An Elf2-like transcription factor acts as repressor of the mouse ecto-5'-nucleotidase gene expression in hepatic myofibroblasts.

Hepatic fibrosis represents a pathological wound healing and tissue repair process triggered in response to chronic liver injury. A heterogeneous population of activated non-parenchymal liver cells, known as liver myofibroblasts, functions as the effector cells in hepatic fibrosis. Upon activation, liver myofibroblasts become fibrogenic, acquiring contractile properties and increasing collagen production capacity, while developing enhanced sensitivity to endogenous molecules and factors released in the local microenvironment. Hepatic extracellular adenosine is a bioactive small molecule, incre…

Massive Accumulation of Myofibroblasts in the Critical Isthmus Is Associated With Ventricular Tachycardia Inducibility in Post-Infarct Swine Heart.

OBJECTIVES: In this study the authors determined the extent of cellular infiltration and dispersion, and regional vascularization in electrophysiologically (EP) defined zones in post-myocardial infarction (MI) swine ventricle. BACKGROUND: The critical isthmus (CI) in post-MI re-entrant ventricular tachycardia (VT) is a target for catheter ablation. In vitro evidence suggests that myofibroblasts (MFB) within the scar border zone (BZ) may increase the susceptibility to slow conduction and VT, but whether this occurs in vivo remains unproven. METHODS: Six weeks after mid-left anterior descendin…

[Role of p38 mitogen activated protein kinase on cyclic stretch in human facial hypertrophic scar fibroblasts differentiation into myofibroblasts].

OBJECTIVE: To explore the signal transduction mechanism of p38 mitogen activated protein kinase (p38MAPK) in human facial hypertrophic scar fibroblast (FB) differentiation into myofibroblasts (MFB). METHODS: Fibroblasts of primary culture were simple randomly assigned into two groups: cyclic stretch (control group) and cyclic stretch pre-treated with SB203580(experimental group). Expression of P-p38MAPK and α-smooth muscle actin (α-SMA) protein were examined using Western blotting and expression of transforming growth factor β1 (TGF-β1) mRNA and α-SMA mRNA were examined using reverse transcri…

Monocyte chemoattractant protein-1 production by intestinal myofibroblasts in response to staphylococcal enterotoxin a: relevance to staphylococcal enterotoxigenic disease.

Food poisoning due to staphylococcal enterotoxins A and B (SEA and SEB) affects hundreds of thousands of people annually. SEA and SEB induce massive intestinal cytokine production, which is believed to be the key factor in staphylococcal enterotoxin enteropathy. MHC class II molecules are the major receptors for staphylococcal enterotoxins. We recently demonstrated that normal human subepithelial intestinal myofibroblasts (IMFs) express MHC class II molecules. We hypothesized that IMFs are among the first cells to respond to staphylococcal enterotoxins and contribute to the cytokine production…

Evidence for modulation of pericryptal sheath myofibroblasts in rat descending colon by transforming growth factor beta and angiotensin II.

BACKGROUND: Absorption of water and Na+ in descending colonic crypts is dependent on the barrier function of the surrounding myofibroblastic pericryptal sheath. Here the effects of high and low Na+ diets and exposure to whole body ionising radiation on the growth and activation of the descending colonic pericryptal myofibroblasts are evaluated. In addition the effect of a post-irradiation treatment with the angiotensin converting enzyme inhibitor Captopril was investigated. METHODS: The levels of Angiotensin II type 1 receptor (AT1), ACE, collagen type IV, transforming growth factor-beta type…

Expression of myosin heavy chain isoforms in mammary epithelial cells and in myofibroblasts from different fibrotic settings during neoplasia.

The expression of smooth muscle (SM) and non-muscle (NM) myosin heavy chain (MyHC) isoforms has been studied in fibroblastic cells of different fibrotic lesions (hypertrophic scars, Dupuytren's nodules and stromal reaction to mammary carcinoma) and in epithelial cells of non-neoplastic and neoplastic mammary glands, using anti-myosin antibodies in immunofluorescence and Western blotting. Two antibodies were specific for SM-MyHC isoforms (SM1 and SM2) and three antibodies were directed against different sequences of NM-MyHC isoforms. Myofibroblasts containing SM-MyHC were present in a variable …

STL1267 inhibits myofibroblast differentiation in a TGFβ1-driven human lung fibroblast model.

Pulmonary fibrosis is a progressive interstitial lung disease characterized by excessive fibroblast-to-myofibroblast transition (FMT) and extracellular matrix (ECM) deposition, largely driven by transforming growth factor-beta 1 (TGFβ1). Existing therapies offer limited efficacy, particularly in advanced disease. Circadian rhythms have recently emerged as key modulators of lung inflammation and fibrosis. In this study, we developed an in vitro model of chronic fibrotic signaling using adenovirus-mediated TGFβ1 overexpression (Ad-TGFβ1) or human recombinant protein TGFβ1 in primary normal human

Pivotal role of FARP2 in TGF-β1-mediated myofibroblast differentiation and corneal fibrosis.

Corneal stromal fibrosis represents a major cause of visual impairment, primarily driven in part by the TGF-β1-mediated transition of stromal fibroblasts into contractile myofibroblasts. FARP2, a mediator of Rho-GTPase signaling and cytoskeletal organization, has been proposed as a potential mediator of this phenotypic transition; however, the specific functional contribution stromal remodeling remains largely undefined) its functional role in corneal stromal remodeling remains unclear. CSFs were isolated from New Zealand rabbits and cultured under standard conditions. Myofibroblast differenti…

Primary Inflammatory Myofibroblastic Tumour of the Breast: A Potential Diagnostic Pitfall on Aspiration Cytology.

Inflammatory myofibroblastic tumour (IMT) is an uncommon mesenchymal neoplasm with intermediate malignant potential, typically arising in the lungs, abdomen, or retroperitoneum. Breast involvement is extremely rare, with limited reported cases. To the best of our knowledge, less than 50 cases of this lesion in the breast have been reported to date and cytological descriptions are very rare. A 47-year-old female presented with a painless mass in the right breast. Imaging revealed a well-defined lobulated soft tissue mass categorised as BIRADS 4A. The cytomorphologic features on fine needle aspi

Aberrant expression of the PREX2-MAGI2 axis modulates myofibroblast differentiation in pulmonary fibrosis.

Myofibroblast differentiation is a pivotal event in idiopathic pulmonary fibrosis (IPF) and is driven by transforming growth factor-β1 (TGF- … foci of IPF-affected lungs. Fluorescence immunostaining revealed PREX2 colocalization with α-SMA in myofibroblasts within fibroblastic foci, corroborating our hypothesis. These findings suggest that MAGI2 acts as a negative regulator of myofibroblast differentiation by stabilizing PTEN, whereas PREX2 is aberrantly expressed in the fibrotic milieu, negatively mod …

PTEN Regulates Myofibroblast Activation in Valvular Interstitial Cells Based on Subcellular Localization.

Aortic valve stenosis (AVS) is characterized by altered mechanics of the valve leaflets, which disrupts blood flow through the aorta and can cause left ventricle hypotrophy. These changes in the valve tissue result in the activation of resident valvular interstitial cells (VICs) into myofibroblasts, which have increased levels of αSMA in their stress fibers. The persistence of VIC myofibroblast activation is a hallmark of AVS. In recent years, the tumor suppressor gene phosphatase and tensin homolog (PTEN) has emerged as an important player in the regulation of fibrosis in various tissues (e.g…

Circular RNA RORβ regulates TGFβR1 in alcohol-induced fibroblast-to-myofibroblast differentiation.

Alcohol exposure augments the expression and signaling of transforming growth factor-beta (TGFβ), leading to fibroproliferation. We showed that inhibition of TGFβ receptor type 1 (TGFβR1) mitigates the effect of alcohol in the lung. We further demonstrated that alcohol modulates TGFβ signaling, partly through its ability to modify microRNA (miRNA or miR) expressions in the lung. The mechanism by which alcohol mediates miRNA expressions and how its connection to TGFβ signaling has not been well elucidated. Circular RNAs (circRNAs or circ) have emerged as potential therapeutic targets due to the

Cellular senescence promotes macrophage-to-myofibroblast transition in chronic ischemic renal disease.

Cellular senescence participates in the pathophysiology of post-stenotic kidney damage, but how it regulates tissue remodeling is incompletely understood. Macrophage-myofibroblast transition (MMT) contributes to the development of tissue fibrosis. We hypothesized that cellular senescence contributes to MMT and renal fibrosis in mice with renal artery stenosis (RAS). INK-ATTAC mice expressing p16INK-4a and green fluorescent protein in senescent cells were assigned to control or unilateral RAS, untreated or treated with AP20187 (an apoptosis inducer in p16INK-4a-expressing cells) for 4 weeks. Rapidly Growing ALK-Negative NTRK3-Positive Inflammatory Myofibroblastic Tumour of the Lung.

Inflammatory myofibroblastic tumours (IMT) are rare mesenchymal neoplasms that account for less than 1% of all lung tumours. Although ~50%-60% of IMT cases are anaplastic lymphoma kinase (ALK)-positive, ALK-negative IMT cases present diagnostic challenges owing to diverse pathological features. Here, we report a rare case of a rapidly growing ALK-negative, neurotrophic tyrosine receptor kinase 3 (NTRK3)-positive pulmonary IMT in a 46-year-old man with a history of pulmonary tuberculosis. A 13-mm nodule detected during routine screening grew to 18 mm within 1 month. Complete surgical resection
