Unmasking phenotypic heterogeneity and extracellular matrix architecture in neuroblastoma through orthotopic xenograft modeling.

Neuroblastoma (NB) is a highly heterogeneous pediatric tumor in which the extracellular matrix (ECM) critically influences tumor growth, cell lineage plasticity, and therapeutic resistance. However, the interplay of ECM remodeling with the traceable origin and fate of tumor cell phenotypes remains poorly understood. Paired-like homeobox 2b (PHOX2B), a pivotal diagnostic marker reflecting neuronal differentiation states of NB, was examined across multiple NB models. Our analysis revealed heterogeneous PHOX2B expression, including PHOX2B-negative foci, indicating pronounced intratumoral heteroge

Effects of 3D-printed exosome-functionalized brain acellular matrix hydrogel on neuroinflammation in rats following cerebral hemorrhage.

BACKGROUND: Exosome-based therapeutics have garnered significant attention for intracerebral hemorrhage (ICH) treatment due to their capacity to regulate metabolic dysregulation, restore cellular homeostasis, and modulate the injury microenvironment via bioactive cargoes such as microRNAs and proteins. However, rapid systemic clearance and enzymatic degradation critically limit their therapeutic efficacy. To address this challenge, we engineered a three-dimensional (3D) bioprinted scaffold capable of encapsulating and sustaining the release of human umbilical cord mesenchymal stem cell-derived

3D bioengineered neural tissue generated from patient-derived iPSCs mimics time-dependent phenotypes and transcriptional features of Alzheimer's disease.

Several iPSC-derived three-dimensional (3D) cultures have been generated to model Alzheimer's disease (AD). While some AD-related phenotypes have been identified across these cultures, none of them could recapitulate multiple AD-related hallmarks in one model. To date, the transcriptomic features of these 3D models have not been compared with those of human AD brains. However, these data are crucial to understanding the pertinency of these models for studying AD-related pathomechanisms over time. We developed a 3D bioengineered model of iPSC-derived neural tissue that combines a porous scaffol

An injectable photo-cross-linking silk hydrogel system augments diabetic wound healing in orthopaedic surgery through spatiotemporal immunomodulation.

BACKGROUND: The complicated hyperglycaemic and chronic inflammation of diabetic wounds in orthopaedic surgery leads to dysregulated immune cell function and potential infection risk. Immune interventions in diabetic wounds face a possible contradiction between simultaneous establishment of the pro-inflammatory microenvironment in response to potential bacterial invasion and the anti-inflammatory microenvironment required for tissue repair. To study this contradiction and accelerate diabetic-wound healing, we developed a photocurable methacryloxylated silk fibroin hydrogel (Sil-MA) system, co-e

Antimicrobial laser-activated sealants for combating surgical site infections.

Surgical-site infections (SSIs) occur in 2-5% of patients undergoing surgery in the US alone, impacting 300 000-500 000 lives each year, and presenting up to 11 times greater risk of death compared to patients without SSIs. The most common cause of SSI is Staphylococcus aureus, and methicillin-resistant S. aureus (MRSA) is the most common pathogen in community hospitals. Current clinical devices used for approximating incisions and traumatic lacerations include sutures, adhesives, tapes, or staples with or without antimicrobial incorporation. However, current closure technologies may not provi

Anisotropic conductive reduced graphene oxide/silk matrices promote post-infarction myocardial function by restoring electrical integrity.

Myocardial infarction (MI) remains the leading cause of death globally, often leading to impaired cardiac function and pathological myocardial microenvironment. Electrical conduction abnormalities of the infarcted myocardium not only induce adverse myocardial remodeling but also prevent tissue repair. Restoring the myocardial electrical integrity, particularly the anisotropic electrical signal propagation within the injured area after infarction is crucial for an effective function recovery. Herein, optimized reduced graphene oxide (rGO) functionalized electrospun silk fibroin (rGO/silk) bioma

Osteoimmunity-Regulating biospun 3D silk scaffold for bone regeneration in critical-size defects.

INTRODUCTION: Silk-based biomaterials have received a great deal of attention in tissue engineering research for bone repair. Current silk-based materials are typically derived from silk protein solutions, but the limited solubility and solution stability of silk protein solution, coupled with problems such as high preparation cost and low productivity, which severely restrict the application of silk-based materials. OBJECTIVE: To address the challenges associated with the complex extraction process and inferior mechanical properties of silk protein or silk fiber-based materials in bone scaff

ROS-responsive 3D biological scaffold delivers hypoxia-primed extracellular vesicles for targeted modulation of neuroinflammation in intracerebral hemorrhage.

BACKGROUND: Emerging evidence suggests that paracrine mechanisms may underlie the therapeutic effects of human umbilical cord mesenchymal stem cell-derived extracellular vesicles (EVs) (hUCMSC-exos) in mitigating neuroinflammation following intracerebral hemorrhage (ICH). Hypoxic preconditioning enhances the paracrine efficacy of hUCMSC-exos. Building on prior studies [1, 2], we developed a ROS-responsive three-dimensional (3D) biological scaffold encapsulating hypoxia-primed EVs (Hypo-Exos) for sustained release under reactive oxygen species (ROS)-rich conditions. METHODS: The 3D biological

3D biological scaffold delivers Bergenin to reduce neuroinflammation in rats with cerebral hemorrhage.

BACKGROUND: Intracerebral hemorrhage (ICH) is a severe form of stroke characterized by high incidence and mortality rates. Currently, there is a significant lack of effective treatments aimed at improving clinical outcomes. Our research team has developed a three-dimensional (3D) biological scaffold that incorporates Bergenin, allowing for the sustained release of the compound. METHODS: This 3D biological scaffold was fabricated using a combination of photoinitiator, GEMA, silk fibroin, and decellularized brain matrix (dECM) to encapsulate Bergenin through advanced 3D bioprinting techniques.

A bionic multichannel nanofiber conduit carrying Tubastatin A for repairing injured spinal cord.

Spinal cord injury is a kind of nerve injury disease with high disability rate. The bioscaffold, which presents a biomimetic structure, can be used as "bridge" to fill the cavity formed by the liquefaction and necrosis of spinal nerve cells, and connects the two ends of the fracture to promote the effective recovery of nerve function. Tubasatin A (TUBA) is a potent selective histone deacetylase 6 (HDAC6) inhibitor, which can inhibit the overexpression of HDAC6 after spinal cord injury. However, TUBA is limited by high efflux ratios, low brain penetration and uptake in the treatment of spinal c

Therapeutic potential of natural polymer-based microgels for cartilage regeneration with implications for osteoarthritis.

Articular cartilage has a limited intrinsic capacity for self-repair because of its avascular, aneural, and alymphatic nature, making cartilage repair a major clinical challenge. This limitation is particularly important in osteoarthritis (OA), where cartilage loss occurs within a complex whole-joint pathological environment involving inflammation, extracellular matrix (ECM) degradation, oxidative stress, chondrocyte senescence, and altered mechanical loading. Cartilage tissue engineering (CTE) has emerged as a promising strategy to restore cartilage structure and function by integrating cells

Silk-coated PLGA non-spherical microparticles for simvastatin delivery in rat maxillofacial bone regeneration.

Maxillofacial bone defects present a major challenge in clinical practice. Bone grafting and bone tissue engineering are limited by various side effects and potential risks, while local drug delivery is regarded as a convenient and efficient strategy for bone repair and regeneration. However, for bone tissue regeneration, a drug delivery system should not only release osteogenic drugs but also possess an appropriate morphology to provide structural support for cell proliferation and differentiation. Polymeric non-spherical counterparts may offer new osteogenic properties through altered cellul

Multifunctional Defense of Achelura yunnanensis Cocoon: High-Strength Tough Silk Fiber, Biomineral Reinforcement, and Protease Degradation Resistance.

Natural silks offer combinations of mechanical performance and biological functionality that synthetic materials still struggle to replicate. Here, we report the structural, mechanical, and biochemical characterization of cocoon silk from Achelura yunnanensis (Lepidoptera: Zygaenidae), a moth that builds a leaf-wrapped cocoon with three integrated defense layers. The silk fiber exhibits an average tensile strength of 1038 ± 434 MPa, a toughness of 81 ± 49 MJ m-3, and an elastic modulus of 22.1 ± 8.8 GPa, approximately twice the respective values for Bombyx mori silk. These properties stem from

Biomimetic trilayered silk-based electrospun scaffolds for regeneration of dura mater.

Dura mater healing is essential to prevent cerebrospinal fluid (CSF) leaks in neurosurgical procedures. Drawing inspiration from the hierarchical structure of native dura mater, we have designed a biomimetic electrospun trilayered scaffold (TLS) utilizing silk fiber to replicate both the structure and function of the original tissue. The electrospun trilayered scaffold comprises three distinct layers: a skull-facing layer constructed from silk fibroin combined with strontium-doped bioactive glass, a gradient inert polyurethane middle layer and a brain-facing layer consisting of polyurethane in
