Chilling Out or Heating Up? Thermal Plasticity of Seminal Fluid Proteins in Drosophila melanogaster.

Seminal fluid proteins (SFPs) play crucial roles in reproduction by shaping female post-mating physiology and behaviour, male sperm competition and sexual conflict between the sexes. However, we know little about how abiotic ecological factors regulate SFP expression and transfer. Here, we use quantitative proteomics in adult flies from a wild Drosophila melanogaster population to test how exposure to natural temperature variation (20°C, 24°C and 28°C) across two time scales (48 h and 13d), and under either no- or high-sperm competition risk, interact to shape SFP production and transfer. Chasing the FoxO in Metabolic Disorders: Novel Considerations for Oxidative Stress, Programmed Cell Death, Wnt, and the Gut Microbiome.

Lifespan is increasing throughout the world leading to a rise in non-communicable diseases in the global population that impacts over 800 million individuals with metabolic disorders, such as diabetes mellitus. Metabolic disease presents a significant challenge for clinical care since multi-organ disease progression ensues despite a broad array of treatment protocols. The pursuit of innovative strategies with mammalian forkhead transcription factors of the "O" class (FoxOs) and intimately related pathways of aging, cellular senescence, telomere integrity, oxidative stress, programmed cell deat

The Impact of Leukemia Inhibitory Factor on Sirtuin1, Ilt-4, and Related microRNAs Expression in a Mouse Model of Recurrent Pregnancy Loss.

PURPOSE: Leukemia Inhibitory Factor (LIF) plays a crucial role in implantation and early pregnancy maintenance. However, its regulatory relationships with molecular markers such as Sirtuin 1 (SIRT1), immunoglobulin-like transcript 4 (ILT-4), and specific microRNAs (miRNA) in recurrent pregnancy loss (RPL) remain underexplored. METHODS: This study investigates the effect of LIF on the expression of Sirt1, Ilt-4, and related miRNAs using a mouse model of RPL. Female CBA/J mice mated with DBA/2 J males received LIF treatment. Gene and miRNA expression levels were assessed via quantitative PCR.

Prevalence of Same-Sex Sexual Behavior in Termites: Persistence Under Mate-Seeking Stress Absence.

Same-sex sexual behavior is an enigma in behavioral ecology as it does not result in reproduction. Previous studies on the evolution of same-sex sexual behavior have primarily focused on factors such as recognition mechanisms, resource constraints, and challenges in securing an opposite-sex partner, while overlooking the investigation into active same-sex sexual behavior without environmental stress. Here, through experimentation, we investigated the stability of same-sex pairs in tandem running, the mating behavior of pairs that form same-sex tandem runs, and the occurrence of same-sex sexual

Floral Biology, Breeding System and Conservation Implications for the Azorean Endemic Azorina vidalii (Campanulaceae).

This study investigates the seasonal and floral phenology, breeding strategies, and floral morphology of Azorina vidalii, an Azorean endemic Campanulaceae with hermaphroditic, protandrous flowers, dichogamy and secondary pollen presentation. Seasonal phenology was recorded in four field populations and floral phenology in a garden population. Reproductive strategies were assessed via controlled hand pollinations in one field population. Floral morphometrics were analysed using 23 floral and five pollen traits from 121 flowers across fourteen populations throughout the Azores archipelago. Non-p

MT1/cAMP/PKA Pathway in Melatonin-Regulated Sperm Capacitation.

Melatonin is mainly synthesized and secreted by pineal gland, and plays multiple functions, including its regulating effects on reproductive processes. Sperm capacitation is necessary for fertilization, but the effect of melatonin on mouse sperm capacitation remains to be elucidated. We thus investigated the roles of melatonin on capacitation by culturing the sperms from mouse cauda epididymis in the medium with different doses of melatonin. The results showed that 10-7 mol/L melatonin significantly enhanced the sperm capacitation by increasing the sperm tyrosine phosphorylation level, percent

Insights on reproduction-related genes in the striped fruit fly, Zeugodacus scutellata (Hendel) (Diptera: Tephritidae).

The striped fruit fly, Zeugodacus scutellata is a significant pest in East and Southeast Asia by damaging Cucurbitaceae blossoms and fruits. To control this pest, a novel strategy to suppress the gene(s) associated with sexually dimorphic phenotypes has been devised and implemented in a laboratory scale. However, comprehensive transcriptomic analysis related to this sex differentiation of Z. scutellata was necessary to determine effective target genes for the genetic control. We performed de novo assembly of the transcript obtained by paired-end sequencing using an Illumina HiSeq platform and

Insulin Elevates ID2 Expression in Trophoblasts and Aggravates Preeclampsia in Obese ASB4-Null Mice.

Obesity is a risk factor for preeclampsia. We investigated how obesity influences preeclampsia in mice lacking ankyrin-repeat-and-SOCS-box-containing-protein 4 (ASB4), which promotes trophoblast differentiation via degrading the inhibitor of DNA-binding protein 2 (ID2). Asb4-/- mice on normal chow (NC) develop mild preeclampsia-like phenotypes during pregnancy, including hypertension, proteinuria, and reduced litter size. Wild-type (WT) and Asb4-/- females were placed on a high-fat diet (HFD) starting at weaning. At the age of 8-9 weeks, they were mated with WT or Asb4-/- males, and preeclampt

Reproductive biology of harvestmen (Arachnida: Opiliones): a review of a rapidly evolving research field.

Harvestmen are a major arachnid order that has experienced a dramatic increase in biological knowledge in the 21st century. The publication of the book Harvestmen: The Biology of Opiliones in 2007 stimulated the development of many behavioral studies. Although the book is relatively recent, our understanding of the reproductive biology of harvestmen is already outdated due to the fast accumulation of new data. Our goal is to provide an updated review of the subject to serve as a benchmark for the following years. In the pre-copulatory phase, we explore the evolution of facultative parthenogene

The role of FGF-4 and FGFR-2 on preimplantation embryo development in experimental maternal diabetes.

OBJECTIVE: Diabetes mellitus can cause spontaneous abortion, neonatal diseases, congenital malformations, and death. There are many studies related to the damage of in vitro hyperglycemia on embryogenesis in literature, but not enough studies on in vivo hyperglycemia effects on embryogenesis. Fibroblast growth factor (FGF) molecules play an essential role in pre-implantation embryo development and diabetes pathogenesis. In our study, we researched whether FGF-4 and FGFR-2 were playing a role in maternal diabetes' effects on embryo development. MATERIAL AND METHODS: Thirty adult virgin female

Oxytocin and CD38 in the paraventricular nucleus play a critical role in paternal aggression in mice.

In mammals, the development of healthy offspring requires maternal care. Behavior by lactating mothers toward other individuals is an important component of maternal aggression. However, it is unclear whether fathers display aggression primed by pups (an external factor), and the protection mechanism is poorly understood. To address this question, we examined paternal aggression in the ICR mouse strain. We found that sires exposed to cues from pups and lactating dams showed stronger aggression toward intruders than did sires that were deprived of family cues or exposed to nonlactating mates. c

Maternal folic acid depletion during early pregnancy increases sensitivity to squamous tumor formation in the offspring in mice.

Gestational nutrition is widely recognized to affect an offspring's future risk of lifestyle-related diseases, suggesting the involvement of epigenetic mechanisms. As folic acid (FA) is a nutrient essential for modulating DNA methylation, we sought to determine how maternal FA intake during early pregnancy might influence tumor sensitivity in an offspring. Dams were maintained on a FA-depleted (FA(-)) or normal (2 mg FA/kg; FA(+)) diet from 2 to 3 days before mating to 7 days post-conception, and their offspring were challenged with chemical tumorigenesis using 7,12-dimethylbenz[a)anthracene a

Alliance of Efflux Pumps with β-Lactamases in Multidrug-Resistant Klebsiella pneumoniae Isolates.

Nosocomial infections caused by Klebsiella pneumoniae are primarily characterized by a high prevalence of extended-spectrum β-lactamases (ESBL's) and a soaring pace of carbapenemase dissemination. Availability of limited antimicrobial agents as a therapeutic option for multidrug-resistant bacteria raises an alarming concern. This study aimed at the molecular characterization of multidrug-resistant K. pneumoniae clinical isolates and studied the role of efflux pumps in β-lactam resistance. Thirty-three isolates confirmed as ESBL-positive K. pneumoniae that harbored resistance genes to major cla

The Safe Recipient of SSC Transplantation Prepared by Heat Shock With Busulfan Treatment in Mice.

Safety is the chief consideration in recipient preparation of spermatogonial stem cell (SSC) transplantation in mammals, especially humans. In this study, we compared the safety of the SSC transplantation recipients that were prepared both by testes heat shock plus testes busulfan injection (heat shock+busulfan(t)) and by busulfan intraperitoneal injection (busulfan i.p.) only. Our results showed that heat shock+busulfan(t) treatment significantly ( p < 0.05) reduced mortality in mice and did not produce bone marrow cell toxicity. Furthermore, heat shock+busulfan(t) treatment directly damaged

Passion, Relational Mobility, and Proof of Commitment: A Comparative Socio-Ecological Analysis of an Adaptive Emotion in a Sexual Market.

Although monogamy, the exclusive bonding with a specific partner, is one characteristic of modern human mating, long-term romantic relationships inherently possess the commitment problem, which is the conflict between maintaining a relationship with a certain partner and seeking attractive alternatives. Frank has argued that love and passion help solve this problem because they make individuals commit voluntarily to the relationship, leading the other party to also be committed with less concern over being cheated on or rejected. Combining this idea with the comparative socio-ecological approa

Allergy and immunology in Africa: Challenges and unmet needs.

The tremendous increase in allergy in the African continent cannot simply be explained by the change in public hygiene. There are many "prehygiene" communities with sewage-contaminated water supplies, helminth infestations, bare footedness, and poor housing, and still there is a high prevalence of allergic disease. Africans can be exposed to many risk factors facilitating severe asthma and wheezing, including airborne viruses, smoke, indoor dampness, cockroaches, and poor access to health care. Although the reporting on food allergy is inadequate to perform systematic reviews or meta-analyses,

Increased Toll-Like Receptor-Myeloid Differentiation Factor 88 Expression at the Maternal-Fetal Interface Is Associated with Spontaneous Abortion.

AIMS: Toll-like receptors (TLRs) form a group of pattern recognition receptors that play a major role in maintaining innate host immunity. Myeloid differentiation factor 88 (MyD88) is an adaptor molecule that is essential for signaling via the TLR family. To analyze the mechanism of the imbalance in the innate immune system mediated by TLRs-MyD88 in spontaneous abortion, we detected the expression of TLR-2, TLR-4, TLR-7, and MyD88 in placentae and deciduas. METHODS: The expression of TLR-2, TLR-4, TLR-7, and MyD88 in the placentae and deciduas of abortion-prone mice (n = 10) and normal pregna

Sensorimotor gating deficits are inheritable in an isolation-rearing paradigm in rats.

Early life experience is a key etiological factor of neuropsychiatric dysfunctions and is associated with developmental origins. Impaired prepulse inhibition (PPI) following an acoustic startle response is acknowledged as a cardinal characteristic in socially deprived weanling rats, which has been employed to investigate the underlying mechanisms of sensorimotor gating abnormalities in certain mental disorders, including schizophrenia. Because impaired PPI is a postnatal malfunction, it is interesting to examine whether it can be passed to the next generation. Isolation-rearing (IR) rats had b

The Transcription Factor NFIL3 Is Essential for Normal Placental and Embryonic Development but Not for Uterine Natural Killer (UNK) Cell Differentiation in Mice.

Mice ablated for the gene encoding the transcription factor Nfil3 lack peripheral natural killer (NK) cells but retain tissue-resident NK cells, particularly in mucosal sites, including virgin uterus. We undertook a time course histological study of implantation sites from syngeneically (Nfil3(-/-)) and allogeneically (BALB/c) mated Nfil3(-/-) females. We also examined implantation sites from Rag2(-/-)Il2rg(-/-) females preconditioned by adoptive transfer of Nfil3(-/-) marrow or uterine cell suspensions to identify the Nfil3(-/-) pregnancy aberrations that could be attributed to nonlymphoid ce

Novel nervous and multi-system regenerative therapeutic strategies for diabetes mellitus with mTOR.

Throughout the globe, diabetes mellitus (DM) is increasing in incidence with limited therapies presently available to prevent or resolve the significant complications of this disorder. DM impacts multiple organs and affects all components of the central and peripheral nervous systems that can range from dementia to diabetic neuropathy. The mechanistic target of rapamycin (mTOR) is a promising agent for the development of novel regenerative strategies for the treatment of DM. mTOR and its related signaling pathways impact multiple metabolic parameters that include cellular metabolic homeostasis
