Forest tree-pathogen interactions under climate change.

PURPOSE: Complete high ulnar nerve injury can cause serious sequelae, including residual sensation and loss of movement and especially dysfunction of the intrinsic muscles of the hand. As a solution to treat complete high ulnar nerve injury, we proposed a new repair method for ulnar nerve injury based on nerve-magnified regeneration. METHODS: Twenty-two patients with complete division of the ulnar nerve at a high level who were treated from May 2013 to December 2016 were divided into two groups. The proposed repair method for complete high ulnar nerve injury was performed in group I (11 patie

Spatiotemporal dynamics of Brucella immune evasion across infection stages.

Brucella, a major zoonotic pathogen, poses a significant threat to global public health and causes substantial economic losses in the livestock industry. It employs diverse and sophisticated immune evasion strategies to circumvent host surveillance, establishing and maintaining chronic infections that are difficult to treat and prone to relapse. While previous reviews have catalogued individual virulence factors-such as the VirB type IV secretion system-and their actions on pathways like TLR4 signaling, most analyses focus on isolated stages or mechanisms, overlooking the integrated, dynamic r

Influence of fungi on epithelial homeostasis and role in inflammatory diseases.

SUMMARYThe skin harbors a diverse fungal community that contributes to both epidermal homeostasis and inflammatory disease. Historically, studies of cutaneous fungi focused primarily on opportunistic infections in immunocompromised hosts. Advances in sequencing technologies and metagenomic analyses have revealed that commensal yeasts of the skin microbiome likely influence host physiology and cutaneous disease severity. In this review, we summarize the current knowledge of host-fungal interactions at the skin epithelium, with particular emphasis on the yeast genera Malassezia and Candida. We d

The Airway Microbiome as a Modulator of Influenza Virus Infection: Mechanistic Insights and Translational Perspectives-Review.

Outcomes of influenza virus infection vary widely across individuals, reflecting not only viral genetics and host factors but also the composition and function of the airway microbiome. Over the past few years, mechanistic work has clarified how specific commensals (for example, Staphylococcus epidermidis and Streptococcus oralis) restrict influenza replication by priming epithelial interferon-λ programs, reshaping intracellular metabolite pools (notably polyamines), dampening host protease activity, and maintaining barrier integrity; meanwhile, pathobionts (notably Staphylococcus aureus and S

Integrin ß4 is a receptor for emerging fungal pathogens from the genera Lomentospora and Scedosporium.

Lomentospora prolificans is an environmental fungus that can cause life-threatening infections when airborne conidia are inhaled. Airway epithelial cells are likely to be the first host cells to interact with L. prolificans during pulmonary infection; however, the fungal and host factors that govern this interaction are completely unknown. Herein, we combined whole fungal cell pulldowns of surface proteins from airway epithelial cells and liquid chromatography-mass spectrometry (LC-MS) to identify host proteins that could potentially serve as host receptors for the fungus. We provide evidence

Tissue-Resident Macrophages: Linking Physiology and Inflammation in Infection.

Macrophages are found in every tissue in the body, and their importance goes far beyond clearing pathogens. Developmental studies have shown that many macrophage populations originate during embryogenesis, persist through self-renewal, and adopt transcriptional and functional programs defined by their tissue environment. These tissue-resident macrophages (TRMs) are embedded within organ physiology, where they regulate, for instance, cardiac conduction, sculpt synaptic circuits, recycle iron, maintain epithelial barriers, and coordinate repair. During infection, TRMs connect to neural, metaboli

Innate Immune and Inflammatory Responses to Dengue Virus Infection.

Dengue is the most common arboviral infection in the world, causing up to 400 million cases per year. Although most cases are asymptomatic, the virus can cause a wide range of symptoms varying from high fever and pain, common to several arbovirus infections, to hemorrhagic fever and shock syndrome, which can often be fatal. Despite the association of some genotypes with disease severity, most symptom varieties can be traced to the interaction of the virus with the immune system. As early as viral entry, Dengue virus co-evolved with humans to evade the innate immune system, especially the antiv

Immune interactions of entomopathogens with mosquito and tick vectors.

Entomopathogenic fungi and bacteria offer promising alternatives to chemical insecticides for controlling mosquitoes and ticks-key vectors of malaria and Lyme disease. However, their efficacy largely depends on immune interactions between pathogens and hosts. Here, we focus on mosquitoes and ticks as representative disease vectors and dissect conserved and divergent immune responses, from physical barriers to cellular defenses and key humoral pathways, with mechanistic insights informed by insect model systems. Furthermore, we elucidate diverse virulence and immune evasion strategies that enab

mSphere of Influence: How a gut microbiota study reshaped my thinking on tick-borne pathogens.

Xin-Ru Wang works in tick biology and intracellular bacterial pathogenesis. In this mSphere of Influence article, she reflects on how "Gut microbiota of the tick vector Ixodes scapularis modulate colonization of the Lyme disease spirochete" by Narasimhan et al. (2014) reshaped her understanding of vector competence by placing pathogen colonization within the ecological context of the tick microbiome. Her laboratory studies Rickettsia-tick cell interactions, including autophagy, apoptosis, and innate immune signaling. Here, she examines how this ecological framework extends to the intracellular

mGem: Subcellular compartments in bacterial pathogens and their role during infection.

Bacterial subcellular compartments and organelles allow for the spatial organization and sequestration of metabolic pathways, toxic intermediates, and key nutrients. The prevalence of these compartments across bacterial phyla, including in prominent human pathogens, has become increasingly defined in the literature. However, the functions of subcellular compartments in bacterial pathogens and their contributions to virulence remain understudied, leaving many questions unanswered. In this review, we summarize current findings regarding the prevalence of proteinaceous and membrane-bound compartm

Curiouser and curiouser: understanding the intersection of inter- and intra-specific diversity during the Borrelia burgdorferi enzootic cycle and Lyme disease.

A consequence of the Borrelia (Borreliella) burgdorferi lifecycle is that a "standard" host-pathogen interaction involving the spirochete is challenging to define. The bacteria continuously cycle between vertebrate and invertebrate hosts, each inflicting substantially different selective pressures on the spirochete. Focusing solely on vertebrate infections, the spirochete can be found in numerous hosts in nature, including deermice, shrews, birds, and lizards. The immune pressures that the bacteria face in each of these hosts are variable and are thought to drive broad intraspecific variation

A co-evolutionary perspective on humans and Mycobacterium tuberculosis in the era of systems biology.

Tuberculosis is once again the most fatal global infectious disease and has killed many more humans than any other pathogen. Despite the identification of Mycobacterium tuberculosis (Mtb) over 140 years ago, we have yet to control the epidemic. A central issue is the complexity of the host-pathogen interaction, with multiple underlying pathways leading to tuberculosis disease. This intricate relationship stems from the prolonged co-evolution of the pathogen with humans, resulting in diverse immunological processes leading to tuberculosis disease. Conversely, Mtb exposure may give a survival ad

Research progress in PANoptosis mechanisms and their interplay with tuberculosis.

PANoptosis is a recently identified, inflammatory programmed cell death pathway that amalgamates features of pyroptosis, apoptosis, and necroptosis, executed via multiprotein complexes called PANoptosomes. Increasing evidence highlights PANoptosis has emerged as a potentially pivotal factor in the pathogenesis of several diseases, including viral and bacterial infections and malignancies. Given its extensive pathophysiological relevance, this review systematically explores the conceptual underpinnings, molecular mechanisms, and structural composition of PANoptosome. Special attention is devote

[Research progress on the Mycobacterium tuberculosis infection of mesenchymal stem cells].

Following Mycobacterium tuberculosis (Mtb) infection, the pathogen can establish a metabolically quiescent or replication-arrested dormant state within the host, leading to asymptomatic latent tuberculosis infection (LTBI). Under specific conditions, these dormant bacilli may reactivate and replicate, resulting in symptomatic clinical infection and constituting a major reservoir for active tuberculosis (TB). Dormant Mtb exhibits reduced susceptibility to conventional anti-TB drugs, and current TB vaccines fail to prevent the establishment or reactivation of dormant Mtb, rendering latent infect

Machine Learning-Based, Label-Free Motion Tracking of Leptospira spp. on Cultured Cells.

Leptospira spp. swim in liquid and crawl on surfaces with two periplasmic flagella. The Leptospira motility is a crucial virulence factor, especially crawling motility, which is possibly associated with the host-dependent severity of leptospirosis. In vitro infection assay using monolayers of cultured host cells provides basic knowledge on host-pathogen interaction in leptospirosis; however, it is difficult to observe leptospires on the host cells due to their similarity in refractive index. Though fluorescent labeling could solve the problem in general, it could negatively affect the leptospi

Diet quality improves survival and mediates larval response to densovirus infection in the Melissa blue butterfly.

Entomopathogenic viruses affect insect populations by reducing fitness and influencing other measures of performance such as development and longevity. Given the huge diversity of viruses, studies are needed to understand the mechanisms underlying how viruses affect their insect hosts. The Junonia coenia densovirus (JcDV) infects a wide range of Lepidoptera and can cause death in the larval and pupal stages. In this study, we used the Melissa blue butterfly (Lycaeides melissa: Lycaenidae) to describe infection progression at the individual level, the effects of infection on development, surviv

Infectious agents in dilated cardiomyopathy: Genetic interactions, autoimmunity, mechanisms, and therapeutic approaches.

Dilated cardiomyopathy (DCM) is a heterogeneous myocardial disorder characterized by left ventricular dilation and systolic dysfunction in the absence of ischemic, hypertensive, or valvular heart disease. Although its precise etiology remains unclear, it is widely recognized as a multifactorial disease arising from complex interactions between genetic predisposition and environmental triggers. Among these, infectious agents have been implicated in the pathogenesis of various subtypes, particularly inflammatory and idiopathic DCM. These agents can contribute to disease onset and progression thr
