Association of a polygenic risk score with low trauma fractures in people with HIV - The swiss HIV cohort study.

BACKGROUND: Polygenic risk scores (PRS) are likely to enter routine clinical care for individual disease risk prediction in the next 10 years. We recently showed that the bone mineral density-associated gSOS-PRS is independently associated with a > 4-fold increased risk of osteoporosis in the Swiss HIV Cohort Study (SHCS). Here we investigate whether this PRS is also associated with low trauma fractures (LTF) in people with HIV in the SHCS. METHODS: Applying a case-control design, cases had a first LTF (1994-2022) and LTF-free controls were matched on age, sex and observation time. We obtaine

Interaction between coffee consumption and polygenic risk score in relation to diabetes: insights from the Maastricht study.

AIMS: This study investigated the associations of polygenic risk score (PRS) and coffee consumption, as well as their interaction, with prediabetes and type 2 Diabetes Mellitus (T2DM) among participants in the southern Netherlands. METHODS: 7668 participants were classified as normal glucose metabolism (NGM), prediabetes, or T2DM based on World Health Organization 2006 criteria. PRS (423 T2DM-related single nucleotide polymorphisms) and coffee consumption (via food frequency questionnaire) were categorized into tertiles (low, medium, and high) based on the population distribution. Multinomial

Benchmarking Alzheimer's disease prediction: personalised risk assessment using polygenic risk scores across various methodologies and genome-wide studies.

BACKGROUND: The success of selecting high risk or early-stage Alzheimer's disease individuals for the delivery of clinical trials depends on the design and the appropriate recruitment of participants. Polygenic risk scores (PRS) show potential for identifying individuals at risk for Alzheimer's disease (AD). Our study comprehensively examines AD PRS utility using various methods and models. METHODS: We compared the PRS prediction accuracy in ADNI (N = 568) and BioFINDER (N = 766) cohorts using five disease risk modelling approaches, three PRS derivation methods, two AD genome-wide association

Health status in patients at risk of inherited arrhythmias and sudden unexpected death compared to the general population.

BACKGROUND: The possibilities in the molecular genetics of long QT syndrome (LQTS) and hypertrophic cardiomyopathy (HCM) has made family screening, with diagnostic and predictive genetic testing part of the health care offer in genetic counselling of inherited arrhythmias, potentially affecting the subjective health among these individuals. The study compared health status among patients at risk of arrhythmia because of family history or clinical diagnosis of LQTS and HCM with reference health status scores of the general population. METHODS: In the period 2005-2007, 127 patients (mean age 45

Measuring heritable contributions to Alzheimer's disease: polygenic risk score analysis with twins.

The heritability of Alzheimer's disease estimated from twin studies is greater than the heritability derived from genome-based studies, for reasons that remain unclear. We apply both approaches to the same twin sample, considering both Alzheimer's disease polygenic risk scores and heritability from twin models, to provide insight into the role of measured genetic variants and to quantify uncaptured genetic risk. A population-based heritability and polygenic association study of Alzheimer's disease was conducted between 1986 and 2016 and is the first study to incorporate polygenic risk scores i

Genome-wide analyses identify 60 loci associated with liver triglyceride content.

We performed a large-scale genome-wide association study meta-analysis of magnetic resonance imaging-derived liver triglyceride content across 71,488 European-ancestry participants from the UK Biobank, approximately doubling the sample size of previous genome-wide association studies1. We identified 60 independent loci, including 31 not previously reported, and estimated the single nucleotide polymorphism-based heritability of liver triglyceride content at 25%. Integrative fine-mapping and gene-prioritization analyses nominated candidate effector genes for 48 loci. Of the 31 newly identified v

Integrating contextual determinants and polygenic risk to examine dementia and late-life cognition in the Multi-Ethnic Study of Atherosclerosis.

INTRODUCTION: Alzheimer's disease and related dementias are influenced by genetic and environmental risk factors. We investigated the relationship between contextual exposures and cognitive outcomes, independent of and in interaction with polygenic risk. METHODS: Using the Multi-Ethnic Study of Atherosclerosis (N = 5687), we assessed the associations of contextual determinants representing the social, chemical, and built environment with incident dementia and late-life cognition using proportional hazards regression and generalized estimating equation models, then evaluated their joint effect

Joint effect of genetic and lifestyle risk factors on type 2 diabetes risk among Chinese men and women.

More than 40 genetic susceptibility loci have been reported for type 2 diabetes (T2D). Recently, the combined effect of genetic variants has been investigated by calculating a genetic risk score. We evaluated 36 genome-wide association study (GWAS) identified SNPs in 2,679 T2D cases and 3322 controls in middle-age Han Chinese. Fourteen SNPs were significantly associated with T2D in analysis adjusted for age, sex and BMI. We calculated two genetic risk scores (GRS) (GRS1 with all the 36 SNPs and GRS2 with the 14 SNPs significantly associated with T2D). The odds ratio for T2D with each GRS point…

Joint exposure to multiple air pollutants, genetic risk and incident psoriasis: a large-scale prospective cohort study.

BACKGROUND: Air pollution and genetic risk have been found to contribute to the onset and development of psoriasis. However, the extent to which genetic susceptibility modifies the effects of air pollutants on the risk of incident psoriasis remains unknown. OBJECTIVES: To assess the association between joint exposure to multiple air pollutants and the risk of psoriasis, and its modification, according to genetic susceptibility. METHODS: This prospective study included 451 064 participants from the UK Biobank who had complete air pollution data and were free of psoriasis at baseline. All part

Early-onset Alzheimer's disease explained by polygenic risk of late-onset disease?

Early-onset Alzheimer's disease (AD) is highly heritable, yet only 10% of cases are associated with known pathogenic mutations. For early-onset AD patients without an identified autosomal dominant cause, we hypothesized that their early-onset disease reflects further enrichment of the common risk-conferring single nucleotide polymorphisms associated with late-onset AD. We applied a previously validated polygenic hazard score for late-onset AD to 193 consecutive patients diagnosed at our tertiary dementia referral center with symptomatic early-onset AD. For comparison, we included 179 participa

Associations of Sleep Pattern and Genetic Risk with Late-Onset Psoriasis: A Large Prospective Cohort Study.

Whether sleep disturbance is associated with the risk of incident psoriasis, and whether this association exists in the presence of different genetic risk levels remain unknown. To examine the sleep-gene interaction in association with risk of psoriasis, we conducted a prospective cohort study that recruited >500,000 participants from 2006 to 2010 using data from the UK Biobank. A sleep pattern was defined by chronotype, sleep duration, insomnia, snoring, and daytime sleepiness and was categorized into 3 groups. In addition, we constructed a weighted genetic risk score of psoriasis and also ca…

Schizophrenia liability shares common molecular genetic risk factors with sleep duration and nightmares in childhood.

Background: Sleep abnormalities are common in schizophrenia, often appearing before psychosis onset; however, the mechanisms behind this are uncertain. We investigated whether genetic risk for schizophrenia is associated with sleep phenotypes. Methods: We used data from 6,058 children and 2,302 mothers from the Avon Longitudinal Study of Parents and Children (ALSPAC). We examined associations between a polygenic risk score for schizophrenia and sleep duration in both children and mothers, and nightmares in children, along with genetic covariances between these traits. Results: Polygenic risk f

A Bidirectional Mendelian Randomization Study to evaluate the causal role of reduced blood vitamin D levels with type 2 diabetes risk in South Asians and Europeans.

CONTEXT: Multiple observational studies have reported an inverse relationship between 25-hydroxyvitamin D concentrations (25(OH)D) and type 2 diabetes (T2D). However, the results of short- and long-term interventional trials concerning the relationship between 25(OH)D and T2D risk have been inconsistent. OBJECTIVES AND METHODS: To evaluate the causal role of reduced blood 25(OH)D in T2D, here we have performed a bidirectional Mendelian randomization study using 59,890 individuals (5,862 T2D cases and 54,028 controls) from European and Asian Indian ancestries. We used six known SNPs, including
