Warmth suppresses and desensitizes damage-sensing ion channel TRPA1.

BACKGROUND: Acute or chronic tissue damage induces an inflammatory response accompanied by pain and alterations in local tissue temperature. Recent studies revealed that the transient receptor potential A1 (TRPA1) channel is activated by a wide variety of substances that are released following tissue damage to evoke nociception and neurogenic inflammation. Although the effects of a noxious range of cold temperatures on TRPA1 have been rigorously studied, it is not known how agonist-induced activation of TRPA1 is regulated by temperature over an innocuous range centred on the normal skin surfac…

Serum levels of substance P are elevated in patients with sickle cell disease and increase further during vaso-occlusive crisis.

As a mediator of neurogenic inflammation and pain, we hypothesized that levels of the neuropeptide Substance P (SP) would be elevated in patients with sickle cell disease (SCD) with vaso-occlusive pain crisis. SP is a known stimulator of tumor necrosis factor-alpha (TNF-alpha) release and a promoter of interleukin-8 (IL-8), which are reported to be increased in SCD. These cytokines enhance adhesion of leukocytes to endothelium and may play a role in vaso-occlusive events. Serum levels of IL-8, TNFalpha, and SP were studied in three groups of children aged 2 to 18 years: 30 well children with S…

Acute ethanol administration results in a protective cytokine and neuroinflammatory profile in traumatic brain injury.

Ethanol intoxication is a common comorbidity in traumatic brain injury. To date, the effect of ethanol on TBI pathogenic cascades and resulting outcomes remains debated. A closed blunt weight-drop murine TBI model has been implemented to investigate behavioral (by sensorimotor and neurological tests), and neuro-immunological (by tissue cytokine arrays and immuno-histology) effects of ethanol intoxication on TBI. The effect of the occurrence of traumatic intracerebral hemorrhage was also studied. The results indicate that ethanol pretreatment results in a faster and better recovery after TBI wi

Metformin as a potential therapeutic agent in broken heart syndrome: Targeting AMPK-dependent cardio-protection and microvascular function.

Broken heart syndrome (Takotsubo or stress cardiomyopathy) is a transient form of acute left ventricular dysfunction commonly triggered by emotional or physical stress. Despite its reversible nature, it may lead to severe complications such as pulmonary edema, arrhythmias, or cardiac arrest. Current management remains largely supportive, with no established pharmacological therapy. Metformin, a well-known anti-diabetic drug, has emerged as a potential cardioprotective agent due to its multifaceted actions on cellular metabolism, vascular function, and neuro-hormonal balance. By activating the

Targeting the neuro-immune axis: immunological mechanisms and therapeutic strategies in male sexual dysfunction.

Male sexual dysfunction (MSD) is a multifactorial condition in which traditional models focusing on vascular, endocrine, and psychogenic factors are insufficient. Emerging evidence highlights dysregulated crosstalk between the nervous and immune systems-the neuroimmune axis-as a potential unifying pathological substrate. This review synthesizes fragmented yet compelling evidence linking neural injury, chronic inflammation (both systemic and neurogenic), and immune dysregulation to the pathogenesis of MSD. We propose a novel integrative framework in which disruption of the neuro-immune axis ser

TRPV1 channels in skin homeostasis and disease: From sensory transduction to therapeutic targeting.

TRPV1 is a non-selective cation channel that is widely expressed in multiple skin cell types, including sensory nerve fibers, keratinocytes, immune cells and fibroblasts. It can be activated by noxious heat, acidic conditions, capsaicin and a range of endogenous inflammatory mediators. Upon activation, TRPV1 mediates Ca2+ influx, leading to neuropeptide release and the initiation of neurogenic inflammation. Under physiological conditions, in addition to its well-established roles in nociception and pruritus transmission, TRPV1 contributes to several aspects of cutaneous homeostasis, including …

Vasoactive properties of calcitonin gene-related peptide in human skin.

AIM: Calcitonin gene-related peptide (CGRP) is a potential mediator of neurogenic inflammation in eczema, psoriasis and rosacea, and also contributes to vasodilatation and oedema in complex regional pain disorder. We investigated the feasibility of administering CGRP and its antagonist CGRP8-37 by iontophoresis in human skin to characterise their vasoactive profiles. METHODS: Two doses of each peptide were administered by iontophoresis (5 and 10 min duration at 0.1 mA) to the forearm skin of 6 healthy young men. Skin blood flow responses over 25 min were assessed using laser Doppler imaging. …

Curcumin ameliorates reserpine-induced pain-depression dyad: behavioural, biochemical, neurochemical and molecular evidences.

An apparent clinical relationship between pain and depression has long been recognized. Depression and pain are often diagnosed in the same patients. The emerging concept for pain-depression pathogenesis is the dysfunction of biogenic amine-mediated pain-depression control and the possible involvement of nitrodative stress-induced neurogenic inflammation. The present study was designed to investigate the effect of curcumin on reserpine-induced pain-depression dyad in rats. Administration of reserpine (1mg/kg subcutaneous daily for three consecutive days) led to a significant decrease in nocice…

Effect of olopatadine hydrochloride, an anti-histamine drug, on rhinitis induced by intranasal instillation of toluene-2,4-diisocyanate in rats.

The main symptoms of allergic rhinitis (AR) are sneezing, rhinorrhea and nasal obstruction. In patients with AR, levels of nerve growth factor (NGF) and vascular endothelial growth factor (VEGF) increase. Olopatadine hydrochloride (olopatadine) is an anti-allergic agent with histamine H1 receptor antagonistic action. To investigate whether olopatadine has an effect on inflammatory reactions, toluene-2,4-diisocyanate (TDI)-sensitized rats were used as an animal model of nasal allergy. Nasal allergy signs (sneezing, rhinorrhea and inflammation) were induced after TDI challenge. Amounts of NGF an

Nitric oxide regulation of calcitonin gene-related peptide gene expression in rat trigeminal ganglia neurons.

Calcitonin gene-related peptide (CGRP) and nitric oxide are involved in the underlying pathophysiology of migraine and other diseases involving neurogenic inflammation. We have tested the hypothesis that nitric oxide might trigger signaling mechanisms within the trigeminal ganglia neurons that would coordinately stimulate CGRP synthesis and release. Treatment of primary trigeminal ganglia cultures with nitric oxide donors caused a greater than four-fold increase in CGRP release compared with unstimulated cultures. Similarly, CGRP promoter activity was also stimulated by nitric oxide donors and…
