Genetic heterogeneity and population structure of Gond-related tribes in the Vidarbha region of Maharashtra.

Genetic heterogeneity in nine polymorphic loci is observed among Gond-related tribes in the Vidarbha region of Maharashtra. Pardhans, with their high ABO*A2 gene frequency (4.01%), low m gene frequency (57%), high P*1 gene frequency (42.7%), and high HbS trait (31.58%), differ significantly from other tribes. Per locus average heterozygosity among the studied tribes ranged from 36.24% to 40.37%, with Pardhans being more heterozygous. Analysis by FST and the empirical relationship between average allele frequencies and the ratio of within-gene to total gene diversity show that the tribes are is

Population and genetic studies suggest a single origin for the Indian deletion beta thalassaemia.

In a study of beta thalassaemia in the Asian Indian immigrant populations in the U.K., 23 out of 125 beta-thalassaemic chromosomes (18%) were of the Indian deletion beta type (600 bp deletion involving the 3' end). The individuals with beta thalassaemia had originated from various parts of India and Pakistan. However, all those individuals with deletion beta thalassaemia were from Sind and the adjacent area of Gujarat. Analysis of restriction fragment length polymorphisms in the beta globin gene cluster showed that all the 23 deletion beta thalassaemia chromosomes had an identical haplotype. T

A statistical test for detection of ancestral genetic contributions to disease occurrence in finite populations.

We have been investigating cancer patterns in the Hutterites, a religious isolate residing on communal farms [Martin et al, 1980a,b,c]. Advantages of this population include extensive and accurate genealogies, essentially complete case ascertainment, and relatively uniform environmental background with regard to geographic location, lifestyle, and diet. However, application of the common epidemiological approach to familial risk assessment based on incidence in relatives of index cases [Haenszel, 1959] posed difficulties because all cases are probands. We also planned to employ matching to con

Genetic and environmental determinants of growth of school-aged children in a rural Colombian population.

Parent-offspring correlations and heritabilities of body measurements from midparent-offspring regressions are presented for school-aged children from the village of Tenza, Colombia (N = 403 families). Parent-child correlations and midparent regressions in this subsistence farming sample, are similar in magnitude to those for well nourished, urban industrial samples, suggesting that the environmental component of variability in body size is the same regardless of the environment. Tenza children are significantly shorter and lighter than upper class Bogota children, and Tenza parents have mean

Genetics of type 2 diabetes and coronary artery disease and their associations with twelve cardiometabolic traits in the United Arab Emirates population.

BACKGROUND: The United Arab Emirates (UAE) population has a high rate of type 2 diabetes mellitus (T2DM) and other metabolic risk factors for coronary artery disease (CAD). Previous studies have indicated strong genetic associations between T2DM and CAD. The objective of this study was to replicate previously reported significant genetic associations for T2DM and CAD which were in a genome-wide significance level in a cohort from the Arab population of the UAE, and to investigate the associations of these loci with twelve cardiometabolic traits that may influence the development of T2DM and CA

Population genetics of Neisseria gonorrhoeae in a high-prevalence community using a hypervariable outer membrane porB and 13 slowly evolving housekeeping genes.

Baltimore, Md., is an urban community with a high prevalence of Neisseria gonorrhoeae. Due to partially protective immune responses, introduction of new strains from other host populations, and exposure of N. gonorrhoeae to antibiotics, the phenotypic and genotypic characteristics of the circulating strains can fluctuate over time. Understanding the overall genetic diversity and population structure of N. gonorrhoeae is essential for informing public health interventions to eliminate this pathogen. We studied gonococci population genetics in Baltimore by analyzing a hypervariable and strongly

Who declines genetic counseling? An assessment of genetic counseling declination among individuals participating in a population-wide genomic screening program.

OBJECTIVES: Population-wide genomic screening (PWGS) programs can identify individuals at increased risk for Hereditary Breast and Ovarian Cancer (HBOC), Lynch Syndrome (LS), and Familial Hypercholesterolemia (FH). However, uptake of follow-up genetic counseling (GC) remains inconsistent, and limited data describe why individuals decline these services. This study aimed to characterize patterns of GC uptake and declination within a statewide PWGS program and understand participant-reported reasons for declining GC as documented by study staff. METHODS: We analyzed GC uptake among 603 PWGS par

Chromosomal genome and population genetic analyses to reveal genetic architecture, breeding history and genes related to cadmium accumulation in Lentinula edodes.

BACKGROUND: Lentinula edodes (Berk.) is the second most productive mushroom in the world. It contains compounds effective for antiviral, antitumor, antioxidant and immune regulation. Although genomes have previously been reported for this species, a high-quality chromosome-level reference for L. edodes is unavailable. This hinders detailed investigation of population genetics, breeding history of strains and genes related to environmental stress responses. RESULTS: A high-quality chromosome-level genome was constructed. We separated a monokaryon from protoplasts of the commercial L. edodes st

Genetic Diversity and Population Genetic Structure Analysis of Plasmodium knowlesi Thrombospondin-Related Apical Merozoite Protein (TRAMP) in Clinical Samples.

The simian malaria parasite Plasmodium knowlesi causes a high number of zoonotic infections in Malaysia. The thrombospondin-related apical merozoite protein (TRAMP) is an essential ligand for binding to the erythrocyte cell surface, whereby it facilitates the invasion. This study is the first attempt to determine the genetic diversity, phylogeography, natural selection and population structure from 97 full-length PkTRAMP gene sequences originating from Malaysia. We found low levels of nucleotide diversity (π~0.0065) for the full-length gene despite samples originating from geographically separ

Genetic and non-genetic factors affecting hemoglobin A2 expression in a large cohort of Thai individuals: implication for population screening for thalassemia.

OBJECTIVE: Increased hemoglobin (Hb) A2 level is an important diagnostic marker for β-thalassemia carrier screening. The level of Hb A2 is also useful for differentiating several thalassemia syndromes. We have examined data bases for reduced Hb A2 expression in a large cohort of Thai subjects. METHODS: A study was done on 1,498 subjects with non-thalassemia and various types of thalassemia and Hb variants to determine the effect of thalassemia genotypes and on 103 women of reproductive age to determine the effect of iron deficiency. Hb analysis was done using capillary electrophoresis, and th

Intraspecific genetic variation in the lymphatic filariasis vector Mansonia dives (Diptera: Culicidae) in Thailand: Hidden species or genetically divergent populations?

Mansonia dives is recognized as a vector for brugian filariasis in Thailand. A recent study analyzing the cytochrome c oxidase subunit I (COI) gene revealed two distinct clades within the Ma. dives population in Thailand. This study aimed to examine the genetic diversity and structure of Ma. dives using the COI gene and the internal transcribed spacer 2 (ITS2) region to determine the presence of distinct species or genetically divergent populations. We analyzed 60 COI and 60 ITS2 sequences from Ma. dives populations in Narathiwat, Ranong, Tak, and Trat. The results showed a nucleotide diversit

A phenome-wide association study of cross-disorder genetic liability in youth genetically similar to individuals from European reference populations.

Etiologic insights into psychopathology may be gained by using hypothesis-free methods to identify associations between genetic risk for broad psychopathology and phenotypes measured during adolescence, including both markers of child psychopathology and intermediate phenotypes such as neural structure that may link genetic risk with outcomes. We conducted an exploratory phenome-wide association study (phenotype n=1,271-1,697) of polygenic risk scores (PRS) for broad spectrum psychopathology (i.e., Compulsive, Psychotic, Neurodevelopmental, and Internalizing) in youth of PCA-selected European

Genetic and non-genetic factors influencing efavirenz population pharmacokinetics among human immunodeficiency virus-1-infected children in Ethiopia.

Despite the potential for efavirenz (EFV) to be an effective alternative antiretroviral agent, its sources of wide inter- and intra-individual pharmacokinetic (PK) variability are not well-characterized in children. We investigated the effects of genetic and non-genetic factors, including demographic, treatment duration, baseline clinical, and biochemical characteristics, on the PKs of EFV through population-PK modeling. Antiretroviral therapy (ART) naïve HIV infected children, 3-16 years (n = 100), were enrolled in Ethiopia and received EFV-based combination ART. EFV concentrations after the

Genetic diversity, population structure, and interspecific differentiation in introduced breeding populations of Eucalyptus sideroxylon and Eucalyptus camaldulensis established in northern Chile.

BACKGROUND: Breeding populations of Eucalyptus sideroxylon and Eucalyptus camaldulensis have been established in the Coquimbo Region of northern Chile, immediately south of the Atacama Desert, owing to their ecological adaptability and tangible economic value. However, their genetic resources remain poorly characterized outside their native ranges. This study aimed to characterize genetic diversity, intraspecific population structure, and interspecific differentiation using a high-density 72K SNP array in a mixed-species breeding trial established in northern Chile. RESULTS: After stringent q

Genetic association of NOS3 rs1799983 variant and hypertension across multiple rural Indonesian populations: a multi-ethnic population-based study.

INTRODUCTION: Hypertension is a complex polygenic disorder where genetic variants become clinically relevant primarily when they contribute to endothelial dysfunction. While the NOS3 rs1799983 variant is known to impair endothelial function, data regarding its genotypic and allelic frequency and association with hypertension in Indonesian rural populations remains limited. The present study aimed to investigate the genotypic and allelic frequency of NOS3 rs1799983 and examine its association with hypertension across multiple ethnic groups in rural Indonesian populations. Allele and extended haplotype polymorphism of HLA-A, -C, -B, -DRB1 and -DQB1 loci in Polish population and genetic affinities to other populations.

Human leukocyte antigen (HLA)-A, -C, -B, -DRB1 and -DQB1 alleles were typed in 200 Polish healthy volunteers recruited for stem cell donor registry, using sequence-specific primer (SSP) and direct sequencing-based methods. Enhanced Bayesian approach of expectation maximization algorithm provided by phase platform was used for extended HLA haplotype inferences. The numbers of identified alleles (four-digit resolution) were 23, 23, 44, 27 and 18 alleles in HLA-A, -C, -B, -DRB1 and -DQB1 loci, respectively, of both northern and southern European frequency characteristics. The most frequent extend

Discovery of a Virulent Population of Meloidogyne graminicola Overcoming the Rice Resistance Gene MG1 and Population Genetic Analysis Based on mtCOI Sequences.

The rice root-knot nematode (Meloidogyne graminicola) is a major pathogen of rice. The resistance gene MG1 in the cultivar Zhonghua 11 (ZH11) confers effective resistance against this pathogen. To date, only one virulent population overcoming ZH11 has been reported, from Cambodia, but whether it overcomes MG1 remains unknown. Here, we collected and identified 31 M. graminicola populations from southern China. Ten of these populations were tested on ZH11. Two of the 10 tested populations were virulent, GZSD1 from Guangdong and HUNSD28 from Hunan, with reproduction factors (Rf) of 3.11 and 1.67,
