A review of the genotoxicity of the industrial chemical cumene.

The purpose of this review is to evaluate the literature on the genotoxicity of cumene (CAS # 98-82-8) and to assess the role of mutagenicity, if any, in the mode of action for cumene-induced rodent tumors. The studies reviewed included microbial mutagenicity, DNA damage/ repair, cytogenetic effects, and gene mutations. In reviewing these studies, attention was paid to their conformance to applicable OECD test guidelines which are considered as internationally recognized standards for performing these assays. Cumene was not a bacterial mutagen and did not induce Hprt mutations in CHO cell cult

Synthesis and Trace-Level Quantification of Mutagenic and Cohort-of-Concern Ciprofloxacin Nitroso Drug Substance-Related Impurities (NDSRIs) and Other Nitroso Impurities Using UPLC-ESI-MS/MS-Method Optimization Using I-Optimal Mixture Design.

Globally, the pharmaceutical industry has been facing challenges from nitroso drug substance-related impurities (NDSRIs). In the current study, we synthesized and developed a rapid new UPLC-MS/MS method for the trace-level quantification of ciprofloxacin NDSRIs and a couple of N-nitroso impurities simultaneously. (Q)-SAR methodology was employed to assess and categorize the genotoxicity of all ciprofloxacin N-nitroso impurities. The projected results were positive, and the cohort of concern (CoC) for all three N-nitroso impurities indicates potential genotoxicity. AQbD-driven I-optimal mixture

[Sister chromatid exchanges and chromosome aberrations as parameters for human risk of cancer development].

Chromosome alterations, which are directly visible changes in the DNA, have close associations to cancer development, non-specific ageing, and heritable genetic status. Human lymphocyte cultures can be used for cytogenetic monitoring of genetic health because many cancers and genetic effects are caused by long-term unhealthy life-styles. We have investigated the sensitivities of lymphocytes from inherited-cancer-prone diseases to the induction of the chromosome alterations by mutagens and carcinogens, and the correlations between the frequency of sister chromatid exchanges (SCEs) in peripheral…

Geographic Range Size Predicts Butterfly Species' Tolerance to Heavy Metals More Than Evolutionary History With Toxic Larval Diets.

Some organisms appear to thrive in contaminated environments, while others are more sensitive, though the causes of this variation are uncle … s. We used this dataset, coupled with butterfly host records, to estimate evolutionary history with mutagens. We found that butterfly species with larger ranges tolerated significantly greater concentrations of lead, arsenic, and cadmium in their tissues. Additionally, species with a history of feeding on relatively more mutagenic host plant families toler …

Evaluation of the genotoxic potential of certain pesticides used in Pakistan.

The mutagenicity of fifteen insecticides, five fungicides, four herbicides, and an acaricide commonly used in Pakistan was evaluated by employing thirteen short-term bioassays. The genetic endpoints used included point or gene mutation, primary DNA damage, and chromosomal effects. Initially, all pesticides were tested in a "core" battery of four in vitro bioassays. A carefully selected group among these chemicals was retested in higher level test systems to confirm the results obtained in the initial phase. Of the pesticides tested, only a small portion consistently displayed mutagenicity acro

Genotoxicity of drinking waters.

Drinking waters in many countries have been shown to contain non-volatile polar mutagenic compounds as well as trihalomethanes. Their nature is uncertain, but one compound, a chlorinated furanone (3-chloro-4-(dichloro-methyl)-5-hydroxy-2(5H)-furanone;MX) seems to be responsible for one third to over one half of the mutagenicity. Its concentration seems to correlate rather well also with the total bacterial mutagenicity. Animal toxicity studies are being used to evaluate the possible carcinogenicity risk of this compound as a first stage in complete risk assessment of the whole complex mixture.

Chemical models for cytochrome P450 as a biomimetic metabolic activation system in mutation assays.

DNA damage is a critical factor in carcinogenesis. The Ames assay is a short-term test that screens for DNA-damaging agents. To be detected in the assay, most carcinogens require oxidation by cytochrome P450, a component of the liver homogenate preparation (S9 mix) that is traditionally used to metabolize promutagens to an active form in vitro. A combination of iron(III) porphyrin plus an oxidant activates many promutagens by mimicking cytochrome P450 metabolism. We previously reported that the mutagenicity of the N-nitrosodialkylamines was detected following reaction with tetrakis(pentafluoro…

UV and ethyl methanesulfonate effects in hyperthermophilic archaea and isolation of auxotrophic mutants of Pyrococcus strains.

The lethal and mutagenic effects of ethyl methanesulfonate (EMS) and UV on nine archaeal strains belonging to each of the two described genera of Thermococcales, Pyrococcus and Thermococcus, were investigated. To test the efficiency of the EMS and UV mutagenesis under a variety of experimental conditions, we chose Pyrococcus abyssi strain GE5 as a model strain. We observed a strong induced mutagenicity in both cases, since the spontaneous mutation frequency (expressed as the frequency of resistance to 5-fluoroorotic acid) increased up to 150-fold with EMS and 400-fold with UV, after mutagen ex

DNA fragility and repair capability are separate genetic phenotypes - studies on in-situ nick translation and chromosome breakage.

Three separate experiments, designed to minimize the DNA repair process following exposures to mutagens, were performed to determine whether DNA repair capability is the sole factor responsible for differential mutagen sensitivity. These experiments included (i) very short bleomycin pulse treatments (2, 5 and 10 min), (ii) ionizing irradiation and (iii) bleomycin treatment with DNA repair inhibited by ethanol. Three lymphoblastoid cell lines, derived (i) from a patient with ataxia telangiectasia (AT), (ii) a patient with lupus erythematosus and (iii) a normal blood donor, were used as test cel…

Preliminary Genotoxicity Assessment of Calcium Phosphate Cement Incorporated with Palm Tocotrienol Using Bacterial Reverse Mutation Assay.

Background/Objectives: Calcium phosphate cement (CPC) is extensively utilised in bone repair owing to its biocompatibility, osteoconductivity, and compositional similarity to native bone. Functionalisation of CPC with palm tocotrienol may enhance its regenerative potential. However, the incorporation of phytochemicals requires safety evaluation to exclude potential genotoxic risks. This study investigated the mutagenic potential of CPC and tocotrienol-enriched CPC (CPC-T3) using the bacterial reverse mutation assay. Methods: Mutagenicity was evaluated in five bacterial strains, including Salmo
