Foetal exposure to maternal stressful events increases the risk of having asthma and atopic diseases in childhood.

BACKGROUND: The natural history of asthma and atopic diseases begins in utero. Studies investigating the influence of foetal exposure to maternal stressful life events during pregnancy (SLEP) on asthma and atopic diseases are lacking. AIM: To test whether the children of mothers who had experienced SLEP are at an increased risk for asthma, atopic eczema and allergic rhinitis. METHODS: The association between maternal SLEP (at least one among: divorce, mourning or loss of the job) and the occurrence of asthma and atopic diseases in childhood was studied in a population (n = 3854) of children,

A longitudinal study on the effects of maternal smoking and secondhand smoke exposure during pregnancy on neonatal neurobehavior.

Maternal smoking during pregnancy is one of the most modifiable causes of morbidity and mortality for both pregnant women and their fetuses. The long-term effects of prenatal exposure to smoke on child behavior and development have been the subject of more extensive research than have the short-term effects. Therefore, the aim of this work is to examine the effects of smoke exposure during pregnancy on neonatal behavior, including in our study a group of mothers exposed to secondhand smoke. The behavior of 282 healthy full-term newborns was assessed using the Neonatal Behavior Assessment Scale

Maternal exposures and risk of spontaneous abortion before and after a community oriented health education campaign.

BACKGROUND: In a district of Veneto (North-east Italy) where numerous females of childbearing age were occupationally exposed to organic solvents in nearly 400 shoe factories, a case-control study found significant associations between maternal exposures (from occupation and risky behavior) and spontaneous abortion (SAB). Thereafter, a health education campaign was undertaken to increase awareness of risk factors for pregnancy in the population. To evaluate the effects of this campaign maternal exposures and SAB risks were compared before and after the campaign. METHODS: Hospital records were…

The influence of maternal exposure to volatile organic compounds on the cytokine secretion profile of neonatal T cells.

Indoor VOC (volatile organic compound) exposure has been shown to be correlated with airway symptoms and allergic manifestations in children. An investigation was conducted within an ongoing birth cohort study (LISA: Lifestyle-Immune System-Allergy) of the association between maternal exposure to VOCs and immune status at birth, in particular the cytokine secretion profile of cord-blood T cells. In a randomly selected group of 85 neonates, cytokine-producing cord-blood T cells were analyzed using intracellular cytokine detection. VOC exposure was measured in children's dwellings by passive sam…

Effects of exposure to pups on maternal behaviour, sexual behaviour and serum prolactin concentrations in male rats.

Male rats showed maternal behaviour within 72 h after the onset of continuous exposure to newborn rat pups. The latency of the behavioural response could be reduced by daily treatment with the dopamine receptor antagonist domperidone (2 X 2.5 mg/rat), which increased serum prolactin concentrations (241.4 +/- 26.5 (S.E.M.) micrograms/l) above those of vehicle-treated males exposed to pups (25.3 +/- 11.7 micrograms/l). Male rats did not respond to exposure to pups by secreting prolactin; keeping endogenous prolactin concentrations at a minimum (2.8 +/- 0.1 micrograms/l) by daily treatment with t

Maternal Gestational and Lactational Exposure to Polystyrene Nanoplastics Triggers Vascular Injury and Lipid Deposition in Mouse Offspring.

As pervasive environmental contaminants, nanoplastics (NPs) have garnered increasing concern due to their potential health hazards. However, the impact of gestational and lactational exposure to NPs on offspring vascular structural injury remains unexplored. In this study, we used BALB/c mice to model gestational and lactational exposure to 100 nm polystyrene nanoplastics (PS-NPs) at doses of 0.1, 1, and 10 mg/L. Our findings revealed that PS-NPs exposure significantly reduced offspring body weight in both sexes. Histopathological analysis demonstrated that exposure induced multiorgan damage i

Maternal obesity and prenatal alcohol exposure are associated with child development: Results from the Safe Passage Study.

A large body of evidence supports the role of the prenatal environment in shaping childhood development. The relative contributions of prenatal alcohol use (PAE), maternal socioeconomic, and nutritional status on child development vary in high- versus low-income settings. We analyzed data from a prospective cohort study among mother-infant dyads from Cape Town (CT), South Africa and the Northern Plains (NP), USA. The Mullen Scales of Early Learning were administered by trained assessors to evaluate cognitive, motor, and language development of 1-year old children. We used multiple linear regre

Periconceptional and prenatal tobacco exposure and early externalizing behavior: Developmental pathways through maternal distress and language delay.

Periconceptional and prenatal tobacco exposure (PPTE) has been associated with early externalizing behaviors, yet the developmental pathways underlying this association remain poorly understood. The purpose of this study was to examine potential mechanisms through which PPTE may relate to child behavior. Data were drawn from a prospective, longitudinal cohort of mother-child dyads recruited at birth. The final sample included 203 mothers, predominantly White, with lower income, education, and partnership rates observed among heavier smoking groups. The study collected socio-demographic informa

Transcriptomic Reprogramming of the Human Placenta Following Maternal Opioid Exposure: Identification of Altered Metabolic and Translational Pathways.

BACKGROUND: Opioid use during pregnancy is linked to preterm birth, fetal growth restriction, and Neonatal Opioid Withdrawal Syndrome. While the placenta is the primary regulator of the uterine environment, the precise molecular mechanisms by which opioids dismantle placental function remain poorly defined. METHODS: The transcriptomic landscape of opioid-exposed human placentas from a unique Hawai'i-based cohort was characterized. RESULTS: Transcriptional signatures were defined by genes involved in translational inhibition and metabolic attenuation. Integrative network modeling predicted a

Maternal exposure to chronic, low-dose nonylphenol in zebrafish: Disruption of ovarian health, reproductive function, and embryogenesis.

Nonylphenol (NP), a non-ionic surfactant and potent endocrine disruptor, is known for its environmental persistence, biotic accumulation potential and toxicity. Nonetheless, mechanisms underlying NP modulation of female fertility with potential impact on embryogenesis in the unexposed offspring remain elusive. This study investigates the effects and toxic mechanisms of maternal exposure to NP at varying concentrations (50 and 100 μg/L) on zebrafish (Danio rerio), specifically focusing on ovarian health, reproductive parameters, and early developmental potential in the F1 generation. Our findin…

Dual impact of microplastic exposure in a mouse model: Impaired uterine receptivity and altered maternal-offspring metabolism.

With the increasing environmental presence of microplastics (MPs), concerns about their potential health risks have intensified. Our previous study detected MP deposition in the human uterus. However, its impact on uterine receptivity and the underlying mechanisms remains poorly understood, and the systemic metabolic consequences of MP exposure are yet to be elucidated. In this study, we investigated the reproductive and metabolic consequences of MP exposure using a mouse model. Female C57BL/6 mice were orally administered polystyrene microplastics (PS-MPs) at 50 mg/kg/day for 14 days (n = 30

Maternal exposure to urban particulate matter induces cardiac developmental toxicity in zebrafish offspring by disrupting mitochondrial homeostasis.

INTRODUCTION: Urban particulate matter (UPM) is a major air pollutant affecting public health, with maternal exposure potentially leading to cardiac developmental disorders in offspring. However, the exact mechanisms underlying the intergenerational effects of UPM remain unclear. OBJECTIVE: This study aimed to investigate the molecular mechanisms involved in cardiac developmental defects caused by maternal UPM exposure in offspring zebrafish. METHODS AND RESULTS: Female zebrafish were exposed to UPM for 21 days to examine intergenerational effects. The results indicated that maternal zebrafi…

Maternal psychological distress modifies the association between prenatal exposure to per- and polyfluoroalkyl substances and infants' neurodevelopment.

Prenatal PFAS exposure and maternal psychological distress may adversely affect child neurodevelopment through shared biological pathways, such as hypothalamic-pituitary-adrenal (HPA) axis disruption and pro-inflammatory responses. However, whether psychological distress modifies PFAS-related neurodevelopmental risks remains unclear. Using data from the Shanghai Birth Cohort including 1779 mother-child pairs, we measured maternal PFAS levels during the first trimester and assessed maternal psychological distress (perceived stress, depression, and anxiety) during the second trimester. Racial disparities in maternal exposure to ambient air pollution during pregnancy and prevalence of congenital heart defects.

Air pollution may be a potential cause of congenital heart defects (CHDs), but racial disparities in this association are unexplored. We conducted a statewide population-based cohort study using North Carolina birth data from 2003 to 2015 (n = 1 225 285) to investigate the relationship between air pollution and CHDs (specifically pulmonary valve atresia/stenosis, tetralogy of Fallot [TOF], and atrioventricular septal defect [AVSD]). Maternal exposure to particulate matter ≤ 2.5 μm in diameter (PM2.5) and ozone during weeks 3 to 9 of pregnancy were estimated using the Environmental Protection A…

Exposure to ethinyl estradiol during lactation reduces maternal mammary gland complexity and restricts pup growth in mice.

The mammary gland undergoes dramatic changes during pregnancy and lactation, remodeling a relatively simple ductal structure into an extensive network of milk secreting alveoli. Estradiol, through activation of estrogen receptor (ER), plays an essential role in the early expansion of the mammary epithelium during pregnancy, but has little role during lactation when circulating levels are low. Yet, ER is present in the epithelium during lactation and exposure to estrogenic endocrine disrupting chemicals (EDCs) could disrupt the lactational capability of the mammary gland. To understand the effe

Similar toxicity potential of glyphosate and glyphosate-based herbicide on cerebellar development after maternal exposure in rats.

Studies on the effects of glyphosate (GlyP) and glyphosate-based herbicides (GBHs) on cerebellar development are extremely limited. This study examined the effects of maternal exposure to GlyP and GBH on rat cerebellar development in male offspring. From day 6 of gestation until day 21 postpartum at weaning, dams were given GlyP at 1.5% or 3.0% in diet or GBH at 1.0% in drinking water (corresponding to 0.36% GlyP). At weaning, GBH exposure was linked to increased numbers of DCX+ migrating granule cells in the cortex and TUNEL+ apoptotic cells in the internal granular layer (IGL), suggesting th…

Maternal exposure to ZIF-8 derails placental function by inducing trophoblast pyroptosis through neutrophils activation in mice.

Adverse environmental factors during maternal gestation pose a threat to pregnancy. Environmental factors, particularly nanoparticles, can impact pregnancy by causing damage to the placenta. Compared to early gestation, foetuses in late gestation are more robustly developed and at lower risk of adverse effects from environmental factors. Delivery systems for targeted therapy during pregnancy is predominantly focused on their application in late gestation. Zeolitic imidazolate framework-8 (ZIF-8) holds great potential for targeted drug therapy. To evaluate the value of ZIF-8 in targeted treatme
