High effective specific activity N-succinimidyl 4-[18F](fluoromethyl)benzoate was prepared using a reversed phase HPLC procedure. Reversed phase HPLC removed several additional impurities not removed by normal phase HPLC, thereby increasing the effective specific activity. Small amount (< 100 micrograms) of sensitive proteins such as erythropoietin can be labeled with this reagent in high yield without aggregation.

A reverse-phase high-performance liquid chromatographic (HPLC) method for the measurement of lamotrigine (LTG) simultaneously with phenobarbitone (PB), phenytoin (PHT), and carbamazepine (CBZ) is described using hexobarbital as the internal standard. Three fluoroketonucleosides (6, 8, and 11) have been synthesized by direct oxidation of the fluoro precursors. The presence of the highly electronegative fluorine atom in the alpha position to the carbonyl group favours hydration leading to the gem-diol form so that the beta-elimination process to afford 6 and 8 was made difficult and failed in the case of the difluoro compound 11. When whole cells (stationary phase) of Yersinia pestis strain EV76 were radiolabelled with Iodogen and 131I, 16 major and 10 minor surface-exposed outer membrane proteins (OMPs) were identified. Labelling with N-hydroxysuccinimidyl 6-biotinylamino-hexanoate (biotin X-NHS) resulted in a complex protein profile detectable after blotting and developing with peroxidase-conjugated avidin. Y. pestis cell fractionation revealed that biotin X-NHS labelled not only OMPs but also proteins of inner cell compartments. Therefore, radiolabelling was the more reliable technique for identifying the OMPs of Y.

A highly sensitive method for the determination of tetracycline antibiotics (TCs) using reversed-phase high-performance liquid chromatography with fluorescence detection is presented. A rapid procedure for the extraction of conjugated bile acids from human fluids using pre-packed octadecyl-bonded silica cartridges is described. The method was compared with the other procedures reported in the literature and was found to produce a higher degree of sample purification and to achieve satisfactory accuracy and reproducibility. The present procedure is applicable to the high-performance liquid chromatographic assay of bile acid conjugates in human bile, gastric juice, serum and urine.

We investigated the structure of glycoconjugates contained within the secretory end-pieces and ductal segments in the rabbit submandibular and sublingual glands. Monomer conjugation of an enzyme followed by copolymerization with free monomer is a useful method of enzyme immobilization. L-asparaginase was conjugated with N-succinimidyl acrylate. An ethanolic solution of perazine (Taxilan), degraded by photooxidation, was examined by HPLC and mass-spectroscopy. Identical degradation and oxidation products were found in vitro as reported to exist in vivo with the exception of perazine-N-oxide and 3-OH-perazine. The formation of the latter two metabolites seems to be bound to the cytochrome P-450 system. A direct, oxygen dependent formation of some metabolites is, however, suggested. Side-effects of phenothiazines such as contact allergies might be related to the formation of radicals or particular oxidation products.

We have synthesized, by the solid phase procedure, then purified and chemically characterized two peptides. They mimic two regions of toxin II of the scorpion Androctonus australis Hector, one around disulfide bridge 12-63 and another at sequence 50-59. Each of these two regions was supposed to include an antigenic site.
