PURPOSE: A woman's skull, exhibiting features of lepromatous leprosy (LL), was recovered from a garden in Hoxne, Suffolk. The absence of post crania and lack of formal excavation meant that diagnosis and dating was uncertain. The aim of this research was to confirm the diagnosis using biomolecular means and second, to place it in context with other British leprosy cases using SNP genotyping and radiocarbon dating.
METHODOLOGY: Bone from the skull was analysed by ancient DNA (aDNA) methods and subjected to radiocarbon dating. As a result, stable carbon and nitrogen isotope values were produced

Purge and trap sample introduction (PTI) has been the premier sampling and preconcentration technique for gas chromatographic determination of volatile organic compounds (VOCs) in drinking water for almost 50 years. PTI affords sub parts-per-billion (ppb) detection limits for purgeable VOCs including fixed gases and higher boiling hydrocarbons and halocarbons. In this study the coupling of solid phase extraction (SPE) to PTI was investigated as a means to substantially increase enrichment and lower detection limits for the emerging contaminant, 1,2,3-trichloropropane (TCP). Water samples (500

The oxidation and nitration of unsaturated fatty acids transforms cell membrane and lipoprotein constituents into mediators that regulate signal transduction. The formation of 9-NO2-octadeca-9,11-dienoic acid and 12-NO2-octadeca-9,11-dienoic acid stems from peroxynitrite- and myeloperoxidase-derived nitrogen dioxide reactions as well as secondary to nitrite disproportionation under the acidic conditions of digestion. Broad anti-inflammatory and tissue-protective responses are mediated by nitro-fatty acids. It is now shown that electrophilic fatty acid nitroalkenes are present in the urine of h

Sodium phenylbutyrate is used in the pharmacological treatment of urea cycle disorders to create alternative pathways for nitrogen excretion. The primary metabolite, phenylacetate, conjugates glutamine in the liver and kidney to form phenylacetylglutamine that is readily excreted in the urine. Patients with urea cycle disorders taking sodium phenylbutyrate have a selective reduction in the plasma concentrations of branched chain amino acids despite adequate dietary protein intake. Moreover, this depletion is usually the harbinger of a metabolic crisis. Plasma branched chain amino acids and oth

We have carried out kinetics studies of spinach carbonic anhydrase (CA) using stopped-flow spectrophotometry at steady state and 13C-NMR exchange at chemical equilibrium. We found that the rate of CO2<-->HCO3- exchange catalyzed by spinach CA at pH 7.0 to be 3-5 times faster than the maximal kcat for either CO2 hydration or HCO3- dehydration at steady state, suggesting a rate-determining H+ transfer step in the catalytic mechanism. Correspondingly, we measured a pH-independent solvent deuterium isotope effect on kcat of approximately 2.0, and found that the rate of catalysis was significantly

Ethylbenzene and toluene degradation under nitrate-, Mn(IV)-, or Fe(III)-reducing conditions was investigated by compound specific stable isotope analysis (CSIA) using three model cultures (Aromatoleum aromaticum EbN1, Georgfuchsia toluolica G5G6, and a Azoarcus-dominated mixed culture). Systematically lower isotope enrichment factors for carbon and hydrogen were observed for particulate Mn(IV). The increasing diffusion distances of toluene or ethylbenzene to the solid Mn(IV) most likely caused limited bioavailability and hence resulted in the observed masking effect. The data suggests further

Urea kinetics were measured on two separate occasions in five adults with normal haemoglobin genotype (HbAA) and in four who were homozygous for sickle cell disease (HbSS). Prime/intermittent doses of [15N15N]urea were given orally on one occasion and intravenously on the other. In three of the nine individuals there appeared to be significant hydrolysis of the oral dose of urea before absorption, leading to spurious results for the urea kinetics. When only the studies in which isotope was given intravenously were considered, there was a difference in the rate at which urea-N was salvaged, wit

We have constructed a strain of Azotobacter vinelandii which has deletions in the genes for both the molybdenum (Mo) and vanadium (V) nitrogenases. This strain fixed nitrogen in medium that did not contain Mo or V. Growth and nitrogenase activity were inhibited by Mo and V. In highly purified medium, growth was limited by iron. Addition of other metals (Co, Cr, Cu, Mn, Ni, Re, Ti, W, and Zn) did not stimulate growth. Like the V-nitrogenase, the nitrogenase synthesized by the double deletion strain reduced acetylene to both ethylene and ethane (C2H6/C2H4 ratio, 0.046). There was an approximatel

Electrochemical nitrate reduction represents a sustainable route for ammonia synthesis, yet its advancement is hampered by two bottlenecks: the fundamental limitation on conversion efficiency stems from inadequate in situ nitrate enrichment and the elusive dynamic evolution of catalytic mechanisms under operational conditions. Here, we report a CuO hollow multishelled structure (CuO-HoMS) that functions as a spontaneous pre-enrichment micronanoreactor capable of enriching, confining, and converting nitrate with exceptional efficiency. The CuO-HoMS with triple shells (3s-CuO-HoMS) achieves a ch

Tris(indol-3-yl)methanes were synthesized from indoles, CO2, and PhMe2SiH in the presence of BPh3 in acetonitrile at 50°C. This is the first CO2 fixation reaction forming the aliphatic methine (>CH─) group. The isotope-labeling experiments clearly demonstrated that the carbon and hydrogen atoms of the methine group originated from carbon dioxide and dimethylphenylsilane, respectively. The mechanism for the multicomponent cascade reaction was elucidated by DFT calculations. One of the CO2-derived products, tris(4-bromo-1-methylindol-3-yl)methane, underwent palladium-catalyzed intramolecular cas

Cold-water coral reefs host diverse microbial communities that play key roles in nutrient cycling and deep-sea productivity. Among them, methylotrophic bacteria metabolize and cycle one-carbon (C1) compounds such as methane, methanol, methylated nitrogen and sulphur, linking food webs and having a central role in the global cycling of climate-active trace gases. To date, few methylotrophic bacteria have been cultured from cold-water coral reefs. In this study, we enriched methylotrophs from seawater, sediment and sponge samples collected in cold-water coral reefs off mid-Norway. Cultivation at

Water column denitrification, a major oceanic nitrogen (N) sink, decreased under past warmer-than-present climates, but the response of N2 fixation, the main oceanic N source, remains unresolved. Here we address this gap using foraminifera-bound N isotope records from the tropical Atlantic spanning the warm late Pliocene, a geological analog for projected 21st century warming. Our records show that tropical North Atlantic N2 fixation was reduced during the late Pliocene and had increased markedly by the late Pleistocene, likely due to an increase in water column denitrification, a process that

The bacterial species "Candidatus Alkanivorans nitratireducens" was recently demonstrated to mediate nitrate-dependent anaerobic oxidation of short-chain gaseous alkanes (SCGAs). In previous bioreactor enrichment studies, the species appeared to reduce nitrate in two phases, switching from denitrification to dissimilatory nitrate reduction to ammonium (DNRA) in response to nitrite accumulation. The regulation of this switch or the nature of potential syntrophic partnerships with other microorganisms remains unclear. Here, we describe anaerobic multispecies cultures of bacteria that couple the

Mercury concentrations in the Laurentian Great Lakes waters are among the lowest reported in the literature, while game fish concentrations approach consumption advisory limits, particularly in Lakes Superior, Huron, and Michigan, indicating efficient methylmercury transfer from water to game fish. To determine if increased transfer efficiency is evident within the lower food web, we measured (2010-2018) mercury and dissolved organic carbon (DOC) in water, and in size-sieved seston, dietary tracers (carbon and nitrogen isotope ratios), phytoplankton methylmercury bioaccumulation, and methylmer

Mercury (Hg) is a contaminant that is impacting ecosystems worldwide. Its toxicity is threatening wildlife and human populations, leading to the necessity of identifying the most affected ecosystems. Therefore, it is essential to identify pertinent bioindicator organisms to monitor Hg contamination. In this study, we determined the stable carbon (δ13C) and nitrogen (δ15N) isotope ratios in the red blood cells (RBCs), and the total Hg concentration in total blood of 72 Melanosuchus niger in French Guiana. The goals of our study were to assess the level of Hg contamination in total blood of Blac

Macroinvertebrates are a key vector in the transfer of methylmercury (MeHg) to fish. However, the factors that affect MeHg concentrations and bioaccumulation in these organisms are not as well understood as for fish, and studies on a broad geographic scale are lacking. In this study, we gathered published and unpublished MeHg and carbon (δ(13)C) and nitrogen (δ(15)N) stable isotope data for freshwater macroinvertebrates from 119 lakes and wetlands across seven Canadian provinces, along with selected physical, chemical and biological characteristics of these systems. Overall, water pH was the m

Despite the global prevalence of both mercury (Hg) contamination and anthropogenic eutrophication, relatively little is known about the behavior of Hg in eutrophic and hypereutrophic systems or the effects of lake trophic status on Hg uptake and trophodynamics. In the current study we explore Hg trophodynamics at 8 tropical East African study sites ranging from mesotrophic to hypereutrophic, in order to assess the influence of lake trophic status on Hg uptake and biomagnification. Comprehensive water, plankton and fish samples were collected for analysis of total mercury (THg) and stable carbo

Mercury (Hg) and the composition of organic matter (OM) in sediments and large zooplankton (> 190 microm) have been analysed in a perturbed lake (Lake 154) in northern Quebec, Canada. We investigated whether methylmercury concentrations ([MeHg]) in flooded sediments can predict [MeHg] in zooplankton. [MeHg] at the uppermost sediment layers located immediately downstream of an inundated system (LA-40) are 10 times higher than at other sediment stations. Zooplankton organisms sampled downstream of LA-40 contain higher [MeHg] (183+/-50 ng g(-1) dry wt.) than those of neighbouring natural lakes (6

Geologic and climatologic studies suggest that conditions on early Mars were similar to early Earth. Because life on Earth is believed to have originated during this early period (3.5 billion years ago), the Martian environment could have also been conducive to the origin of life. To investigate this possibility we must first define the attributes of an early Martian biota. Then, specific geographic locations on Mars must be chosen where life may have occurred (i.e. areas which had long standing water), and within these distinct locations search for key signatures or bio-markers of a possible
