Effects of Saikosaponin A on post-traumatic stress disorder-like behaviors, hippocampal monoaminergic neurotransmission, and the NF-κB inflammatory pathway in a rat model.

Post-traumatic stress disorder (PTSD) is a debilitating psychiatric condition with limited effective treatments. Saikosaponin A (SSa), a bioactive triterpenoid from Bupleurum chinense, exhibits neuroprotective and anti-inflammatory properties, yet its therapeutic potential for PTSD remains unexplored. This study investigated whether SSa ameliorates PTSD-like behaviors in rats and examined the underlying mechanisms. A PTSD rat model was established using single prolonged stress (SPS) combined with footshock(FS). Rats were randomly assigned to control, model, SSa treatment, and fluoxetine-positi

Saikosaponin-d attenuated lipopolysaccharide-induced depressive-like behaviors via inhibiting microglia activation and neuroinflammation.

Saikosaponin-d (SSd), a triterpenoid saponins compound extracted from Radix Bupleuri, has been demonstrated to effectively alleviate chronic mild stress-induced depressive behaviors in rats, but the underlying molecular mechanisms are still uncertain. Increasing evidence indicated that microglia activation and inflammatory responses were involved in the pathogenesis of depression. Thus, we desired to induce inflammation-related depressive-like behaviors in mice by injecting lipopolysaccharide (LPS) to investigate whether the antidepressant effect of SSd is related to inhibiting inflammation. The clickable activity-based probe of anti-apoptotic calenduloside E.

CONTEXT: Calenduloside E (CE), one of the primary natural products found in Aralia elata (Miq.) Seem. (Araliaceae), possesses prominent anti-apoptotic potential. A previous study found that one of the anti-apoptotic CE targets is heat shock protein 90 AB1 (Hsp90AB1) by probe CE-P, while the other targets of CE still need to be identified with more efficient probes. OBJECTIVE: This study investigates CE analogue (CEA) as one clickable activity-based probe for use in exploring anti-apoptotic CE targets. MATERIALS AND METHODS: Pretreatment of HUVECs with CEA (1.25 μM) for 8 hr, followed by ox-L

Triterpene derivative improves the renal function of streptozotocin-induced diabetic rats: a follow-up study on maslinic acid.

Introduction: Reports indicate that oral administration of plant-derived maslinic acid (MA) exhibits hypoglycemic and renoprotective effects in streptozotocin (STZ)-induced diabetic rats. Challenges with triterpenes such as MA include low bioavailabilty which affects treatment efficacy in experimental animals. The goal of this study was to synthesize the MA derivative phenylhydrazine (PH-MA) in an effort to improve the efficacy of MA. Methods: Separate groups of non-diabetic and STZ-induced diabetic rats (n = 6) were anesthetized and the jugular vein cannulated for the infusion of 0.077 M NaCl

Mechanisms contributing to adverse cardiovascular events in patients with type 2 diabetes mellitus and stage 4 chronic kidney disease treated with bardoxolone methyl.

BACKGROUND: Bardoxolone methyl, an Nrf2-activating and nuclear factor-κB-inhibiting semisynthetic oleanane triterpenoid compound, was evaluated in a phase 3 trial (BEACON) in patients with type 2 diabetes mellitus (T2DM) and stage 4 chronic kidney disease (CKD). The trial was terminated because of an increase in heart failure events in the bardoxolone methyl group, many of which appeared related to fluid retention. Thus, additional analyses were conducted to explain these serious adverse events. METHODS: Patients (n = 2,185) were randomized to receive once-daily bardoxolone methyl (20 mg) or

CDDO-Me protects against space radiation-induced transformation of human colon epithelial cells.

Radiation-induced carcinogenesis is a major concern both for astronauts on long-term space missions and for cancer patients being treated with therapeutic radiation. Exposure to radiation induces oxidative stress and chronic inflammation, which are critical initiators and promoters of carcinogenesis. Many studies have demonstrated that non-steroidal anti-inflammatory drugs and antioxidants can reduce the risk of radiation-induced cancer. In this study, we found that a synthetic triterpenoid, CDDO-Me (bardoxolone methyl), was able to protect human colon epithelial cells (HCECs) against radiatio

In vitro and in silico evaluation of anti-inflammatory triterpene saponins from Elsholtzia penduliflora W.W. Smith (Lamiaceae).

Elsholtzia penduliflora W.W. Smith (Lamiaceae), a medicinal herb traditionally used in Vietnam, was investigated for its anti-inflammatory potential. Phytochemical analysis of its aerial parts, extracted using 80% ethanol, yielded 15 oleanane-type saponins (1-15) using chromatographic techniques. Among these, penduloside E (4), penduloside C (5), and kajiichigoside F1 (10) were prioritised for bioactivity testing based on structural characteristics and preliminary anti-inflammatory screening. These compounds at a concentration of 3 µM reduced PGE2 production by 48.9-54.3% and downregulated COX

Bardoxolone Derivatives as Novel Pseudo-Natural Necroptosis Inhibitors by Destabilizing HSP90 Client Proteins.

Targeting necroptosis has been confirmed as an efficient treatment strategy for inflammatory diseases. 2-Cyano-3,12-dioxo-olean-1,9-diene-28-carboxylic acid (CDDO) was previously identified as a pseudonatural-product necroptosis inhibitor. However, CDDO was inactive in murine cells and less active in human cells. In this study, 27 derivatives of CDDO were synthesized by structural modification in A and D/E rings, among which ZYH-23 had the best activity. It could effectively block necroptosis in both human and murine cells and soon alleviate SIRS-induced hypothermia and death by remarkably dec

Bredemolic Acid Ameliorates Selected Liver Function Biomarkers in a Diet-Induced Prediabetic Rat Model.

BACKGROUND: Prediabetes is an intermediary hyperglycaemic state that precedes type 2 diabetes mellitus (T2DM) in which abnormal metabolism of glucose and lipids occurs in organs such as the liver. Evidence has shown that, about 70% of T2DM patients develop hepatic dysfunction which is found to begin during the prediabetic stage. Bredemolic acid, a pentacyclic triterpene, has been found to improve insulin sensitivity in diet-induced prediabetic rats. The effects of this compound on liver function, however, are unknown. This study was therefore designed to investigate the effects of BA on liver

Targeting the Keap1-Nrf2 Axis in COPD: Comparative analysis of electrophilic and peptide-based Nrf2 activators in airway and immune cells.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) is characterized by sustained oxidative stress, inflammation, and epithelial damage. The transcription factor Nrf2 is a master regulator of antioxidant and cytoprotective defenses, and its dysregulation has been implicated in COPD pathogenesis. METHODS: We first investigated Nrf2 expression and its downstream antioxidant genes in lung tissue and neutrophils from healthy donors and COPD patients, and examined their association with disease severity (GOLD stage). We then compared the efficacy of two mechanistically distinct Nrf2 activator

Discovery of New CDDO-Imidazole Derivatives as Potential Antitumor Agents.

In the search for new CDDOs (2-cyano-3,12-dioxooleana-1,9(11)-dien-28-oic acid derivatives) with higher antitumor activity in vivo, thirty CDDO-imidazole derivatives (1-30) were designed and synthesized. Then, 8 was selected due to its superior anti-proliferative activity against three cancer cell lines (B16F10, A549, and HCT116) and its lower toxicity in zebrafish embryos compared to the other evaluated compounds. Further study found that 8 induced apoptosis in HCT116 cells by downregulating Bcl-2, upregulating Bax, and activating caspase-3 to kill cancer cells. Notably, 8 exhibited significa

Design, synthesis, and biological evaluation of C-28 carboxyl derivatives of Hederacoside C as anti-inflammatory agents for acute lung injury.

Hederacoside C, a bioactive triterpenoid saponin isolated from the dried roots of the Ranunculaceae plant Pulsatilla chinensis, has demonstrated therapeutic efficacy in murine models of nephritis, acute lung injury, and inflammatory bowel disease. However, its further development as a drug candidate has been hampered by hemolytic toxicity and poor oral bioavailability. Based on published evidence that the free carboxyl group at the C-28 position is closely associated with hemolytic activity, a series of C-28 carboxyl derivatives was rationally designed and synthesized with the aim of attenuati
