Natural compounds from freshwater mussels disrupt fungal virulence determinants and influence fluconazole susceptibility in the presence of macrophages in Cryptococcus neoformans.

UNLABELLED: Cryptococcus neoformans is a human fungal pathogen responsible for fatal infections, especially in patients with a depressed immune system. Overexposure to antifungal drugs due to prolonged treatment regimens and structure-similar applications in agriculture have weakened the efficacy of current antifungals in the clinic. The rapid evolution of antifungal resistance urges the discovery of new compounds that inhibit fungal virulence determinants, rather than directly killing the pathogen, as alternative strategies to overcome disease and reduce selective pressure toward resistance.

Therapeutic vaccine using a monoclonal antibody against a 70-kDa glycoprotein in mice infected with highly virulent Sporothrix schenckii and Sporothrix brasiliensis.

Sporotrichosis is a chronic granulomatous mycosis caused by the dimorphic fungi that comprise the Sporothrix complex. The latter are widely distributed in nature, developing a saprophytic mycelial form on plant debris and soil. Formerly, the S. schenckii species was thought to be the only species capable of causing sporotrichosis. However, in recent years, the existence of a group of highly genotypically and phenotypically variable species has been reported as etiologic agents of this mycosis. Recently, it has become important to study aspects such as virulence and the immune response against

1,3-β-d-Glucan synthase of Paracoccidioides brasiliensis: recombinant protein, expression and cytolocalization in the yeast and mycelium phases.

Paracoccidioides brasiliensis is a thermo-dimorphic human pathogenic fungus that in the mycelium phase lives at 23°C in environment and in the yeast phase at 37°C in the host tissues. In P. brasiliensis, the main polymers that compound the cell wall are chitin, 1,3-β-D-glucan and 1,3-α-glucan. They make a primary barrier responsible for the structural integrity and form of the cell wall. In P. brasiliensis, just one homologue of 1,3-β-D-glucan synthase gene (PbFKS1) was found. Here, the active recombinant protein (PbFks1pc) containing the catalytic region was obtained in Escherichia coli. In a

Structural switching of Arabidopsis QSOX1 defines stress-dependent chaperone and defense functions.

Quiescin sulfhydryl oxidase 1 (QSOX1) is a redox-responsive enzyme in Arabidopsis thaliana, previously characterized for its chaperone function under abiotic stress. Its role in pathogen defense, however, has remained unclear. Here, we show that recombinant QSOX1 exhibits structure-dependent antifungal activity. The low molecular weight (LMW) form suppressed the growth of diverse fungal pathogens at concentrations of 20-40 μg/mL, whereas the high molecular weight (HMW) form displayed little or no inhibitory effect. Confocal and scanning electron microscopy revealed that LMW-QSOX1 disrupts fung

Identification and characterization of an aspartyl protease from Cryptococcus neoformans.

Cryptococcosis, caused by Cryptococcus neoformans, is an invasive infection often occurring in AIDS patients. Potent therapy against HIV, which includes protease inhibitors (PIs), has beneficial effects also on opportunistic infections by pathogens such as C. neoformans and C. albicans. PIs inhibit growth of C. albicans by affecting the activity of its aspartyl proteases. We identified, cloned and sequenced a cDNA from C. neoformans encoding for a putative aspartyl protease (CnAP1), and the corresponding genomic region. The gene cnap1 codifies for a protein of 505 aa, with a canonical aspartyl

Ultrastructural Study of Cryptococcus neoformans Surface During Budding Events.

Cryptococcus neoformans is a fungal pathogen that causes life-threatening infections in immunocompromised individuals. It is surrounded by three concentric structures that separate the cell from the extracellular space: the plasma membrane, the cell wall and the polysaccharide (PS) capsule. Although several studies have revealed the chemical composition of these structures, little is known about their ultrastructural organization and remodeling during C. neoformans budding events. Here, by combining the latest and most accurate light and electron microscopy techniques, we describe the morpholo

Trapping conformational states of a flavin-dependent N-monooxygenase in crystallo reveals protein and flavin dynamics.

The siderophore biosynthetic enzyme A (SidA) ornithine hydroxylase from Aspergillus fumigatus is a fungal disease drug target involved in the production of hydroxamate-containing siderophores, which are used by the pathogen to sequester iron. SidA is an N-monooxygenase that catalyzes the NADPH-dependent hydroxylation of l-ornithine through a multistep oxidative mechanism, utilizing a C4a-hydroperoxyflavin intermediate. Here we present four new crystal structures of SidA in various redox and ligation states, including the first structure of oxidized SidA without NADP(H) or l-ornithine bound (re

Structural analysis of substrate recognition loop flexibility in D-arabinose dehydrogenase from Candida auris.

Candida auris is an emerging fungal pathogen that poses a significant threat to global health due to its multidrug resistance and ability to persist in healthcare settings. A key factor contributing to its survival and virulence is its capacity to combat oxidative stress, a process primarily driven by oxidative stress-related enzymes. One such enzyme, d-arabinose dehydrogenase from C. auris (CaAldO), plays a crucial role in the biosynthesis of d-erythroascorbic acid (EASC), an essential antioxidant that shields fungal cells from oxidative damage. CaAldO catalyzes the oxidation of d-arabinose t

Molecular characterization and virulence of fungal pathogens associated with mass mortalities in hilsa Shad (Tenualosa ilisha).

The rising incidence of invasive microbial infections and the expanding spectrum of pathogens make early and accurate identification of the causative pathogen a daunting task. Accurate diagnosis enables the identification and characterization of microbes and offers new possibilities for the control and prevention of infectious diseases. For the first time, we isolated and characterized three fungal species, viz., Mucor circinelloides, Fusarium equiseti, and F. incarnatum from hilsa (Tenualosa ilisha). The characterization was based on fungus morphology (in vitro culture and microscopy), molecu

Chemogenomic profiling predicts antifungal synergies.

Chemotherapies, HIV infections, and treatments to block organ transplant rejection are creating a population of immunocompromised individuals at serious risk of systemic fungal infections. Since single-agent therapies are susceptible to failure due to either inherent or acquired resistance, alternative therapeutic approaches such as multi-agent therapies are needed. We have developed a bioinformatics-driven approach that efficiently predicts compound synergy for such combinatorial therapies. The approach uses chemogenomic profiles in order to identify compound profiles that have a statisticall

Current advancements and future perspectives of 1,2,3-triazoles to target lanosterol 14α-demethylase (CYP51), a cytochrome P450 enzyme: A computational approach.

Antifungal resistance has become a significant challenge, necessitating the development of novel antifungal agents. Resistance often arises from prolonged and widespread use of existing treatments, leading to mutations in fungal enzymes that reduce drug efficacy. Amongst various scaffolds, 1,2,3-triazoles have emerged as antifungal agents due to their ability to bind effectively to fungal enzymes. This review examines the binding interactions of 1,2,3-triazoles with lanosterol 14α-demethylase (CYP51), an enzyme in Candida albicans (PDB IDs:5TZ1and5V5Z), highlighting their potential in fighting

Fusarium fruiting body microbiome member Pantoea agglomerans inhibits fungal pathogenesis by targeting lipid rafts.

Plant-pathogenic fungi form intimate interactions with their associated bacterial microbiota during their entire life cycle. However, little is known about the structure, functions and interaction mechanisms of bacterial communities associated with fungal fruiting bodies (perithecia). Here we examined the bacterial microbiome of perithecia formed by Fusarium graminearum, the major pathogenic fungus causing Fusarium head blight in cereals. A total of 111 shared bacterial taxa were identified in the microbiome of 65 perithecium samples collected from 13 geographic locations. Within a representat

Structure-Activity Relationship Profiling of Substituted N-Phthalimidopiperidine Analogs as Broad-Spectrum Antifungal Agents.

Invasive fungal infections are a major and escalating global health concern, frequently resulting in life-threatening systemic infection. The combination of limited therapeutic options, increasing mortality, and the rapid emergence of multidrug-resistant fungal pathogens highlights the critical need for novel antifungal agents. With this goal in mind, a high-throughput screening assay identified SLB1122168, and a subsequent structure-activity relationship study led to more potent analogs, including SSD2032594, which demonstrated fungicidal activity with a minimum inhibitory concentration of 1-

Exploring Long Arm Amide-Linked Side Chains in the Design of Antifungal Azole Inhibitors of Sterol 14α-Demethylase (CYP51).

The rise in fungal drug resistance has exacerbated the treatment of invasive fungal infections, most commonly caused by Candida. This research describes the synthesis of extended "long-arm" azole antifungals that were evaluated against wild-type and resistant fungal species. Biphenyl derivative 22 was the most effective derivative, displaying potent inhibitory activity against Saccharomyces, Candida, and Cryptococcus CYP51 enzymes, including in resistant strains, in comparison with posaconazole. The X-ray crystal structure of S-22 complexed with S. cerevisiae CYP51 showed a hydrogen bond betwe

Using genomics to understand the mechanisms of virulence and drug resistance in fungal pathogens.

Fungal pathogens pose an increasingly worrying threat to human health, food security and ecosystem diversity. To tackle fungal infections and improve current diagnostic and therapeutic tools it is necessary to understand virulence and antifungal drug resistance mechanisms in diverse species. Recent advances in genomics approaches have provided a suitable framework to understand these phenotypes, which ultimately depend on genetically encoded determinants. In this work, we review how the study of genome sequences has been key to ascertain the bases of virulence and drug resistance traits. We fo

Cell Rupture and Morphogenesis Control of the Dimorphic Yeast Candida albicans by Nanostructured Surfaces.

Nanostructured surfaces control microbial biofilm formation by killing mechanically via surface architecture. However, the interactions between nanostructured surfaces (NSS) and cellular fungi have not been thoroughly investigated and the application of NSS as a means of controlling fungal biofilms is uncertain. Cellular yeast such as Candida albicans are structurally and biologically distinct from prokaryotic microbes and therefore are predicted to react differently to nanostructured surfaces. The dimorphic opportunistic fungal pathogen, C. albicans, is responsible for most cases of invasive

Functional analysis of fungal polyketide biosynthesis genes.

Fungal polyketides have huge structural diversity from simple aromatics to highly modified complex reduced-type compounds. Despite such diversty, single modular iterative type I polyketide synthases (iPKSs) are responsible for their carbon skeleton construction. Using heterologous expression systems, we have studied on ATX, a 6-methylsalicylic acid synthase from Aspergillus terreus as a model iPKS. In addition, iPKS functions involved in fungal spore pigment biosynthesis were analyzed together with polyketide-shortening enzymes that convert products of PKSs to shorter ketides by hydrolytic C-C

Spontaneous feline sporotrichosis: a fine structural study.

Fine structural details of the parasitic yeastlike phase of Sporothrix schenckii contained in biopsy tissue from a naturally-occurring case of disseminated feline sporotrichosis are described and illustrated by electron microscopy. Both free and phagocytosed fungal cells were observed. The fungal cells were contained within an extracellular, electron transparent vacuolar area which was bounded by a limiting membrane of probable host origin. The yeastlike cells were characterized by a conspicuous layer of osmiophilic microfilaments which occurred along the outermost surface of the cell wall. In

The effect of nine-year nitrogen addition on foliar fungal diseases of plant groups in an alpine meadow.

INTRODUCTION: Foliar fungal diseases have become one of the non-negligible drivers of accelerated grassland degradation, and global change processes such as increased nitrogen (N) deposition can significantly regulate the occurrence and development of foliar fungal diseases in meadows. However, little is known about how increased nitrogen deposition affects foliar fungal diseases in alpine meadows, and there is limited understanding of the relative importance of different plant groups in the process of N enrichment affecting plant diseases. METHODS: Here, based on a 9-year continuous nitrogen
