Characterization and expression analysis of UBC gene family provide insights into the potential roles in pigment biosynthesis in pepper fruit.

BACKGROUND: Ubiquitin-conjugating enzymes (UBCs) are a critical part of the ubiquitin-proteasome pathway and play diverse roles in growth, development, and abiotic stress responses in pepper (Capsicum annuum L.). Although UBC genes have been reported in several plant species, genome-wide characterization of this gene family remains unexplored in pepper. RESULTS: In this study, 56 putative CaUBCs were identified in pepper, which were clustered into 11 groups based on phylogenetic analysis, including ubiquitin E2 enzyme variant (UEV), SOMO (HUS), and COP10. CaUBCs within each subgroup showed re

Metabolomics and transcriptomics combined with physiology reveal key metabolic pathway responses in tobacco roots exposed to NaHS.

Hydrogen sulfide (H2S) has emerged as a novel endogenous gas signaling molecule, joining the ranks of nitric oxide (NO) and carbon monoxide (CO). Recent research has highlighted its involvement in various physiological processes, such as promoting root organogenesis, regulating stomatal movement and photosynthesis, and enhancing plant growth, development, and stress resistance. Tobacco, a significant cash crop crucial for farmers' economic income, relies heavily on root development to affect leaf growth, disease resistance, chemical composition, and yield. Despite its importance, there remains

Coordinated Regulation of Iron Homeostasis, Antioxidant Defenses and Hormone Signaling Underlies Pepper Responses to Iron Deficiency and Excess.

Iron availability strongly influences crop performance, yet the mechanisms in non-model horticultural species remain unclear. We combined morphological, physiological, and root transcriptome analyses to dissect the response of Capsicum annuum L. cultivar CA#8 to iron deficiency, low iron, and excess iron. Seedlings grown hydroponically under four iron supplies (0, 5, 25, and 150 μM EDTA-Fe) showed distinct shoot and root phenotypes: iron deficiency caused interveinal chlorosis and strong inhibition of root growth, excess iron induced leaf wrinkling with white spots and reduced root development

Deletion of TaBSK3-2B attenuates BR signaling to regulate the Rht4 semi-dwarf phenotype and drought tolerance in wheat.

The application of Green Revolution alleles Rht-B1b and Rht-D1b in wheat semi-dwarf breeding has remarkably improved lodging resistance and increased global wheat production. However, these gibberellin (GA)-insensitive genes require higher water and fertilizer inputs and impair seedling emergence and early vigor, prompting the search for alternative GA-sensitive dwarfing genes to breed environmentally friendly wheat varieties for sustainable wheat production. Here, we reported the map-based cloning of Rht4 and functional characterization of its potential causal gene TaBSK3-2B, which encodes a

Bacillus velezensis promotes growth and flavonoid accumulation in Anoectochilus roxburghii via phytohormone signaling.

Beneficial Bacillus agents play crucial roles in promoting crop growth and metabolic processes by modulating phytohormone levels and increasing the availability of soil nutrients. This study investigated the phytohormone signaling pathways to elucidate the underlying mechanisms by which Bacillus velezensis promotes root development, nutrient uptake, and the accumulation of bioactive compounds in Anoectochilus roxburghii Lindl. It was found that B. velezensis promoted fine root development by upregulating genes involved in the synthesis of 3-indoleacetic acid (IAA) and downregulating genes invo

Genome-wide identification, classification, and expression analysis of heat shock transcription factor family in switchgrass (Panicum virgatum L.).

Switchgrass is one of the most promising bioenergy crops and is generally cultivated in arid climates and poor soils. Heat shock transcription factors (Hsfs) are key regulators of plant responses to abiotic and biotic stressors. However, their role and mechanism of action in switchgrass have not been elucidated. Hence, this study aimed to identify the Hsf family in switchgrass and understand its functional role in heat stress signal transduction and heat tolerance by using bioinformatics and RT-PCR analysis. Forty-eight PvHsfs were identified and divided into three main classes based on their

A CCaMK-CYCLOPS-DELLA Complex Activates Transcription of RAM1 to Regulate Arbuscule Branching.

Intracellular arbuscular mycorrhiza symbiosis between plants and glomeromycotan fungi leads to formation of highly branched fungal arbuscules that release mineral nutrients to the plant host. Their development is regulated in plants by a mechanistically unresolved interplay between symbiosis, nutrient, and hormone (gibberellin) signaling. Using a positional cloning strategy and a retrotransposon insertion line, we identify two novel alleles of Lotus japonicus REDUCED ARBUSCULAR MYCORRHIZA1 (RAM1) encoding a GRAS protein. We confirm that RAM1 is a central regulator of arbuscule development: arb

The sunflower HaHB4 gene confers enhanced tolerance to heat stress at the pre-anthesis stage in three different wheat cultivars.

The sunflower gene HaHB4 was previously described as conferring drought tolerance to Cadenza wheat plants, as assessed in 37 field trials. HaHB4-wheat is the first transgenic wheat trait approved for global markets. In this work, we show that transgenic plants also exhibit heat tolerance in the pre-anthesis stage. During 2023 and 2024, the experiments were conducted in open-air conditions, using the modern cultivars Algarrobo and Feroz by introgression of the original Cadenza HaHB4. The newly obtained transgenic wheat plants, particularly Algarrobo, as well as the original Cadenza HaHB4, maint

Genome-wide identification of alcohol dehydrogenase (ADH) gene family in oilseed rape (Brassica napus L.) and BnADH36 functional verification under salt stress.

BACKGROUND: Alcohol dehydrogenase (ADH) is an enzyme that binds to zinc, facilitating the interconversion of ethanol and acetaldehyde or other corresponding alcohols/aldehydes in the pathway of ethanol fermentation. It plays a pivotal role in responding to environmental stress. However, the response of the ADH family to abiotic stress remains unknown in rapeseed. RESULT: In this study, we conducted a comprehensive genome-wide investigation of the ADH family in rapeseed, encompassing analysis of their gene structure, replication patterns, conserved motifs, cis-acting elements, and response to

Genetic Analysis and Fine Mapping of a New Rice Mutant, Leaf Tip Senescence 2.

Premature leaf senescence significantly reduces rice yields. Despite identifying numerous factors influencing these processes, the intricate genetic regulatory networks governing leaf senescence demand further exploration. We report the characterization of a stably inherited, ethyl methanesulfonate(EMS)-induced rice mutant with wilted leaf tips from seedling till harvesting, designated lts2. This mutant exhibits dwarfism and early senescence at the leaf tips and margins from the seedling stage when compared to the wild type. Furthermore, lts2 displays a substantial decline in both photosynthet

Overexpression of the Poa pratensis GA2ox gene family significantly reduced the plant height of transgenic Arabidopsis thaliana and Poa pratensis.

Gibberellin (GAs) is an important plant hormone that plays a key role in plant growth and development. Gibberellin 2-oxidase (GA2ox) catalyzes the inactivation of biologically active GA or their direct precursors. In this study, five GA2ox genes were isolated from the wild type Poa pratensis 'Baron', named PpGA2ox3, PpGA2ox4, PpGA2ox5, PpGA2ox8, and PpGA2ox9. Phylogenetic tree analysis showed that PpGA2ox3, PpGA2ox4, PpGA2ox5, and PpGA2ox8 belong to class I GA2ox genes, while PpGA2ox9 belongs to class III GA2ox genes. They expressed in all tissues of Poa pratensis, in each plant tissue and gro

Characterization of cassava ORANGE proteins and their capability to increase provitamin A carotenoids accumulation.

Cassava (Manihot esculenta Crantz) biofortification with provitamin A carotenoids is an ongoing process that aims to alleviate vitamin A deficiency. The moderate content of provitamin A carotenoids achieved so far limits the contribution to providing adequate dietary vitamin A levels. Strategies to increase carotenoid content focused on genes from the carotenoids biosynthesis pathway. In recent years, special emphasis was given to ORANGE protein (OR), which promotes the accumulation of carotenoids and their stability in several plants. The aim of this work was to identify, characterize and inv

Molecular characterization of rice sphingosine-1-phosphate lyase gene OsSPL1 and functional analysis of its role in disease resistance response.

Our results indicate that overexpression of OsSPL1 in transgenic tobacco plants attenuated disease resistance and facilitated programmed cell death. Long-chain base phosphates including sphingosine-1-phosphate have been shown to act as signaling mediators in regulating programmed cell death (PCD) and stress responses in mammals. In the present study, we characterized a rice gene OsSPL1, encoding a putative sphingosine-1-phosphate lyase that is involved in metabolism of sphingosine-1-phosphate. Expression of OsSPL1 was down-regulated in rice plants after treatments with salicylic acid, benzothi

Overexpression of OsF3H modulates WBPH stress by alteration of phenylpropanoid pathway at a transcriptomic and metabolomic level in Oryza sativa.

The whitebacked planthopper (WBPH), has become a devastating pest for rice crops, causes serious yield losses each year, and urgently needs biological control. Here, we developed a WBPH-resistant rice cultivar by overexpressing the OsF3H gene. A genetic functional analysis of the OsF3H gene confirmed its role in facilitating flavonoid contents and have indicated that the expression of the OsF3H gene is involved in regulation of the downstream genes (OsDFR and OsFLS) of the flavonoid pathway and genes (OsSLR1 and OsWRKY13) involved in other physiological pathways. Comparative analysis of DDR-related genes and microRNA expression during rice germination: Implications for salinity susceptibility screening.

Soil salinity poses a significant threat to the agri-food sector and particularly to rice cultivation. High salinity during germination induces overproduction of reactive oxygen species (ROS) that cause lesions in the DNA resulting in reduced vigor. MicroRNAs (miRNAs) are known to modulate stress response in plants, however, studies focusing on its relation with the expression of the DNA damage response (DDR)-related genes are not thoroughly explored. In this regard, the aim of this work was to investigate the link between the expression of miRNAs and putative targeted DDR-related genes in res

Targeted Gene Expression Modulation Using CRISPR/dCas9 to Investigate Pathogenic Outcomes in Tomato.

CRISPR (clustered regularly interspaced short palindromic repeats) has become integral to modern biological research, with the Streptococcus pyogenes CRISPR/Cas9 system serving as the most extensively used tool for precise, site-specific genome editing across a wide range of organisms and cell types. Compared with earlier genome-editing platforms, such as zinc-finger nucleases (ZFNs) and transcription activator-like effector nucleases (TALENs), CRISPR/Cas9 offers greater simplicity, precision, versatility, and scalability. Beyond introducing stable DNA modifications, this system can be reengin

Rice cold tolerance: Physiological responses, molecular regulatory networks, and improvement strategies.

Low-temperature stress is a critical abiotic constraint that severely impairs rice yield and quality, posing a substantial threat to global food security-especially as rice, a staple food for more than half of the world's population, is inherently susceptible to cold stress. This review systematically summarizes the typical phenotypic and physiological impairments induced by cold stress across key rice growth stages, and elaborates on the multi-layered molecular regulatory networks underpinning rice cold tolerance, which integrate the entire process of cold signal perception, intracellular tra

PAL-mediated SA biosynthesis pathway contributes to nematode resistance in wheat.

The pathogen cereal cyst nematode (CCN) is deleterious to Triticeae crops and is a threat to the global crop yield. Accession no. 1 of Aegilops variabilis, a relative of Triticum aestivum (bread wheat), is highly resistant to CCN. Our previous study demonstrated that the expression of the phenylalanine ammonia lyase (PAL) gene AevPAL1 in Ae. variabilis is strongly induced by CCN. PAL, the first enzyme of phenylpropanoid metabolism, is involved in abiotic and biotic stress responses. However, its role in plant-CCN interaction remains unknown. In the present study, we proved that AevPAL1 helps t
