Occurrence of ovulation after intracerebroventricular infusion of substance P in 6-OHDA pretreated female rats.

Substance P (SP) infused into the third cerebral ventricle blocks spontaneous ovulation in female rats, probably through catecholaminergic neurons. The studies presented in this paper were undertaken to investigate whether SP exerts its suppressing effect on ovulation in 6-hydroxydopamine (6-OHDA) pretreated female rats. After 8-12 days following 6-OHDA pretreatment female rats were infused, on the day of proestrus, with vehicle and all animals were found to ovulate or with a solution containing 5 nmol of SP and 89% of female rats were then found to ovulate. In the group pretreated with vehicl

Naltrindole retards tolerance development to morphine-induced effects on EEG and EEG power spectra.

In the present study, EEG and EEG power spectra were used to assess the effects of naltrindole, a selective delta opioid antagonist, on the development of tolerance to morphine. Adult female Sprague-Dawley rats were implanted with cortical EEG electrodes and permanent indwelling i.c.v. and i.v. cannulas. Twice daily for 7 days, rats were pretreated with either i.c.v. naltrindole (20 nmol) or i.c.v. water, 20 min before i.v. morphine (10 mg/kg) injections. The treatments produced EEG slow-wave bursts and associated behavioral stupor. The amount and duration of these effects decreased less rapid

[Effects of thyrotropin-releasing hormone (TRH) on status epilepticus in rats].

Recent studies have demonstrated that intramuscular administration of thyrotropin-releasing hormone (TRH) or its analogue improves various clinical aspects of intractable epilepsy such as Lennox-Gastaut syndrome, West syndrome, and myoclonus epilepsy. Other clinical studies reported efficient property of intravenous TRH against status epilepticus. However, it is also true that intravenous TRH produces epileptic seizures in patients with epilepsy or organic brain damage. Thus, the utility of intravenous TRH for the treatment of status epilepticus seems to be equivocal. To further explore the pr

[The effect of the intracerebroventricular administration of vasopressin on the arterial pressure and heart rate of Brattleboro and Long-Evans rats].

Vasopressin (VP) was administered in the lateral cerebral ventricles of homozygous rats with congenital diabetes insipidus of Brattleboro (BB) strain as well as of normal animals of Long Evans strain (LE). The following parameters were studied: the latent period for achievement of maximal pressor response in seconds (LP), absolute value of the increase in arterial pressure in comparison with the initial V mmHg (delta AP) and cardiac frequency (CF). The initial values of AP in BB were lower than those values found in LE (BB-9, 10 +/- 0.57 kPa; LE-11, 31 +/- 0.53 kPa; p less than 0.01, while CF

Intra-cerebral ventricular infusion of 5-iodo-2-deoxyuridine (IUDR) as a radiosensitizer in the treatment of a rat glioma.

The efficacy of 5-iodo-2-deoxyuridine (IUDR) as a radiosensitizer when administered by continuous infusion into the cerebral spinal fluid (CSF) of the lateral cerebral ventricle was evaluated in a 9L gliosarcoma rat brain tumor model. Stereotactic implantation of a 5 x 10(4) tumor cell suspension into the left caudate nucleus was carried out in four groups of 10 rats each. Control animals had a median survival of 16.9 days (range 16-21 days). IUDR, 8.4 mg over 7 days administered by continuous infusion into the left lateral ventricle produced a slight survival advantage (median survival 21.5 d

Analysis of the action of angiotensin II on the baroreflex control of heart rate in conscious rabbits.

There is considerable evidence that angiotensin II (Ang II) attenuates the baroreflex control of heart rate (HR), but the mechanism and site of this action have not been precisely defined. In the present study the effects of systemically and centrally administered Ang II on the baroreflex control of HR were investigated in conscious, chronically prepared rabbits. Baroreflex curves (HR vs. mean arterial pressure) were generated with iv infusions of phenylephrine or nitroprusside. Background infusion of Ang II at 10 ng/kg.min increased mean arterial pressure from 77.3 +/- 3.0 to 94.3 +/- 4.1 mm

Ingestion, body weight and activity of rats receiving repeated intracerebroventricular infusions of rat satietin.

Satietin is a putative satiety agent that is found in a variety of species including man and the rat. In the first experiment male Sprague-Dawley rats were fitted with chronic third ventricle cannulas and placed in activity wheels. After recovery, animals were intracerebroventricularly (ICV) infused with either sterile rat satietin (r-SAT) (100 micrograms/rat) dissolved in 10 microliters saline (n = 6) or sterile saline (n = 8). Infusions were repeated the next two days. Infusions of r-SAT had no effect on the rats' water intake or activity but did suppress (p less than 0.05) their food consum

Suppression of the pressor effect of centrally administered thyrotropin-releasing hormone under halothane, pentobarbital and flunitrazepam anaesthesia.

Intracerebroventricular (i.c.v.) administration of thyrotropin-releasing hormone (TRH) caused an increase in blood pressure (BP) and heart rate (HR) in conscious rats. The pressor effect was greatly diminished by adrenalectomy as well as after pretreatment with phentolamine, an alpha-receptor antagonist or with mecamylamine, a ganglion blocker, suggesting that centrally administered TRH increases BP mainly by stimulating sympathetic activity. Under halothane (0.8%), pentobarbital (33 mg/kg, i.p.) and flunitrazepam (0.8 mg/kg, i.v.) anaesthesia, the pressor effect of TRH was almost completely b

6-Hydroxydopamine-induced aggression in cats: effects of various drugs.

The effects of intracerebroventricular injections (ICV) in the unanesthetized cat of antimuscarinic drugs, ganglionic blocking agents, alpha and beta adrenergic blocking substances, dopamine and 5-hydroxytryptamine (5-HT) antagonists, and an antihistamine on aggressive behavior produced by 6-hydroxydopamine injected similarly was investigated. It was found that atropine, hyoscine, hexamethonium, mecamylamine, yohimbine, phenoxybenzamine, propranolol, practolol, chlorpromazine, haloperidol, antazoline and methysergide exerted virtually no effect on the pattern of aggressive responses evoked by

Intracisternal bombesin induced inhibition of gastric secretion is not mediated through prostaglandin or opioid pathways.

Neuromedin B, a mammalian decapeptide which shares homology with the carboxyl terminal fragment of ranatensin R and bombesin, injected intracisternally (880 pmol) did not modify gastric acid secretion stimulated either by pylorus ligation in conscious rats or by pentagastrin infusion in urethane-anesthetized rats. Intracisternal bombesin (300 pmol) further enhanced PGE2-induced inhibition of gastric acid secretion and suppressed prostaglandin-induced stimulation of gastric secretory volume. Gastric response to intracisternal bombesin was not modified by indomethacin (10 mg/kg) or naloxone pret

Altered body temperature of rabbits after central injection of beta-endorphin and other peptides.

After injection of 5 micrograms into a lateral cerebral ventricle, 6 of 12 peptides induced mean changes of 0.3 degree C or more in rectal temperature of rabbits. When beta-endorphin was studied further with 1.25-5 micrograms in 10, 23 and 30 degrees C environments, it induced dose-related hypothermia in the cold, hyperthermia in the heat and progressively smaller increases in body temperature with increasing doses at 23 degrees C.

Evaluation of neurotensin's thermolytic action by ICV infusion with receptor antagonists and a Ca++ chelator.

To investigate the mechanisms responsible for the thermolytic action of neurotensin (NT), cannulae for intracerebroventricular (ICV) injection were implanted stereotaxically in Sprague-Dawley rats. Postoperatively, body temperature in the unrestrained rat was monitored continuously by a colonic thermistor probe. NT in a dose of 1.5-4.5 micrograms or CSF carrier vehicle was infused bilaterally in the cerebral ventricle (ICV) in a volume of 7.5 microliters. With the rats kept at an ambient temperature of 22 degrees C, NT given ICV produced a dose-dependent fall in core temperature of greater tha

The effect of intracerebroventricular administration of catecholamines and their antagonists on rectal temperature of Mastomys natalensis.

Noradrenaline (NA), adrenaline (ADR), isoprenaline (ISO) and dopamine (DA) were given through a chronically implanted cannula in the lateral cerebral ventricle of Mastomys natalensis. Low doses of NA (0.05-0.25 microgram) reduced rectal temperature while larger doses (0.35 microgram upwards) produced dose-dependent hyperthermia. The hypothermic effect was antagonised by alpha-adrenoceptor and the hyperthermia by beta-adrenoceptor antagonists. alpha-Methyl noradrenaline produced less hyperthermia but it antagonised the hyperthermic effect of NA. Adrenaline (0.1-10 microgram) was ineffective per

[Effect of cathionine (alpha-aminopropiophenone) on rat EEG activity].

We have studied the effect induced by the injection of Cathinone (alpha-aminopropiophenone) into the cerebral ventricle. This substance has recently been isolated from the leaves of the Catha Edulis. The chemical structure of Cathinone is similar to that of amphetamine. However, Cathinone does not modify cerebral electric activity in sense of a typical activation seen with amphetamine. Cathinone has evoked, at high doses, seizures comparable to epileptic activity.

Increased pressor responsiveness to enkephalin in spontaneously hypertensive rats: the role of vasopressin.

1. The cardiovascular effects of an enkephalin analogue were examined in spontaneously hypertensive and normotensive Wistar-Kyoto rats. (D-Ala2)-methionine enkephalin caused a biphasic increase in blood pressure and an increase in heart rate after intracerebroventricular injection. 2. The initial pressor response to (D-Ala2)-methionine enkephalin was greater in hypertensive than in normotensive rats. No difference was noted between groups during the secondary pressor response. Heart rate increases paralleled the secondary increase in blood pressure. 3. Naloxone pretreatment abolished the secon

Effect of angiotensin II and glucocorticoids on plasma angiotensinogen concentration in the dog.

The increase in plasma angiotensinogen concentration produced by angiotensin II infusion in the dog is accompanied by an increase in plasma corticosteroid concentration and is prevented by adrenalectomy. The present study was undertaken to test the possibility that the angiotensinogen response to angiotensin is mediated via an increase in glucocorticoid secretion. Infusion of angiotensin II (3-6 mug/h for 5 h) into the third cerebral ventricle of five dogs increased plasma corticosteroid concentration from 3.5+/-0.6 to 11.8+/-1.5 mug/dl (P less than 0.025) but did not increase plasma corticost

Interaction between the effects of spinal heating and cooling and of injections into a lateral cerebral ventricle of noradrenaline, 5-hydroxytryptamine and carbachol on thermoregulation in sheep.

1. A study has been made of the interactions of the thermoregulatory effects of spinal cord heating and cooling and of the injections into the cerebral ventricle of noradrenaline, 5-hydroxytryptamine (5-HT) and carbamylcholine in sheep. 2. The interactions of spinal cord heating and the injections into the cerebral ventricle of noradrenaline, 5-HT and carbamylcholine were very similar to those of hypothalamic heating or of high ambient temperature and the injections into the cerebral ventricle of these substances. These results are interpreted as evidence of the synaptic convergence of the pat

Effect of cerebral intraventricular insulin on pancreatic insulin secretion in the dog.

The effect of cerebral intraventricular insulin on pancreatic insulin secretion was investigated. An extracorporeal pancreatic blood circuit was established after laparotomy to monitor blood flow and insulin concentration directly from the superior pancreaticoduodenal vein. Phentolamine was infused throughout (0.2 mg./min. intravenously) to block alpha-adrenergic effects of any catecholamine secretion induced by surgical stress. Glucose (1.5 mg./kg./min. intravenously) was infused to maintain a constant baseline stimulation of insulin secretion. Six dogs received insulin and six control dogs r
