Dynamic feedback control of oncogenic tyrosine kinase signaling in acute leukemia.

CD25 is a subunit of the interleukin-2 (IL-2) receptor on T cells and natural killer (NK) cells. Acute leukemias with oncogenic tyrosine kinases often include CD25+ leukemia subpopulations, which portend poor clinical outcomes for patients; however, acute leukemia cells do not respond to IL-2. Here, we identified CD25 and its phosphorylation by protein kinase Cδ (PKCδ) as central elements of a feedback loop that stabilized fluctuations in oncogenic tyrosine kinase signaling in acute lymphoblastic and myeloid leukemia. Genetic ablation of CD25 in murine and patient-derived xenograft (PDX) model

CRISPR-mediated Correction of Oncogenic AS-NMD in Splicing Factor Mutant Cancer.

Alternative splicing coupled to nonsense-mediated mRNA decay (AS-NMD) evolved as a master regulator of gene expression. Dysregulated AS-NMD has been identified as the root of many human maladies, from developmental defects to deadly cancer. Poison exons (PEs) are highly conserved alternative exons that contain a premature termination codon and elicit AS-NMD when included in a transcript. Cancer cells often exploit the inclusion of PEs to downregulate tumor suppressors or the exclusion of PEs to upregulate oncoproteins. Therefore, PEs have drawn significant attention as a novel therapeutic aven

Oncogenic Role of BCL6 Rearrangement: A Focus on Follicular and Marginal Zone Lymphomas.

The BCL6 gene is a transcriptional regulator essential for germinal center B-cell function, modulating their activation, survival, cell cycle, and differentiation into plasma or memory B-cells. In indolent B-cell lymphomas, particularly follicular lymphoma and a subset of marginal zone lymphomas, BCL6 rearrangements are increasingly recognized as recurrent but heterogeneous events. In follicular lymphoma, their clinicopathologic significance differs according to the BCL2 status. While BCL6 rearrangements have limited impact in conventional BCL2-rearranged follicular lymphoma, BCL2-negative/BCL

Choline and DNA methylation: impact across metabolic, oncogenic and neurodevelopmental disorders.

BACKGROUND: Aberrant DNA methylation patterns are increasingly recognized as contributors to a wide range of conditions, including metabolic … outcomes. In hepatocellular carcinomas, choline modulates methylation of tumor suppressor genes and oncogenes, interacting with one-carbon metabolism enzymes and the Wnt/β-catenin pathway. During development, gestational choline supplementation induces long-lasting epigenetic changes across multiple systems, affecting genes involved in stress regulation, ne …

Virulence of Marek's disease virus in Japan is linked to polymorphisms in the meq oncogene.

Marek's disease virus (MDV) causes lymphomas and neurological disorders in chickens. Although vaccination largely controls outbreaks, highly virulent strains continue to emerge. The major MDV-encoded oncoprotein is Meq, functions as a transcription factor. Amino acid polymorphisms in Meq have been reported to influence virulence. Despite routine vaccination, MD still occurs sporadically in Japan. Japanese isolates harbour characteristic Meq polymorphisms, but their impact on MDV virulence remains unclear. We investigated the transcriptional regulation by Meq from Japanese isolates and evaluate

B-MYB (MYBL2): from cell cycle regulator to an oncogenic player.

B-MYB (MYBL2) is a transcription factor of the MYB family that plays critical roles in cell cycle progression, proliferation, and survival. Through the DREAM-MMB-FOXM1 network, B-MYB coordinates the expression of genes required for mitosis and cytokinesis (G2/M genes), while genes required for DNA replication during S phase are regulated by E2F-DP complexes. Initially identified as a regulator of normal cell cycle processes, B-MYB has emerged as a key oncogenic driver across multiple cancer types. This review addresses the physiological roles of B-MYB, the mechanisms underlying its oncogenic a

Targeting MYC-Driven Cancers: From Oncogenic Addiction to Therapeutic Vulnerability.

The MYC family oncoproteins, including MYC, MYCN, and MYCL, are potent drivers of tumorigenesis across a broad range of human cancers, frequently linked to aggressive tumor behavior, poor prognosis, and therapy resistance. They function as master transcriptional regulators that orchestrate gene expression programs governing nearly all aspects of tumor development. Despite their pivotal oncogenic role, they have long been considered "undruggable" due to their intrinsically disordered structure, lack of enzymatic activity, and the difficulty of targeting their protein-DNA and protein-protein int

Endogenous retroviruses in aging and cancer: from genomic defense to oncogenic activation.

Nearly half of the human genome consists of transposable elements, among which endogenous retroviruses, remnants of ancient retroviral infections, represent some of the most evolutionarily intriguing due to their paradoxical functional duality. While research has documented functional ERV exaptation in key biological processes, these elements have also been associated with age-related diseases, particularly cancer. This apparent contradiction presents an evolutionary question: why would potentially disruptive elements persist in genomes over evolutionary time? Here we review the complex relati

The oncogenic role of miR-155 on oral cancer progression and treatment strategies.

Oral squamous cell carcinoma (OSCC) is a prevalent and aggressive malignancy within head and neck cancers, often associated with risk factors such as tobacco use, alcohol consumption, and HPV infection. MicroRNAs (miRNAs), small non-coding RNAs that inhibit target genes, are a promising new focus as biomarkers or therapeutic targets for cancer treatment. miR-155 has emerged as a significant regulator in various cancers, including OSCC, where its altered expression is linked to tumor initiation, progression, and resistance to treatment. miR-155 is a well-studied miRNA that has an oncogenic role

ALG3 as a PanCancer Oncogene: Bioinformatics Analysis and Identification of Small-Molecule Inhibitors.

INTRODUCTION: Glycosylation plays a crucial role in cellular processes such as recognition and signaling, and its dysregulation is associated with tumor progression. Alpha-1,3-mannosyltransferase (ALG3) is a key enzyme in N-glycosylation, and its aberrant expression has been implicated in various malignancies. However, the mechanisms underlying ALG3-driven oncogenesis and the identification of potential ALG3 inhibitors remain largely unexplored. This study aims to comprehensively investigate the oncogenic role of ALG3 across different cancer types and identify potential inhibitors through bioi…

Inhibition of GSK3 and TSC2 Mediates the Oncogenic Activity of AKT in Hepatocellular Carcinoma.

UNLABELLED: Aberrant activation of AKT is a key oncogenic driver in hepatocellular carcinoma (HCC). As AKT activates multiple downstream signaling pathways, the key mechanisms mediating AKT-driven tumorigenesis must be elucidated to develop optimal treatment strategies. Using an Akt/NRas-induced HCC mouse model, we found that AKT promotes tumorigenesis by targeting tuberous sclerosis complex 2 (TSC2) and glycogen synthase kinase 3 α/β (GSK3α/β) rather than forkhead box O protein family members. Loss of either TSC2, leading to mTORC1 activation, or both GSK3 isoforms cooperated with activated N

HPV42 - a human papillomavirus classified as a low-risk type with oncogenic potential.

Human papillomavirus (HPV) type 42 is a rare HPV type currently classified as "low-risk" and belongs to the alpha genus. It has primarily been detected in benign vulvar papillomas and is predominantly associated with benign lesions such as anogenital warts. However, in very rare cases, HPV42 has also been identified in invasive genital tumors, particularly cervical carcinomas. In the case series presented here, HPV42 was detected in anogenital warts, a digital squamous cell carcinoma in situ, and a lymphogenously metastasized digital papillary adenocarcinoma (DPA). In the latter tumor entity,

[Human papillomaviruses : origin, oncogenic factors and markers for cervical cancer screening].

The Human papillomaviruses (HPV) have existed in the human population since the archaic hominids. Over the course of human migration and evolution, HPVs have co-evolved with humans on all continents to become today the leading cause of cervical cancer. HPVs are classified by genera, species, genotype, lineage, sub-lineage and variants. Among more than 200 HPV genotypes, HPV16 is the most common and the most oncogenic at high-risk (HPV-HR). If viral oncogenesis is governed by numerous factors and mechanisms involving the virus and its host, the E6/E7 oncogenic proteins of HR-HPV play a central …

Unraveling the role of γδ T cells in the pathogenesis of an oncogenic avian herpesvirus.

Marek's disease virus (MDV) is an oncogenic alphaherpesvirus that causes deadly lymphomas in chickens. In chickens, up to 50% of all peripheral T cells are gamma delta (γδ) T cells. Until now, their role in MDV pathogenesis and tumor formation remains poorly understood. To investigate the role of γδ T cells in MDV pathogenesis, we infected recently generated γδ T cell knockout chickens with very virulent MDV. Strikingly, disease and tumor incidence were highly increased in the absence of γδ T cells, indicating that γδ T cells play an important role in the immune response against MDV. In the ab

Prospects and Challenges in Developing mRNA Vaccines for Infectious Diseases and Oncogenic Viruses.

mRNA vaccines have emerged as an optimistic technological platform for vaccine innovation in this new scientific era. mRNA vaccines have dramatically altered the domain of vaccinology by offering a versatile and rapid approach to combating infectious diseases and virus-induced cancers. Clinical trials have demonstrated efficacy rates of 94-95% in preventing COVID-19, and mRNA vaccines have been increasingly recognized as a powerful vaccine platform. Although mRNA vaccines have played an essential role in the COVID-19 pandemic, they still have several limitations; their instability and degradat

A Tetravalent Bispecific Antibody Selectively Inhibits Diverse FGFR3 Oncogenic Variants.

The receptor tyrosine kinase FGFR3 is frequently mutated in bladder cancer and is a validated therapeutic target. Although pan-FGFR tyrosine kinase inhibitors (TKI) have shown clinical efficacy, toxicity and acquired resistance limit the benefit of these agents. While antibody-based therapeutics can offer superior selectivity than TKIs, conventional ligand-blocking antibodies are usually ineffective inhibitors of constitutively active receptor tyrosine kinases. Furthermore, the existence of multiple oncogenic variants of FGFR3 presents an additional challenge for antibody-mediated blockade. He
