Pharmacological modulation of directed network communication and neural hubs in action-effect integration.

BACKGROUND: Acting intentionally requires individuals to anticipate the effects of their actions. Recent work has revealed the neural oscillatory dynamics underlying the establishment of action-effect bindings, which are vital to anticipating action effects. However, the neurobiological basis of these processes is elusive. METHODS: Healthy adult participants (N = 54) engaged in a double-blind, counter-balanced, placebo-controlled experiment in which they worked in an experiment able to examine how action effects are planned, anticipated, and processed under placebo and methylphenidate conditi

Transcranial magneto-acoustic stimulation suppresses the power and coherence of beta-oscillations in parkinsonian rat model with improvement motor performance.

OBJECTIVE: Transcranial magneto-acoustic stimulation (TMAS) is an innovative non-invasive technique designed for neural modulation, utilizing coupled magnetic and acoustic fields to regulate brain neural activity. Alterations in the electrophysiological activity between the motor cortex (MC) and subthalamic nucleus (STN) are implicated in the motor impairments of Parkinson's disease (PD). METHOD: This study aimed to investigate the alterant electrophysiological activities in the STN-MC pathway. Using TMAS on the STN of PD rats, changes in beta-frequency(12-30 Hz) power and coherence in the mo

Immune pathway activation in neurons triggers neural damage after stroke.

Ischemic brain injury is a severe medical condition with high incidences in elderly people without effective treatment for the resulting neural damages. Using a unilateral mouse stroke model, we analyze single-cell transcriptomes of ipsilateral and contralateral cortical penumbra regions to objectively reveal molecular events with single-cell resolution at 4 h and 1, 3, and 7 days post-injury. Here, we report that neurons are among the first cells that sense the lack of blood supplies by elevated expression of CCAAT/enhancer-binding protein β (C/EBPβ). To our surprise, the canonical inflammato

Challenges of neural interfaces for stroke motor rehabilitation.

More than 85% of stroke survivors suffer from different degrees of disability for the rest of their lives. They will require support that can vary from occasional to full time assistance. These conditions are also associated to an enormous economic impact for their families and health care systems. Current rehabilitation treatments have limited efficacy and their long-term effect is controversial. Here we review different challenges related to the design and development of neural interfaces for rehabilitative purposes. We analyze current bibliographic evidence of the effect of neuro-feedback i

Decreased investigatory head scanning during exploration in learning-impaired, aged rats.

"Head scanning" is an investigatory behavior that has been linked to spatial exploration and the one-trial formation or strengthening of place cells in the hippocampus. Previous studies have demonstrated that a subset of aged rats with normal spatial learning performance show head scanning rates during a novel, local-global cue-mismatch manipulation that are similar to those of young rats. However, these aged rats demonstrated different patterns of expression of neural activity markers in brain regions associated with spatial learning, perhaps suggesting neural mechanisms that compensate for a

Brain activity underlying face and face pareidolia processing: an ERP study.

BACKGROUND AND PURPOSE: Face pareidolia is described as an interpretation of any unrelated object seen for the first time as a face. It is still unclear how to face pareidolia is processed. In this study, the neural basis of face and face pareidolia processing was investigated through recording event-related potentials (ERPs). METHODS: The ERPs were recorded from 35 right-handed and healthy participants in response to faces and face pareidolia. Amplitudes and latencies of N170, vertex-positive potential (VPP), and N250 components were analyzed, and current source density (CSD) maps relevant t

Simplification of neural networks for skin lesion image segmentation using color channel pruning.

Automatic analysis of skin abnormality is an effective way for medical experts to facilitate diagnosis procedures and improve their capabilities. Efficient and accurate methods for analysis of the skin abnormalities such as convolutional neural networks (CNNs) are typically complex. Hence, the implementation of such complex structures in portable medical instruments is not feasible due to power and resource limitations. CNNs can extract features from the skin abnormality images automatically. To reduce the burden of the network for feature extraction, which can lead to the network simplicity,

Mitochondrial dysfunction is implicated in retinoic acid-induced spina bifida aperta in rat fetuses.

Neural tube defects (NTDs) are the most common and severe congenital malformations, which result from failure of the neural tube to close during embryonic development. The etiology of NTDs is complex, caused by interactions between genetic defects and environmental factors, but the exact mechanisms of this disease are still not fully understood. We herein employ a Seahorse Bioscience microplate-based extracellular flux (XF) analyzer to determine mitochondrial function and quantify respiratory coupling to various bioenergetic functions using specific pharmacological inhibitors of bioenergetic p

Genomic approaches to the assessment of human spina bifida risk.

Structural birth defects are a leading cause of mortality and morbidity in children world-wide, affecting as much as 6% of all live births. Among these conditions, neural tube defects (NTDs), including spina bifida and anencephaly, arise from a combination of complex gene and environment interactions that are as yet poorly understood within human populations. Rapid advances in massively parallel DNA sequencing and bioinformatics allow for analyses of the entire genome beyond the 2% of the genomic sequence covering protein coding regions. Efforts to collect and analyze these large datasets hold

Noggin and Sonic hedgehog are involved in compensatory changes within the motoneuron-depleted mouse spinal cord.

Sonic hedgehog and Noggin are morphogenetic factors involved in neural induction and ventralization of the neural tube, but recent findings suggest that they could participate in regeneration and functional recovery after injury. Here, in order to verify if these mechanisms could occur in the spinal cord and involve synaptic plasticity, we measured the expression levels of Sonic hedgehog, Noggin, Choline Acetyltransferase, Synapsin-I and Glutamate receptor subunits (GluR1, GluR2, GluR4), in a motoneuron-depleted mouse spinal cord lesion model obtained by intramuscular injection of Cholera toxi

Ectopic expression of the immune adaptor protein CD3zeta in neural stem/progenitor cells disrupts cell-fate specification.

Immune signaling and neuroinflammatory mediators have recently emerged as influential variables that regulate neural precursor/stem cell (NPC) behavior and function. In this study, we investigated whether the signaling adaptor protein CD3ζ, a transmembrane protein involved in T cell differentiation and function and recently shown to regulate neuronal development in the central nervous system (CNS), may have a role in NPC differentiation. We analyzed the expression profile of CD3ζ in embryonic rat brain during neurogenic periods and in neurosphere-derived neural cells, and we investigated the a

Hangman's fracture: arguments in favor of surgical therapy for type II and III according to Edwards and Levine.

BACKGROUND: The optimal therapy for traumatic fractures of the neural arch of the axis, the so-called hangman's fractures, is still controversial. Indications for surgery depend on the type of hangman's fracture and/or additional injuries of the intervertebral disc or ligaments. METHODS: Sixteen patients sustaining fractures of the neural arch of the axis were treated by the screw fixation technique evolved by Judet. The passive mobility of the cervical spine was analyzed postoperatively by the radiographic evaluation method of Dvorak and coworkers and Penning. The angulation and anterior tra

Bayesian bilevel operator learning with low-rank adaptation for efficient uncertainty quantification of PDE inverse problems.

Uncertainty quantification in PDE inverse problems is essential in many applications. Scientific machine learning and AI enable data-driven learning of model components while preserving physical structure, and provide the scalability and adaptability needed for emerging imaging technologies and clinical insights. We develop a Bilevel Local Operator Learning framework for Bayesian inference in PDEs (B-BiLO). At the upper level, we sample parameters from the posterior via Hamiltonian Monte Carlo, while at the lower level we fine-tune a neural network via low-rank adaptation (LoRA) to approximate

Long-term Learning Induces Plastic Changes in Frontostriatal Circuits.

Neural activity in frontal-striatal circuits underlies reinforcement learning. Traditional theories suggest that reinforcement signals, which drive learning, strengthen connections within the basal ganglia. This strengthening is believed to shift information processing from cortical regions to subcortical regions as learning becomes established over time. To examine this hypothesis, we trained macaques to associate multiple sets of images with their values. Selecting different images led to either an increase (+2, +1) or a decrease (-1, -2) in the number of tokens, which subsequently determine
