Quantitative genetic analysis of injury liability in infants and toddlers.

A threshold model of latent liability was applied to infant and toddler twin data on total count of injuries sustained during the interval from birth to 36 months of age. A quantitative genetic analysis of estimated twin correlations in injury liability indicated strong genetic dominance effects, but no additive genetic variance was detected. Because interpretations involving overdominance have little research support, the results may be due to low order epistasis or other interaction effects. Boys had more injuries than girls, but this effect was found only for groups whose parents were promp

Conversations with French medical geneticists. A personal perspective on the origins and early years of medical genetics in France.

The history of the beginnings of medical genetics in France is discussed, based on the personal perspective provided by recorded interviews with 16 early French workers in the field. The weakness of French genetics overall up to the beginning of the Second World War meant that post-war medical genetics had to start from new, with its origins largely derived from the medical fields of child health and the prevention of genetic disorders, rather than from basic science. The key people responsible for initiating these developments were Robert Debré and Maurice Lamy at Hôpital Necker in Paris and

CRISPR-Cas9 and precision editing technologies linking functional genomics to clinical translation in genetic diseases.

BACKGROUND: CRISPR-Cas9 and derivative precision-editing platforms increasingly connect pathogenic variant interpretation with functional genomics and therapeutic development in genetic diseases. This narrative review focuses on a variant-mechanism-driven framework for matching editing strategies to mutation structure, functional consequence, disease-model evidence, delivery feasibility, safety risk, and translational readiness. MAIN BODY: The review summarizes how monogenic, polygenic, coding, non-coding, mitochondrial, and complex disease contexts influence the choice of canonical Cas9 edit

Scoping review of Australian prioritisation frameworks impacting consumer access to clinical genetics: protocol.

INTRODUCTION: Timely access to care is critical, yet prolonged waiting lists across healthcare systems cause avoidable harm and inequity, visible to clinicians who experience moral distress when patients wait unsafely. While equity is embedded in policy, performance metrics may obscure these realities. Clinical genetics exemplifies the issue: delays in assessment, diagnosis and reproductive planning are compounded by inconsistent prioritisation processes across states and subspecialties. There is no national consensus on urgency criteria, and frameworks often overlook consumer values such as t

Ocular gene therapy targeting retinal angiogenesis and vascular leakage: translational and clinical evidence from a systematic review and meta-analysis.

INTRODUCTION: Retinal diseases driven by pathological angiogenesis and vascular leakage, including neovascular age-related macular degeneration and diabetic retinopathy, impose substantial visual and treatment burdens because current anti-vascular endothelial growth factor (anti-VEGF) therapy requires repeated intravitreal administration. Ocular gene therapy can provide durable intraocular expression of therapeutic proteins and sustained pathway-level disease control. METHODS: This systematic review and meta-analysis was prospectively registered in the International Prospective Register of Sy

The PreGen Research Program: Implementing Prenatal Genomic Testing in Australia-A Commentary.

Prenatal genomic sequencing, which can provide a significantly increased diagnostic rate for fetal structural anomaly (FSA) compared with karyotype and microarray, is not available uniformly across Australia. PreGen, a 5-year translational research program, has identified significant barriers to implementation including access to funding, the availability of genomic testing, access to termination of pregnancy services and the availability of specialist genomic centres. A federal item number for prenatal genomic testing would increase equitable test availability and reduce delays to diagnoses b

A qualitative investigation of Ehlers-Danlos syndrome genetics triage.

The Ehlers-Danlos syndromes (EDS) are a group of hereditary connective tissue disorders for which patients struggle to find a timely and accurate diagnosis. Overburdened genetics practices have been identified as a barrier to care. Triage is a tool utilized by genetics practices to address workload issues and prioritize high-risk patients. Through a demographic survey and semi-structured interviews, this study aimed to explore what triages exist for EDS referrals to genetics practices across the United States and how genetic counselors perceive their triages. Inductive coding and conventional

"I can't help them like I used to": Evolving prenatal genetic counseling practices in a post-Roe U.S.

This exploratory empirical study documents how prenatal genetic counseling practice was evolving one year after the 2022 Supreme Court decision overturning the constitutional right to abortion in Dobbs v. Jackson Women's Health Organization. This study captures a moment when prenatal genetic counselors, clinicians who provide education and counseling about genetic and other fetal health issues before and during pregnancy, were navigating new state abortion restrictions. Qualitative interviews were conducted with 27 genetic counselors in 15 states representing a range of abortion regulations. T

Challenges with scale-up of GeneXpert MTB/RIF® in Uganda: a health systems perspective.

BACKGROUND: Many high burden countries are scaling-up GeneXpert® MTB/RIF (Xpert) testing for tuberculosis (TB) using a hub-and-spoke model. However, the effect of scale up on reducing TB has been limited. We sought to characterize variation in implementation of referral-based Xpert TB testing across Uganda, and to identify health system factors that may enhance or prevent high-quality implementation of Xpert testing services. METHODS: We conducted a cross-sectional study triangulating quantitative and qualitative data sources at 23 community health centers linked to one of 15 Xpert testing si

Ethnography/ethics.

This paper situates discussion of the ethics of ethnographic research against the background of a theoretical and methodological debate about the relationship between ethics and method, and about the relationships between research methods and their objects. In particular, the paper investigates the implications of folding together the ethical and the empirical in research and argues that this requires the development of new ethico-ethnographic methods for the investigation of ethico-moral objects. The paper falls into three main parts. The first considers calls for what has come to be known as

Direct-to-consumer genetic testing for addiction susceptibility: a premature commercialisation of doubtful validity and value.

Genetic research on addiction liability and pharmacogenetic research on treatments for addiction have identified some genetic variants associated with disease risk and treatment. Genetic testing for addiction liability and treatment response has not been used widely in clinical practice because most of the genes identified only modestly predict addiction risk or treatment response. However, many of these genetic tests have been commercialized prematurely and are available direct to the consumer (DTC). The easy availability of DTC tests for addiction liability and lack of regulation over their …

Mexico City Prospective Study: cohort profile update.

To fully understand the causes of human disease worldwide, large blood based prospective studies are needed in a range of populations with different healthcare settings and exposures to environmental and genetic risk factors. The Mexico City Prospective Study, updated in this cohort profile, was established in the late 1990s to investigate the sociodemographic, lifestyle, physical, and biological (including genetic) determinants of disease among around 150 000 adults in Mexico City. The study has over 20 years of mortality follow-up, cohort-wide biochemical and genetic data, repeat survey data

Genomics May Be the Key to Understanding Endurance Training Pillars.

Endurance performance is primarily determined by three key physiological pillars: maximal oxygen uptake (VO2max), anaerobic threshold, and economy of movement. Recent research has suggested physiological resilience as a potential fourth dimension, referring to an athlete's ability to sustain performance despite accumulating fatigue. While the role of genetic factors in endurance has been widely studied, their influence on these pillars, particularly on fatigue resistance and long-term adaptation, remains an area of growing interest. This narrative review explores the genomic basis of endurance

Community leaders' perspectives on engaging African Americans in biobanks and other human genetics initiatives.

There is limited information about what African Americans think about biobanks and the ethical questions surrounding them. Likewise, there is a gap in capacity to successfully enroll African Americans as biobank donors. The purposes of this community-based participatory study were to: (a) explore African Americans' perspectives on genetics/genomic research, (b) understand facilitators and barriers to participation in such studies, and (c) enlist their ideas about how to attract and sustain engagement of African Americans in genetics initiatives. As the first phase in a mixed methods study, we

[Genetic factors are often found inl Alzheimer disease. An extensive twin study to clarify the heredity-environment relationship].

Genetic and environmental risk factors are central concerns in dementia research. Through twin studies one can study the relative importance of genetic and environmental risk factors. One such study, based on the Swedish Twin Registry, shows that genetic effects are of considerable importance for developing Alzheimer's disease, even late in life, but that environmental factors are also important. For vascular dementia, the results indicate that non-genetic factors are of primary importance.

Genetic Panel Screening of Nearly 100 Mutations Reveals New Insights into the Breed Distribution of Risk Variants for Canine Hereditary Disorders.

BACKGROUND: The growing number of identified genetic disease risk variants across dog breeds challenges the current state-of-the-art of population screening, veterinary molecular diagnostics, and genetic counseling. Multiplex screening of such variants is now technologically feasible, but its practical potential as a supportive tool for canine breeding, disease diagnostics, pet care, and genetics research is still unexplored. RESULTS: To demonstrate the utility of comprehensive genetic panel screening, we tested nearly 7000 dogs representing around 230 breeds for 93 disease-associated variant

Central Sympathetic Nerve Activation-Mediated Hypertension: Target Mechanisms and Multimodal Interventions-From Basic Research to Clinical Translation.

Hypertension is the leading global risk factor for cardiovascular diseases, and its pathogenesis is closely linked to excessive sympathetic activation, which markedly elevates the risk of stroke, heart failure and other adverse cardiovascular events. Traditional therapies mainly target peripheral mechanisms, whereas the clinical efficacy of renal denervation highlights the critical role of central regulation in sympathetic hyperactivity. This review focuses on the core sympathetic nuclei including the rostral ventrolateral medulla (RVLM) and paraventricular nucleus (PVN), with epigenetic regul

Epigenetic mechanisms in preeclampsia: translational therapeutic strategies and precision-medicine perspectives.

Preeclampsia is increasingly understood not only as a clinical syndrome of hypertension and organ dysfunction, but also as a disorder in which placental gene regulation, maternal vascular adaptation, and inflammatory signaling are shaped by abnormal epigenetic control. While several reviews have described epigenetic biomarkers in preeclampsia, the therapeutic implications of these mechanisms remain less clearly integrated. This review, therefore, focuses on the translational potential of epigenetic pathways as therapeutic entry points, with particular attention to DNA methylation, histone regu
