The risk of determining risk with multivariable models.

PURPOSE: To review the principles of multivariable analysis and to examine the application of multivariable statistical methods in general medical literature. DATA SOURCES: A computer-assisted search of articles in The Lancet and The New England Journal of Medicine identified 451 publications containing multivariable methods from 1985 through 1989. A random sample of 60 articles that used the two most common methods--logistic regression or proportional hazards analysis--was selected for more intensive review. DATA EXTRACTION: During review of the 60 randomly selected articles, the focus was

Critical phenomena in the majority voter model on two-dimensional regular lattices.

In this work we studied the critical behavior of the critical point as a function of the number of nearest neighbors on two-dimensional regular lattices. We performed numerical simulations on triangular, hexagonal, and bilayer square lattices. Using standard finite-size scaling theory we found that all cases fall in the two-dimensional Ising model universality class, but that the critical point value for the bilayer lattice does not follow the regular tendency that the Ising model shows.

Artificial multiple criticality and phase equilibria: an investigation of the PC-SAFT approach.

The perturbed-chain statistical associating fluid theory (PC-SAFT) is studied for a wide range of temperature, T, pressure, p, and (effective) chain length, m, to establish the generic phase diagram of polymers according to this theory. In addition to the expected gas-liquid coexistence, two additional phase separations are found, termed "gas-gas" equilibrium (at very low densities) and "liquid-liquid" equilibrium (at densities where the system is expected to be solid already). These phase separations imply that in one-component polymer systems three critical points occur, as well as equilibri

Generating survival times to simulate Cox proportional hazards models.

Simulation studies present an important statistical tool to investigate the performance, properties and adequacy of statistical models in pre-specified situations. One of the most important statistical models in medical research is the proportional hazards model of Cox. In this paper, techniques to generate survival times for simulation studies regarding Cox proportional hazards models are presented. A general formula describing the relation between the hazard and the corresponding survival time of the Cox model is derived, which is useful in simulation studies. It is shown how the exponential

Modeling longitudinal data with nonparametric multiplicative random effects jointly with survival data.

In clinical studies, longitudinal biomarkers are often used to monitor disease progression and failure time. Joint modeling of longitudinal and survival data has certain advantages and has emerged as an effective way to mutually enhance information. Typically, a parametric longitudinal model is assumed to facilitate the likelihood approach. However, the choice of a proper parametric model turns out to be more elusive than models for standard longitudinal studies in which no survival endpoint occurs. In this article, we propose a nonparametric multiplicative random effects model for the longitu

Dimensionality of hallucinogen and inhalant/solvent abuse and dependence criteria: implications for the Diagnostic and Statistical Manual of Mental Disorders-Fifth Edition.

BACKGROUND: Prior research has demonstrated the dimensionality of Diagnostic and Statistical Manual of Mental Disorders-Fourth Edition (DSM-IV) alcohol, nicotine, cannabis, cocaine and amphetamine abuse and dependence criteria. The purpose of this study was to examine the dimensionality of hallucinogen and inhalant/solvent abuse and dependence criteria. In addition, we assessed the impact of elimination of the legal problems abuse criterion on the information value of the aggregate abuse and dependence criteria, another proposed change for DSM-IV currently lacking empirical justification. Nonparametric inference on the E/O ratio in model validation.

In preventing diseases such as cancers and osteoporosis, statistical models are often used to identify subjects with high risks. The ratio of the expected (or predicted) number of cases in the target population and the observed numbers of cases (the E/O ratio) is a useful quantity to evaluate the goodness of fit of the model. The model is usually evaluated on a sample taken from the target population and, in the literature, statistical inferences on the E/O ratio often assume that the expected number is a constant and the observed number follows a Poisson distribution. In this paper, we introd

Item response mixture modeling: application to tobacco dependence criteria.

This paper illustrates new hybrid latent variable models that are promising for phenotypical analyses. The hybrid models combine features of dimensional and categorical analyses seen in the conventional techniques of factor analysis and latent class analysis. The paper focuses on the analysis of categorical items, which presents especially challenging analyses with hybrid models and has recently been made practical in the Mplus program. The hybrid models are typically seen to fit data better than conventional models of factor analysis (IRT) and latent class analysis. An illustration is given i

Comparing the sensitivity of cross-over and parallel study designs for QTc assessment: An analysis based on a single large study of moxifloxacin in 48 nonhuman primates.

The cardiovascular safety pharmacology (SP) study conducted to satisfy ICH S7A and S7B has commonly used a cross-over study design where each animal receives all treatments. In an increasing number of cases, cross-over designs are not possible and parallel studies have to be used. These can seldom be as large as 8 animals/treatment to match an n = 8 cross-over. Animals in parallel designs receive only one treatment. Parallel studies will have a different sensitivity to detect changes. This sensitivity is a critical question in using nonclinical QTc evaluations to support an integrated proarrhy

DyVAESurv: A VAE-Enhanced Model for Dynamic Survival Analysis.

Survival analysis plays a crucial role in discovering the relationship between covariates and event times, which is widely applied in individual risk prediction. Traditional methods often rely on static features, resulting in the difficulty of capturing time dependencies, while dynamic survival models have limitations in capturing heterogeneity among individuals. To address these issues, a deep learning model, integrating original features with latent representations generated by a variational autoencoder (VAE), is proposed to better characterize individual survival trajectories. The model emp

The Use of Real-World Data for Studies of Dynamic Disease Processes.

Obtaining valid real-world evidence about intervention effects from observational cohorts or administrative health records data is challenging. Visits to healthcare providers tend to occur more often during periods of increased disease activity and symptom exacerbation, or upon disease progression. Treatments likewise tend to change when it is apparent that disease activity has increased or a meaningful progression has occurred. This creates a dual problem in which patient visits are disease-related and treatments changes are driven by disease condition and clinical presentation. Disease-relat

Predicting the incidence of syphilis in China using Baidu search index and an ARIMAX model.

BACKGROUND: Existing studies have confirmed the value of Internet search data in predicting infectious disease trends. This study aims to develop a predictive model using the Baidu Index to examine the relationship between the search volume of syphilis-related keywords and the prevalence of syphilis in China. METHODS: We collected daily reported syphilis case counts and Baidu search volumes for syphilis-related keywords from January 2011 to March 2025. Keywords highly correlated with syphilis incidence were identified using Spearman rank correlation and time series cross-correlation analysis,

Implementation of a dynamic model updating pipeline provides a systematic process for maintaining performance of prediction models.

OBJECTIVES: We describe the steps for implementing a dynamic updating pipeline for clinical prediction models and illustrate the proposed methods in an application of 5-year survival prediction in cystic fibrosis. STUDY DESIGN AND SETTING: Dynamic model updating refers to the process of repeated updating of a clinical prediction model with new information to counter performance degradation. We describe 2 types of updating pipeline: "proactive updating" where candidate model updates are tested any time new data are available, and "reactive updating" where updates are only made when performance

Runtime verification in uncertain environment based on probabilistic model learning.

Runtime verification (RV) is a lightweight approach to detecting temporal errors of system at runtime. It confines the verification on observed trajectory which avoids state explosion problem. To predict the future violation, some work proposed the predictive RV which uses the information from models or static analysis. But for software whose models and codes cannot be obtained, or systems running under uncertain environment, these predictive methods cannot take effect. Meanwhile, RV in general takes multi-valued logic as the specification languages, for example the true, false and inconclusiv

The κ-statistics approach to epidemiology.

A great variety of complex physical, natural and artificial systems are governed by statistical distributions, which often follow a standard exponential function in the bulk, while their tail obeys the Pareto power law. The recently introduced [Formula: see text]-statistics framework predicts distribution functions with this feature. A growing number of applications in different fields of investigation are beginning to prove the relevance and effectiveness of [Formula: see text]-statistics in fitting empirical data. In this paper, we use [Formula: see text]-statistics to formulate a statistica

A dynamic Mover-Stayer model for recurrent event processes subject to resolution.

In studies of affective disorder, individuals are often observed to experience recurrent symptomatic exacerbations warranting hospitalization. Interest may lie in modeling the occurrence of such exacerbations over time and identifying associated risk factors. In some patients, recurrent exacerbations are temporally clustered following disease onset, but cease to occur after a period of time. We develop a dynamic Mover-Stayer model in which a canonical binary variable associated with each event indicates whether the underlying disease has resolved. An individual whose disease process has not re

Modelling tuberculosis trends in the USA.

We present a mathematical transmission model of tuberculosis in the USA. The model is calibrated to recent trends of declining incidence in the US-born and foreign-born populations and is used in assessing relative impacts of treatment of latently infected individuals on elimination time, where elimination is defined as annual incidence <1 case/million. Provided current control efforts are maintained, elimination in the US-born population can be achieved before the end of this century. However, elimination in the foreign-born population is unlikely in this timeframe even with higher rates of t

Quantitative assessment of age-related macular degeneration using parametric modeling of the leakage transfer function: preliminary results.

Age-related macular degeneration (AMD) is a major cause of blindness and visual impairment in older adults. The wet form of the disease is characterized by abnormal blood vessels forming a choroidal neovascular membrane (CNV), that result in destruction of normal architecture of the retina. Current evaluation and follow up of wet AMD include subjective evaluation of Fluorescein Angiograms (FA) to determine the activity of the lesion and monitor the progression or regression of the disease. However, this subjective evaluation prevents accurate monitoring of the disease progression or regression

Statistical analysis of comparative field trials of acute rodenticides.

The present method of analysis of data from comparative field trials of acute rodenticides was compared with two alternative methods of analysis, using computer simulation techniques. One of the proposed alternative analyses, the use of post-treatment census takes as a percentage of pre-treatment census takes in an analysis of variance, was found to be more accurate, to avoid a theoretical difficulty associated with the present method, and is computationally much simpler.
