OBJECTIVE: In animals, nicotine, the primary psychoactive constituent of tobacco smoke, reduces neurogenesis and increases cell loss in both hippocampus and cortex. Accordingly, tobacco smoking has been linked to reduced performance on cognitive paradigms requiring attention and working memory in humans. However, few prior studies have tested for evidence of structural brain alterations in human tobacco smokers. In this study, proton magnetic resonance spectroscopy was used to assess the effects of chronic smoking on neuronal integrity of the hippocampus and anterior cingulate cortex (ACC).

Previous neuroimaging studies showed that use of marijuana can alter patterns of cortical activation during rest or a task challenge. We used functional magnetic resonance imaging to examine whether recent cannabis abuse contributed to stress-induced blood-oxygen-level-dependent (BOLD) contrast in a group of cocaine-dependent individuals. Emotional stress was induced using the script-guided imagery paradigm, in which subjects imagined being in a real-life stressful situation and, as a control, in a neutral situation, while BOLD signals of their brain were acquired with a 1.5 T scanner. A young Chinese male was admitted for a generalised tonic-clonic seizure preceded by a week-long history of fever. Subsequently, he developed continuous myoclonic jerks in all four limbs, with clear left sided predominance, and no accompanying clouding of consciousness. Contrast MRI of the brain demonstrated a venous angioma in the right cingulate gyrus. Over the next few days, the clinical picture evolved, with focal motor status involving primarily the left lower limb and the abdomen. These movements resolved with anticonvulsant therapy. This case illustrates generalised myoclonus arising fr

A middle-aged man suffering from acute carbon monoxide intoxication was clinically assessed to be in an akinetic and mute state. In order to elucidate regional cerebral disturbances, brain metabolism was investigated with fluoro-deoxyglucose positron emission tomography ((18)FDG-PET) 5.5 months after intoxication. Significantly reduced metabolic rates of glucose were revealed in selected brain regions, especially in both the frontal and anterior cingulate cortices, as well as in the subcortical white matter. Frontal and cingulate cortices showed a preserved metabolism of 35-53%, whereas the re

Previous research has demonstrated that patients with schizophrenia have an impaired ability to monitor erroneous responses to stimuli internally. Event-related potential (ERP) studies of error-eliciting tasks indicate that, in healthy adults, the commission of an erroneous response is associated with a fronto-centrally distributed negative voltage component termed the error negativity (Ne) or error-related negativity (ERN). In patients with schizophrenia, the Ne/ERN elicited by errors of commission (EoC) is reduced in amplitude compared with that elicited in healthy participants. Functional M

The objective of the present study was to provide a pathophysiological model of the development of schizophrenia. The method used was a selective review of recent findings, including those in our own department, concerning the clinical symptoms, cognitive characteristics and morphological brain changes in schizophrenia. A four-syndrome classification was proposed in which 'alienation syndrome' is separated from delusion syndrome. Memory organization deficit in schizophrenia patients was correlated with reduced activation of the left inferior frontal regions. Magnetic resonance imaging and stat

The various structural components of the cortical degeneration of Alzheimer's disease were defined and graded. The severity of the degenerative process was thus mapped in different cortical areas where neuronal numbers and cortical width were also measured and compared with controls. Contrary to the general opinion that the degenerative process is rather uniformly diffuse, though accentuated in the medial temporal and frontal cortex, we found a consistent and more elaborate pattern with clearcut regional differences. Thus the degeneration involved, besides basal medial temporal limbic areas, t

1. All lesions resulted in degeneration of the short intracortical association fibers in cortical layer I and of the short subcortical fibers which extended to the corona radiata before ending in the deeper layers of the overlying neopallium. 2. From all the lesions fibers were traced through the corona radiata to the subcallosal or the so-called superior fronto-occipital association bundle. This bundle had projection fibers to the orbitofrontal cortex. 3. From the lesion in the orbitofrontal neopallium, the orbitofrontal-pyriform connections were established. Such fibers coursed on the dorsal

CD33 is a susceptibility locus for late-onset Alzheimer's disease (AD). However, how the neural mechanism of CD33 affects cognition in the AD spectrum population remains unclear. We aimed to investigate the primary and interactive effects of the CD33 (rs3865444) genotype on brain function in patients with AD using global functional connectivity density (gFCD) mapping via resting-state functional magnetic resonance imaging. Furthermore, we used a conditional process analysis to identify the relationship among the CD33 genotype, gFCD, and cognition performance across the AD spectrum population.

The nocebo effect, opposite of the better-known placebo effect, in which anticipation of harm worsens pain and other symptoms, is increasingly thought to be responsible for poor clinical outcomes. In humans, nocebo hyperalgesia (i.e., increased pain sensitivity) is blocked by proglumide, a cholecystokinin (CCK) receptor antagonist. Yet, the neural circuitry underlying nocebo hyperalgesia remains unidentified, largely due to a lack of appropriate animal models. Here, we developed distinct mouse models of CCK-dependent nocebo hyperalgesia in which the expectation of pain was elicited by contextu

Physical neglect is associated with poorer cognitive functioning in patients with schizophrenia, including deficits in facial emotion recognition. Recent research by our group showed the relationship between physical neglect and facial emotion recognition is mediated by inflammation. While this mediation effect was observed at the level of behaviour, examining the relationship between physical neglect, inflammation and neural activation during facial emotion recognition would help confirm brain regions impacted and support behavioural findings, but these relationships are unclear. OBJECTIVE: Transcranial magnetic stimulation (TMS) of the dorsolateral prefrontal cortex (DLPFC) may achieve its antidepressant effect via transsynaptic activation of the subgenual cingulate (SGC). SGC activation by DLPFC TMS can be quantified by concurrent EEG (TMS-EEG) via source localization, while the connection between the DLPFC and SGC can be quantified by resting state functional connectivity (RSFC). Here, the relationship between SGC source activation and DLPFC-SGC anticorrelation is characterized in depression patients.
METHODS: TMS-EEG and fMRI were collected from 42 depression pati

Adverse Childhood Experiences (ACE), including abuse and neglect, can have lasting negative effects on health, decreasing lifespan and increasing the risk of chronic diseases. While research on ACE's impact on brain biochemistry is limited, Magnetic Resonance Spectroscopy (MRS) provides a non-invasive way to study these alterations. This study aims to identify neurochemical patterns linked to ACE exposure using J-edited MRS methods. 43 female participants (18 Low-ACE and 25 High-ACE), aged 19 to 31, were recruited. ACE exposure was assessed using the Maltreatment and Abuse Chronology of Exposu

BACKGROUND: Volumetric changes in the superior temporal gyrus and anterior cingulate cortex have been repeatedly reported in studies on schizophrenia. Tractography and functional magnetic resonance imaging studies have suggested that alterations in connectivity involving the superior temporal gyrus and the anterior cingulate cortex are relevant to psychotic symptoms of schizophrenia.
METHODS: We analyzed nanometer-scale three-dimensional structures of brain tissues of the superior temporal gyrus and the anterior cingulate cortex in eight schizophrenia and eight control cases and evaluated str

BACKGROUND: Adverse childhood experiences (ACEs) represent a strong influence on the developing brain and profoundly affect corticolimbic circuits, thereby contributing to vulnerability for mental disorders. Individual differences in resilience-related behavior, such as physical activity, may mitigate these effects.
METHODS: This retrospective study examined whether self-reported lifetime physical activity (LPA) modulates the relationship between ACEs and resting-state functional connectivity (rs-FC) of key limbic regions among 75 adults (mean age = 31.8 years, 82.7% female). Interaction mode

Objective. This feasibility study provides a technical validation of a functional magnetic resonance imaging-based (fMRI)-neurofeedback system for personalised neuromodulation of reward neurocircuitry in a larger study of cannabis use disorder. We adapted an fMRI-neurofeedback system in 7 Tesla scanner to neuromodulate the top 33% voxels of interest (VOIs) within the anterior cingulate cortex (ACC). We investigated characteristics of the neurofeedback signal and its specificity to VOIs compared to control regions (cerebrospinal-fluid [CSF], precentral gyrus, superior frontal gyrus, and middle t

AIMS: The misfolding and accumulation of the protein α-synuclein (αSyn) into cytoplasmic inclusions termed Lewy bodies (LBs) and Lewy neurites is the defining neuropathological feature of LB diseases, such as Parkinson's disease (PD) and dementia with Lewy bodies (DLB). The loss of neurons and/or synapses has been postulated to underlie the clinical syndrome of DLB. The present study sought to elucidate the relationship between LB burden and neuronal and synaptic loss in DLB.
METHODS: Post-mortem brain tissue from the cingulate gyrus and inferior temporal gyrus, two regions vulnerable to LB p

Recent evidence indicates that the intraparietal sulcus (IPS) may play a causal role in action stopping, potentially representing a novel neuromodulation target for inhibitory control dysfunctions. Here, we leverage intracranial recordings in human subjects to establish the timing and directionality of information flow between IPS and prefrontal and cingulate regions during action stopping. Prior to successful inhibition, information flows primarily from the inferior frontal gyrus (IFG), a critical inhibitory control node, to IPS. In contrast, during stopping errors the communication between I

BACKGROUND: The phantom sound of tinnitus can be an extremely debilitating condition. The thalamocortical dysrhythmia (TCD) hypothesis is a feature of subjective tinnitus; however, its consistency in characterizing different types of tinnitus remains unclear.
METHOD: We compared theta-beta/gamma coupling in multichannel EEG recordings from subjects with bothersome tinnitus (BT), non-bothersome tinnitus (NBT), and healthy controls (HC). Additionally, we used these EEG features to distinguish BT from NBT by employing the k-nearest neighbor (KNN) model.
RESULTS: Theta-beta/gamma phase-amplitude
