Heterologous expression of rat P450 2E1 in a mammalian cell line: in situ metabolism and cytotoxicity of N-nitrosodimethylamine.

GM0637, a human fibroblast cell line, was transfected with pCMV2E1, an expression vector containing the full length cDNA for rat cytochrome P450 2E1 (P450 2E1), and with pCMVneo, which contained vector alone, and the selected clones were designated GM2E1 and GMneo, respectively. Western blot analysis showed that GM2E1, but not GMneo, expressed a protein that reacted with anti-human P450 2E1 antibody. The 7-ethoxycoumarin O-deethylase,p-nitrophenol hydroxylase, and N-nitrosodimethylamine (NDMA) demethylase activities of the P450 in these cells were measured in monolayer cell cultures without pr

Comparative studies on prostanoid receptors in human non-pigmented ciliary epithelial and mouse fibroblast cell lines.

To examine the expression of functional prostanoid receptors in the human non-pigmented ciliary epithelial (ODMC1-2) and mouse fibroblast cell lines (NIH 3T3) we have measured the generation of the second messengers, cyclic AMP, inositol phosphates and the mobilization of intracellular calcium ([Ca2+]i) following stimulation by prostaglandin receptor agonists. The amount of cyclic AMP generated was measured by a protein binding method. Radiolabeled inositol phosphates were separated using ion exchange columns and quantified by counting the radioactivity. For intracellular calcium measurements,

Interleukin-1alpha and tumour necrosis factor-alpha modulate airway smooth muscle DNA synthesis by induction of cyclo-oxygenase-2: inhibition by dexamethasone and fluticasone propionate.

1. Previous studies have established that glucocorticoids inhibit airway smooth muscle DNA synthesis. The effects of a combination of the pro-inflammatory cytokines, interleukin-1alpha (IL-1alpha) and tumour necrosis factor-alpha (TNF-alpha) on the inhibition of DNA synthesis by glucocorticoids in human cultured airway smooth muscle have now been investigated, since these cytokines are chronically expressed in asthmatic airways. 2. Thrombin (0.3 u ml(-1)) and basic fibroblast growth factor (bFGF, 300 pM) stimulated increases in DNA synthesis which were concentration-dependently inhibited by de

Hypoxic Regulation of Notch1 Promotes Mitochondrial Fission and Scleral Remodeling in Myopia.

BACKGROUND: The progression of myopia is typically accompanied by hypoxia-induced remodeling of the scleral extracellular matrix (ECM). In this study, we focused on the regulatory mechanisms underlying Notch signaling pathway activation of mitochondrial dynamics under conditions of scleral hypoxia, along with its effects on the scleral ECM. METHODS: Three-week-old male guinea pigs were used to establish form-deprived myopia (FDM) models, and human scleral fibroblasts (HSFs) were cultured in a hypoxic environment. To examine the intrinsic associations among factors, we used short hairpin RNAs

Genetic causes of isolated and severe fetal growth restriction in normal chromosomal microarray analysis.

OBJECTIVE: To investigate the genetic burden in fetuses with isolated and severe fetal growth restriction (FGR) using Trio whole-exome sequencing (WES) with a normal chromosomal microarray. METHOD: This retrospective study analyzed WES results of singleton fetuses with isolated and severe FGR, whose estimated fetal weight (EFW) was less than the third percentile by Hadlock formula, in a tertiary center between March 2016 and March 2022. Cases with abnormal chromosomal microarray analysis (CMA) and TORCH results were excluded. RESULTS: Fifty-one fetuses with isolated and severe FGR and negati

Tumor necrosis factor alpha and interleukin 1 beta suppress myofibroblast activation via nuclear factor kappa B signaling in 3D-cultured mitral valve interstitial cells.

Mitral valve disease is a major cause of cardiovascular morbidity throughout the world. Many different mitral valve pathologies feature fibrotic remodeling, often accompanied by an inflammatory state. Mitral valve fibrosis is mediated by valvular interstitial cells (VICs), which reside in the valve leaflets and often differentiate into myofibroblast-like cells during disease conditions. In this study, we investigated the effects of tumor necrosis factor alpha (TNF-α) and interleukin 1 beta (IL-1β) on mitral VICs, since these pro-inflammatory cytokines have been shown to exert pleiotropic effec

TSG-6 inhibits hypertrophic scar fibroblast proliferation by regulating IRE1α/TRAF2/NF-κB signalling.

TNF-stimulated gene (TSG-6) was reported to suppress hypertrophic scar (HS) formation in a rabbit ear model, and the overexpression of TSG-6 in human HS fibroblasts (HSFs) was found to induce their apoptotic death. The molecular basis for these findings, however, remains to be clarified. HSFs were subjected to TSG-6 treatment. Treatment with TSG-6 significantly suppressed HSF proliferation and induced them to undergo apoptosis. Moreover, TSG-6 exposure led to reductions in collagen I, collagen III, and α-SMA mRNA and protein levels, with a corresponding drop in proliferating cell nuclear antig

Properties of Oligomeric Interaction of the Cytomegalovirus Core Nuclear Egress Complex (NEC) and Its Sensitivity to an NEC Inhibitory Small Molecule.

Herpesviral nuclear egress is a regulated process shared by all family members, ensuring the efficient cytoplasmic release of viral capsids. In the case of human cytomegalovirus (HCMV), the core of the nuclear egress complex (NEC) consists of the pUL50-pUL53 heterodimer that builds hexameric lattices for capsid binding and multicomponent interaction, including NEC-associated host factors. A characteristic feature of NEC interaction is the N-terminal hook structure of pUL53 that binds to an alpha-helical groove of pUL50, thus termed as hook-into-groove interaction. This central regulatory eleme

E-cadherin inactivation shapes tumor microenvironment specificities in invasive lobular breast cancer.

Invasive lobular breast carcinoma (ILC) shows specific stromal features, and a high tumor-infiltrating lymphocyte (TIL) content being associated with poor patient prognosis. Here, we reveal the underlying mechanism by performing single-cell analysis, immunohistochemistry, deconvolution of bulk RNA-sequencing in a large female ILC series and functional assays. We show that E-cadherin (CDH1)-loss in breast cancer cells prevents differentiation of FAP+ inflammatory cancer-associated fibroblasts (iCAF) into FAP+ myofibroblastic CAF, leading to iCAF accumulation in ILC. In turn, FAP+ iCAF attract T

Enhanced MiR-711 transcription by PPARγ induces endoplasmic reticulum stress-mediated apoptosis targeting calnexin in rat cardiomyocytes after myocardial infarction.

MicroRNA 711 (miR-711) levels in the heart change dynamically after myocardial infarction (MI). As peroxisome proliferator-activated receptor gamma (PPARγ) can upregulate miR-711 in adipocytes and cardiac fibroblasts, this study examined the precise mechanism of PPARγ-mediated miR-711 upregulation and its role in the heart in the early stages after MI. In a rat model of MI induced by left anterior descending coronary artery ligation, immunohistochemical and western blot analyses revealed increased PPARγ expression in cardiomyocyte nuclei after MI. PPARγ modulated miR-711 levels in neonatal rat

Uncovering the gastroprotective properties of araucaria brown propolis and its active compound, abietic acid.

Although Araucaria brown propolis is a singular propolis with potential therapeutic applications, its anti-ulcer activity remains unexplored. This study investigated the gastroprotective effects of Araucaria brown propolis hydroalcoholic extract (HeBPA, 30- 300 mg/kg and its isolated compound, abietic acid (AA) in corresponding doses to its content in HEBPA. The extract's anti-ulcer activity was evaluated using acidified ethanol-induced gastric ulcers and pylorus ligation to assess gastric antisecretory activity. Additionally, the study examined histological, oxidative, and inflammatory parame

A structurally defined galactoglucan from Arisaema erubescens with a multi-target tumor-associated protein binding domain.

As targeted therapy assumes an increasingly pivotal role in comprehensive cancer treatment, there is a growing imperative to develop natural drugs that exhibit low toxicity and minimal side effects. Polysaccharides derived from the traditional Chinese herbal medicine Arisaema erubescens have been reported to show antitumor potential, however the key structural features and specific molecular targets responsible for their pharmacological effects are still vague. To address this question, a homogeneous polysaccharide TNX05 (Mw ≈ 9 kDa) was obtained and characterized from Arisaema erubescens. Str

Heart-derived factors and organ cross-talk in settings of health and disease: new knowledge and clinical opportunities for multimorbidity.

Cardiovascular disease affects millions of people worldwide and often presents with other conditions including metabolic, renal and neurological disorders. A variety of secreted factors from multiple organs/tissues (proteins, nucleic acids and lipids) have been implicated in facilitating organ cross-talk that may contribute to the development of multimorbidity. Secreted proteins have received the most attention, with the greatest body of research related to factors released from adipose tissue (adipokines), followed by skeletal muscle (myokines). To date, there have been fewer studies on prote

Chemotherapy-induced CAF-associated stromal remodeling in a humanized stroma pancreatic ductal adenocarcinoma organoid xenograft model: a comparison between GP and GS.

Pancreatic ductal adenocarcinoma features a dense, desmoplastic tumor microenvironment (TME) dominated by cancer-associated fibroblasts (CAFs). While gemcitabine plus nab-paclitaxel (approximated as gemcitabine plus paclitaxel [GP]) and gemcitabine plus S-1 (GS) are standard regimens with distinct clinical indications, their specific effects on tumor suppression and CAF-associated stromal remodeling remain unclear. Using a humanized, three-dimensional (3D) S2-013 organoid xenograft model integrating human mesenchymal stem cells (MSCs) and human endothelial cells, we investigated the histopatho

Effects of combined training on quality of life in individuals with overweight-related type 2 diabetes: a pilot study of correlations with metabolic, inflammatory and thermogenic markers.

BACKGROUND: There is still a lack of information regarding the impact of training on the domains that constitute quality of life (QoL), and the relationship between QoL with hormonal, inflammatory and pro-thermogenic/anti-inflammatory components. Thus, the present work aimed to evaluate the effects of combined training (CT) on the QoL of individuals with overweight-related type 2 diabetes mellitus (T2DM) and to correlate (baseline values and CT-induced delta percentage changes (Δ%)) QoL variables with hormonal, inflammatory, and pro-thermogenic/anti-inflammatory components. METHODS: Middle-ag

FR-167653, a selective p38 MAPK inhibitor, exerts salutary effect on liver cirrhosis through downregulation of Runx2.

Liver cirrhosis remains a difficult-to-treat disease with a substantial morbidity and mortality rate. There is an emerging body of data purporting a pivotal role of the activated p38 mitogen-activated protein kinase (MAPK) in the process of cirrhosis. Several anticirrhotic agents have been developed over the past few years, and most of them exert their effects by indirectly inhibiting the p38 pathway. Effect of a selective p38 inhibitor is yet to be reported. In this study, we evaluated the salutary effect of FR-167653 (FR), a selective p38 inhibitor, in a carbon tetrachloride (CCl(4))-induced

Cytokine mRNA repertoire of articular chondrocytes from arthritic patients, infants, and neonatal mice.

Articular chondrocytes from nine arthritic patients, five infants, and Balb/c neonatal mice were analyzed for the presence of various cytokine mRNAs by a reverse transcriptase polymerase chain reaction (RT-PCR). Four cytokine mRNAs, interleukin (IL)-6, IL-8, IL-11, and macrophage colony stimulating factor (M-CSF), were detected in all human chondrocytes, regardless of source. IL-10, IL-12p35, and tumor necrosis factor alpha (TNF-alpha) transcripts were found in at least 12 of the 14 human samples. Fetal macrosomia and placental expression of fibroblast growth factor 21 and peroxisome proliferator-activated receptor alpha.

Macrosomia (birth weight >4000 g) is a product of endocrine dysfunction in utero leading to fetal overgrowth and can lead to maternal and infant morbidity. Fibroblast growth factor 21 (FGF21) and its transcription factor, peroxisome proliferator-activated receptor alpha (PPARα), are found in the placenta and are associated with abnormal metabolic states. Their relationship to the placental dysregulation that leads to macrosomia is unknown. We sought to evaluate the relationship between protein expression of FGF21 and PPARα in placental samples from infants with macrosomia compared to controls

Chemical chaperone treatment reduces intracellular accumulation of mutant collagen IV and ameliorates the cellular phenotype of a COL4A2 mutation that causes haemorrhagic stroke.

Haemorrhagic stroke accounts for ∼20% of stroke cases and porencephaly is a clinical consequence of perinatal cerebral haemorrhaging. Here, we report the identification of a novel dominant G702D mutation in the collagen domain of COL4A2 (collagen IV alpha chain 2) in a family displaying porencephaly with reduced penetrance. COL4A2 is the obligatory protein partner of COL4A1 but in contrast to most COL4A1 mutations, the COL4A2 mutation does not lead to eye or kidney disease. Analysis of dermal biopsies from a patient and his unaffected father, who also carries the mutation, revealed that both d

An intact and functional soluble form of the insulin receptor is secreted by cultured cells.

We have recently reported that the insulin receptor (IR), a tetrameric transmembrane protein located on the surface of target cells, is present as a soluble form in human plasma. In the present study we investigated whether human cells in culture release an intact and functional form of the IR. We found that IRs are secreted into the incubation medium by four cell lines (IM-9 human lymphoblasts, MCF-7 human breast cancer cells, HepG2 human hepatoma cells, and 3T3 mouse fibroblasts transfected with human IRs). IR secretion was further characterized in IM-9 cells. IR release was time, temperatur
