Genes of cyanobacterial origin in plant nuclear genomes point to a heterocyst-forming plastid ancestor.

Plastids are descended from a cyanobacterial symbiosis which occurred over 1.2 billion years ago. During the course of endosymbiosis, most genes were lost from the cyanobacterium's genome and many were relocated to the host nucleus through endosymbiotic gene transfer (EGT). The issue of how many genes were acquired through EGT in different plant lineages is unresolved. Here, we report the genome-wide frequency of gene acquisitions from cyanobacteria in 4 photosynthetic eukaryotes--Arabidopsis, rice, Chlamydomonas, and the red alga Cyanidioschyzon--by comparison of the 83,138 proteins encoded i

Niche differentiation of bacterial communities at a millimeter scale in Shark Bay microbial mats.

Modern microbial mats can provide key insights into early Earth ecosystems, and Shark Bay, Australia, holds one of the best examples of these systems. Identifying the spatial distribution of microorganisms with mat depth facilitates a greater understanding of specific niches and potentially novel microbial interactions. High throughput sequencing coupled with elemental analyses and biogeochemical measurements of two distinct mat types (smooth and pustular) at a millimeter scale were undertaken in the present study. A total of 8,263,982 16S rRNA gene sequences were obtained, which were affiliat

Two residues in the T-loop of GlnK determine NifL-dependent nitrogen control of nif gene expression.

X-ray crystallographic analysis of the Escherichia coli P(II) protein paralogues GlnB and GlnK has shown that they share a superimposable structural core but can differ in conformation of the T-loop, a region of the protein (residues 37-54) that has been shown to be important for interaction with other proteins. In Klebsiella pneumoniae GlnK has been shown to have a clearly defined function in regulating NifL-mediated inhibition of NifA activity in response to the nitrogen status, and GlnB, when expressed from the chromosome, does not substitute for GlnK. Because the T-loops of K. pneumoniae a

Functional methylthio-alkane reductases are widely distributed for bacterial organic sulfur acquisition.

UNLABELLED: Methylthio-alkane reductase (MAR) is a relative of nitrogenase that cleaves methylthio-alkanes for methanethiol liberation and s … ia. Functional MAR enzymes are only observed in one phylogenetic group, Group IV-C, from the larger nitrogen fixation-like family of sequences, indicating that the remaining phylogenetic groups catalyze alternative and, in many cases, unknown reactions. Finally, either the bona fide NifB of R. rubrum, which synthesizes NifB-co, or the homologous MarB associated wit …

Response to Nutrient Stress in the Industrial Model Bacterium Cupriavidus necator: A Thermal Proteome Profiling (TPP) Investigation.

The facultative chemolithoautotroph Cupriavidus necator is capable of heterotrophic growth on diverse carbon sources, or of autotrophic growth using CO2 fixation with H2 as an energy source. Under stress conditions, it produces biodegradable polyesters (polyhydroxyalkanoates, PHAs) as a storage material occupying a high proportion of the total biomass. This metabolic versatility means that C. necator is under intense study for sustainable biotechnology processes; however, a relative lack of understanding of the overall regulatory architecture has limited its application. The major mechanisms b

Adaptable Immunofluorescence Protocol for Muscle Fiber Typing in FFPE Human and Mouse Skeletal Muscle and Intact Mouse Hindlimbs.

Skeletal muscle fiber type composition affects muscle function, metabolism, and disease vulnerability. In addition, muscle fiber type analysis informs disease diagnosis and underlying pathophysiology. Multiple methodologies can be used to assess muscle fiber type; however, immunofluorescence (IF) for myosin heavy chain (MyHC) isoforms is the most widely used modern approach due to its relative ease, time-effectiveness, single-cell resolution, and capacity to preserve spatial positioning within the native tissue architecture. Here, we present a protocol for IF for MyHC labeling on formalin-fixe

A genome-resolved cyanobacterial consortium from the Atacama Desert reveals novel bacterial diversity, stress-resilience potential, chromatic flexibility, and photoprotection.

The Atacama Desert is one of the most extreme terrestrial environments on Earth, characterized by hyper-aridity, intense solar and ultraviolet radiation, oxidative stress, and metal-rich substrates. Here, we characterized an enriched cyanobacteria-associated consortium obtained from a high-altitude, river-influenced site in Quebrada Talabre, Atacama Desert. Deep shotgun metagenomic sequencing, achieving 99.7% estimated community coverage, enabled the reconstruction of 14 high-quality metagenome-assembled genomes representing five bacterial phyla. Genome-resolved analyses revealed multiple puta

Role of phosphate solubilizing Burkholderia spp. for successful colonization and growth promotion of Lycopodium cernuum L. (Lycopodiaceae) in lateritic belt of Birbhum district of West Bengal, India.

Profuse growth of Lycpodium cernuum L. was found in phosphate deficient red lateritic soil of West Bengal, India. Interaction of vesicular-arbuscular mycorrhiza (VAM) with Lycopodium rhizoids were described earlier but association of PGPR with their rhizoids were not studied. Three potent phosphate solubilizing bacterial strains (P4, P9 and P10) associated with L. cernuum rhizoids were isolated and identified by 16S rDNA homologies on Ez-Taxon database as Burkholderia tropica, Burkholderia unamae and Burkholderia cepacia respectively. Nitrate regulates rhizobial and mycorrhizal symbiosis in common bean (Phaseolus vulgaris L.).

Nitrogen-limited conditions are considered to be a prerequisite for legume-rhizobial symbiosis, but the effects of nitrate-rich conditions on symbiotic status remain poorly understood. We addressed this issue by examining rhizobial (Rhizobim tropici) and arbusclar mycorrhizal (Glomus intraradices) symbiosis in Phaseolus vulgaris L. cv. Negro Jamapa under nitrate pre-incubation and continuous nitrate conditions. Our results indicate that nitrate pre-incubation, independent of the concentration, did not affect nodule development. However, the continuous supply of nitrate at high concentrations i

Dinitrogen Activation and Functionalization Affording Chromium Diazenido and Hydrazido Complexes.

ConspectusThe activation and functionalization of N2 to form nitrogen-element bonds have long posed challenges to industrial, biological, and synthetic chemists. The first transition-metal dinitrogen complex prepared by Allen and Senoff in 1965 provoked researchers to explore homogeneous N2 fixation. Despite intensive research in the last six decades, efficient and quantitative conversion of N2 to diazenido and hydrazido species remains problematic. Relative to a plethora of reactions to generate N2 complexes, their functionalization reactions are rather rare, and the yields are often unsatisf

Electron microscopic analysis of the mouse and human alveolar epithelial glycocalyx using high-pressure freezing and freeze substitution.

Alveolar epithelial cells (AECs) are covered apically by the alveolar lining layer (ALL). The alveolar epithelial glycocalyx (AEGCX), which is part of the ALL, consists of a mixture of glycoproteins, proteoglycans, and glycosaminoglycans. In comparison with the endothelial glycocalyx (GCX), much less is known about the composition and structure of the AEGCX, although it is presumed to be involved in many cellular processes. The challenges of in vivo visualization of the AEGCX, coupled with the distorting effects of aldehyde fixation, make it difficult to accurately determine its structural org
