Apolipophorin-III (ApoLp-III), a multifunctional protein with lipid transport and immune defense functions, widely exists in insects. Although the function of ApoLp-III as a pattern recognition receptor (PRR) in immunity has been relatively studied, the immune response mediated by ApoLp-III is still vague. To understand whether ApoLp-III is involved in the activation of the prophenoloxidase-activating system (PPO-AS), we examined the production of nitric oxide (NO), and the synthesis of antimicrobial peptides after immune recognition. The larvae of lepidopteran pest Ostrinia furnacalis were us

Systemic autoinflammatory diseases (SAIDs) are associated with a dysregulation of innate immunity leading to recurrent or chronic inflammation. There are both well-characterized monogenic forms, such as familial Mediterranean fever, cryopyrinopathies and VEXAS syndrome, and multifactorial forms defined by classification criteria, such as Still's disease, Schnitzler syndrome and SITRAME. However, a significant proportion of patients, estimated at up to 73% in some series, present with an autoinflammatory phenotype without any identified pathogenic variant or established diagnosis based on class

Mitochondria, known as the powerhouse of cells, play a crucial role in host innate immunity during flavivirus infections such as Dengue, Zika, West Nile, and Japanese Encephalitis Virus. Mitochondrial antiviral signaling protein (MAVS) resides on the outer mitochondrial membrane which is triggered by viral RNA recognition by RIG-I-like receptors (RLRs). This activation induces IRF3 and NF-κB signaling, resulting in type I interferon (IFN) production and antiviral responses. Upon flavivirus infection, mitochondrial stress and dysfunction may lead to the release of mitochondrial DNA (mtDNA) into

Malignant pleural effusion (MPE) is a common complication of lung adenocarcinoma (LADC) which is associated with a dismal prognosis. We investigated the prognostic role of PD-L1 and other immunomodulators expression in the immune compartment of MPE immune composition. MPE cytologic cell blocks of 83 LADC patients were analysed for the mRNA expression of 770 cancer-immune genes by the NanoString nCounter platform. The expression of relevant immune cell lineage markers was validated by immunohistochemistry (IHC) using quantitative pathology. The mRNA immune profiling identified four MPE patient

Asthma is a common inflammatory disease involving cross-talk between innate and adaptive immunity. We reveal that antibacterial innate immunity protein, peptidoglycan recognition protein (Pglyrp)1, is involved in the development of allergic asthma. Pglyrp1(-/-) mice developed less severe asthma than wild-type (WT) mice following sensitization with house dust mite (allergen) (HDM). HDM-sensitized Pglyrp1(-/-) mice, compared with WT mice, had diminished bronchial hyperresponsiveness (lung airway resistance); numbers of eosinophils, neutrophils, lymphocytes, and macrophages in bronchoalveolar lav

Pig farmers are exposed to organic material in pig barns on a daily basis and have signs of an ongoing chronic airway inflammation and increased prevalence of chronic inflammatory airway diseases, predominantly chronic bronchitis. Interestingly, the inflammatory response to acute exposure to organic dust is attenuated in farmers. The aim of the study was to closer characterize innate immunity features in blood and airways in farmers and in naïve, non-exposed, controls. The expression of pattern recognition receptors (TLR2, TLR4 and CD14) whose ligands are abundant in pig barn dust and adhesion

Mannan-binding lectin (MBL) is a C-type serum lectin, which is believed to play an important role in the innate immunity against a variety of pathogens. MBL can bind to sugar determinants of a wide variety of microorganisms, neutralize them and inhibit infection by complement activation through the lectin pathway and opsonization by collectin receptors. Given that small intestine is a predominant site of extrahepatic expression of MBL, here we addressed the question whether MBL is involved in mucosal innate immunity. The carbohydrate recognition domain (CRD) genes of mouse MBL-C (mMBL-C) were

Duck virus hepatitis caused by duck hepatitis A virus (DHAV) is an acute and contagious disease. To better understand the pathogenic mechanism of DHAV-3 in ducklings, an infection experiment was performed. Our results showed that typical symptoms were observed in the infected ducklings. DHAV-3 could infect many tissues, leading to pathological lesions, especially on the livers and spleen, and the host immune responses are activated in infection. Real-time quantitative PCR demonstrated that expression of many innate immune-related genes was mostly up-regulated in the livers and spleen, and anti

Antimicrobial-resistant infections present significant threats to public health, predicted to cause 10 million fatalities annually by 2050. Of these infections, one of the most frequent is methicillin-resistant Staphylococcus aureus (MRSA). During therapeutic exposure to last-line antibiotics, including daptomycin, MRSA often acquires genetic mutations conferring resistance. Resistance is typically mediated through structural alterations to the bacterial cell wall and membrane, which we have previously shown, concomitantly impede the activation of responding dendritic cells (DCs). Here, we dem

Susceptibility to infection after solid organ transplantation (SOT) reflects interactions among epidemiologic exposures, allograft-specific factors, pharmacologic immunosuppression, and host innate and adaptive immune functions. Substantial variability in susceptibility to viral, bacterial, and fungal infections occurs among recipients despite comparable immunosuppressive regimens. Accumulating evidence demonstrates that allelic variation, including single-nucleotide polymorphisms, occurs in genes involved in multiple aspects of innate immune function and influence infectious risk after SOT. The single domain von Willebrand factor type C (SVWC) appears in small secreted peptides that are arthropod-specific and are produced following environmental stress or pathogen exposure. Most research has focused on proteins with SVWC domain that are induced after virus infection and are hypothesized to function as "cytokines" to regulate the innate immune response. The expansion of SVWC genes in insect species indicates that many other functions remain to be discovered. Research in shrimp has elucidated the adaptability of Vago-like peptides in the innate immune response against bacteria, fun

INTRODUCTION: Sentinel cells of the airway epithelium are repetitively exposed to RNA virus infections. Here, the production of pathogen-associated molecular patterns (PAMPs) activates the innate immune response (IIR), inducing antiviral defenses, activating adaptive immunity and stimulating epithelial repair. The question whether repeated exposures produce innate "memory", where heightened inflammatory and injury/repair responses are produced upon subsequent exposure, is unknown.
METHODS: We establish a reproducible model of innate memory in tumor protein (TP63)+/keratin (KRT)5+ human small

The threat of new viral outbreaks has heightened the need for ready-to-use vaccines that are safe and effective. Here we show that a subcutaneous vaccine consisting of live Zika virus electrostatically entrapped in a self-adjuvanting hydrogel recruited immune cells at the injection site and provided mice with effective protection against a lethal viral challenge. The hydrogel prevented the escape of the viral particles and upregulated pattern recognition receptors that activated innate antiviral immunity. The local inflammatory niche facilitated the engulfment of the virus by immune cells infi

Pattern-triggered immunity (PTI) is a key plant defense mechanism initiated by the recognition of pathogen-associated molecular patterns (PAMPs) by cell-surface receptors, including leucine-rich repeat receptor proteins (LRR-RPs). Upon PAMP perception, LRR-RPs associate with SOBIR1 and recruit SERK coreceptors, such as BAK1, into the receptor complex to initiate PTI. The lipase-like protein EDS1 and PAD4 are central components of effector-triggered immunity (ETI). Small molecule (SM) phosphoribosyl-AMP/ADP, generated by TIR-domain NADase activity, promotes the recruitment of helper NLRs of the

The discovery of signal receptors of congenital immunity (signal PRR) not only provided a novel view of basic aspects of pathogenesis of chronic inflammatory diseases but also created a basis for the development of additional diagnostic criteria for these pathologies and new pharmaceuticals for their treatment. Reduced expression and function of PRR due to mutations/polymorphisms or epigenetic disturbances of regulation can be regarded as immunodeficient conditions manifest as severe infectious inflammatory diseases. In contrast, excessive expression and activation of PRR as a rule leads to ch

Tissue-resident-memory T cells (TRM) populate the body's barrier surfaces, functioning as frontline responders against reencountered pathogens. Understanding of the mechanisms by which CD8TRM achieve effective immune protection remains incomplete in a naturally recurring human disease. Using laser capture microdissection and transcriptional profiling, we investigate the impact of CD8TRM on the tissue microenvironment in skin biopsies sequentially obtained from a clinical cohort of diverse disease expression during herpes simplex virus 2 (HSV-2) reactivation. Epithelial cells neighboring CD8TRM

Cigarette smoking is the leading preventable cause of chronic diseases (e.g., COPD, cardiovascular disease, cancer), largely driven by persistent immune-inflammatory mechanisms. This review synthesizes the molecular and cellular cascades linking cigarette smoke (CS) exposure to chronic pathology. CS constituents, particularly ROS/RNS, induce rapid oxidative stress that overwhelms antioxidant defenses and generates damage-associated molecular patterns (DAMPs). These DAMPs activate pattern recognition receptors (PRRs) and the NLRP3 inflammasome, initiating NF-κB signaling and the release of pro-

Pseudomonas aeruginosa causes high morbidity and mortality in nosocomial infections, and newly approved antibiotics have been declining for decades. A green and universal deprotonation-driven strategy is used to screen the guanylic acid-metal ion coordination polymer nanoparticles (GMC), instead of the failure of binding occurs when specific metal ion participation. We find that the precise pH-dependent oxidase-like activity of GMC-2 orchestrates a duple symphony of immune modulation for Pseudomonas aeruginosa biofilm infections. Specifically, GMC-2-mediated reactive oxygen species (ROS) regul

The health and well-being of a host are deeply influenced by the interactions with its gut microbiota. Contrasted environmental conditions, such as diseases or dietary habits, play a pivotal role in modulating these interactions, impacting microbiota composition and functionality. Such conditions can also lead to transitions from beneficial to detrimental symbiosis, viewed as alternative stable states of the host-microbiota dialogue. This article introduces a novel mathematical model exploring host-microbiota interactions, integrating dynamics of the colonic epithelial crypt, microbial metabol

Metal-induced hypersensitivity is driven by T-cell sensitization to metal ions. Although numerous metals are associated with the development of diffuse parenchymal lung disease, beryllium-induced hypersensitivity is the best-studied to date. This review focuses on the interaction between innate and adaptive immunity that leads to the development of chronic beryllium disease. After beryllium exposure, activation of the innate immune system occurs through the engagement of pattern-recognition receptors. This activation leads to cell death, release of alarmins, and activation and migration of den
