was employed. Low-intensity electroacupuncture was applied to the acupoints Large Iintestine 10 and Stomach 36 immediately after reperfusion. Neurological deficits and structural damage were ass…

Electroacupuncture Alleviates Cerebral Ischemia-Reperfusion Injury by Downregulating IL-17 A and Inhibiting Neurotoxic Astrocyte Activation.

Cerebral ischemia-reperfusion injury (CIRI) represents a critical pathological mechanism underlying ischemic stroke, yet effective therapeutic interventions remain limited. Neurotoxic astrocytes, activated by inflammatory mediators such as interleukin-17 A (IL-17 A), exacerbate neuronal damage. Although electroacupuncture (EA) has demonstrated neuroprotective properties, its influence on IL-17 A signaling and subsequent astrocyte-mediated neurotoxicity in CIRI remains unclear. This study aims to investigate whether EA mitigates CIRI by downregulating IL-17 A to suppress the activation of neuro…

Electroacupuncture Serum Protects against Blood-brain Barrier Damage after Ischemic Stroke by Regulating Pericytes in vitro.

BACKGROUND: Electroacupuncture (EA) exerts a protective role in Blood-brain Barrier (BBB) damage after ischemic stroke, but whether this effect involves the regulation of the pericytes in vitro is unclear. METHODS: The in vitro BBB models were established with brain microvascular endothelial cells (BMECs) and pericytes, and the co-cultured cells were randomly divided into three groups: the control group, oxygen-glucose deprivation/reoxygenation (OGD/R) group and EA group. OGD/R was performed to simulate cerebral ischemia-reperfusion in vitro. EA serum was prepared by EA treatment at the "Renz…
