Recent advances in malaria parasite cultivation and their application to studies on host-parasite relationships: a review.

The continuous cultivation of the erythrocytic stages of Plasmodium falciparum was achieved in 1976 and techniques have now also been develo … e have been no significant advances in the in vitro cultivation of the sporogonic stages of malaria parasites for the last 15 years, although studies indicate that the in vitro cultivation of these stages from gamete to sporozoite stage is theoretically possible. The application of cultivation techniques to the study of parasite epidemiology is discussed. To …

Emerging antimalarial drug targets to combat drug-resistant malaria parasites.

The emergence and rapid spread of multidrug resistance in Plasmodium parasites, particularly in the Greater Mekong region and increasingly in Sub-Saharan Africa. Resistance has led to an urgent need to develop novel anti-malaria therapies with an effective mechanism. Several non-classical targets, including Na++ pump PfATP4 (Plasmodium falciparum ATPase 4), PI4K (Phosphatidylinositol 4-kinase) and PfNCR1(Plasmodium falciparum Niemann-Pick Type C1-Related protein), are highly active against the asexual stage of Plasmodium. We summarise the progress achieved on key emerging targets over past dec…

Parasites of fish and other aquatic organisms in the Anthropocene.

The Anthropocene is the most recent period in Earth's history and is characterized by humankind-driven alterations to all ecosystems. Climate change, pollution, overfishing, invasive species and habitat degradation continue to reshape aquatic ecosystems, and like all living organisms in our changing world, parasites are likely to be affected at different levels. This Collection features research and review papers on some of these topics: parasites co-introduced with their fish hosts illustrate that biological invasions must be studied case-by-case, as some fish hosts lose their parasites upon …

Functional and protein interaction analysis of Nop7 in trypanosomatid parasites.

Nucleolar protein 7 (Nop7), also known as Pescadillo or PES1, is a conserved factor required for ribosome biogenesis, cell proliferation, and nucleolar organization in opisthokonts; however, its function in early-branching eukaryotes remains poorly understood. Here, we characterize Nop7 in the kinetoplastid parasites Trypanosoma brucei and Leishmania major, organisms that possess atypical ribosomes containing extensively fragmented 28S-type ribosomal RNA. Structural in silico analyses revealed lineage-specific features in trypanosomatid Nop7 proteins that may support specialized protein-protei…

Assessment of zoonotic parasites in sand sports courts in Brazil.

The popularity of sand sports has increased significantly, driven by their recreational appeal combined with health benefits. However, because these sports are typically played barefoot in areas where dogs and cats may roam freely, the risk of human exposure to zoonotic parasites-such as the causative agents of cutaneous and visceral larva migrans (Ancylostoma caninum, A. braziliense, and Strongyloides stercoralis)-increases. This study aimed to investigate the presence of potentially zoonotic helminth larvae in the sand of public sports courts in the municipality of Ilhéus, Bahia, Brazil, a c…

Inventory of helminth parasites in Caatinga anurans from Northeastern Brazil.

Biological inventories are essential tools for understanding parasite diversity. In this study, we describe the helminth parasite communities associated with anuran amphibians from five localities within the Caatinga domain of Northeastern Brazil. A total of 1,198 individuals, belonging to 22 species and six anuran families (Bufonidae, Hylidae, Leptodactylidae, Microhylidae, Odontoprhynidae, and Phyllomedusidae), were examined. Of these, 629 specimens (52.5%) were parasitized, and 27,004 helminths were recovered. The helminths represented 34 taxa, including 24 Nematoda, 7 Digenea, 1 Cestoda, a

Avian haemosporidian parasites affecting non-descript village chickens in Africa.

Smallholder farmers in most of the rural areas in African countries rear non-descript village chickens for petty cash, food provision and for performing rituals. Village chicken production systems are regarded as low input- low output because the chickens receive minimum care and produce average to less eggs and meat. The chickens receive minimal biosecurity and are often left to scavenge for feed and thus exposes them to potential vector parasites that can transmit parasites such as haemoparasites. Haemosporidian parasites (Haemosporidia, Apicomplexa) are blood parasites infecting avian speci…

Mosquito immune responses to Plasmodium parasites that limit malaria transmission.

The mosquito immune system is a major barrier that malaria parasites must overcome for their successful development and disease transmission. At each developmental stage in the vector, Plasmodium parasites can be potentially targeted by the mosquito innate immune responses, which involve epithelial, humoral, and cellular components. The immune response to Plasmodium ookinetes can be powerful and some of the underlying effector mechanisms are well characterized. However, the defense responses to oocysts and sporozoites appear to be less effective and are less well understood. Plasmodium parasit…

Alba protein-mediated gene and protein regulation in protozoan parasites.

The success of protozoan parasites relies heavily on regulation of gene and protein expression to facilitate their persistence in harsh and often changing environments. These parasites display biology that is highly divergent from model eukaryotes, unfortunately leaving our understanding of these parasites' critical regulatory mechanisms incomplete. Alba proteins, a highly diverse group of DNA/RNA-binding proteins, are found across all domains of life and it has become increasingly apparent that these proteins play key regulatory roles in many protozoan parasite species including Plasmodium, L…

Molecular Survey of Hemosporidian Parasites in Owls in Mainland Portugal.

Owls play an important role as avian hosts for hemosporidians, overlapping habitat use and activity with the arthropod vectors, which results in susceptibility to morbidity and, in some cases, mortality upon infection. This study aimed to analyze the hemosporidians in tissues collected from individual dead owls (n=240), in various regions of mainland Portugal, primarily obtained from wildlife rehabilitation centers (WRCs). Hemosporidian species were detected using nested PCRs, targeting the cytochrome b gene of the mitochondria. For species and lineage identification, 19 positive PCR samples w

[Infectious cysts and pseudocysts: When parasites want to mimic tumors!].

Parasites can form true tissular cysts, with cavity(ies) delimited by a wall, covered by a specialized parasitic epithelium. In humans, these tissue cysts are caused by cestodes (segmented flatworms), which cannot complete their full cycle (e.g., in hydatidosis, alveolar echinococcosis or cysticercosis). Parasites can also form tissue pseudocysts, the wall of which consists of a resorptive granulomatous reaction to the presence of the parasite. In humans, these pseudocysts are caused by both nematodes (roundworms) and cestodes. These pathogens are rarely encountered by pathologists, and their …

Parasites in the bloodstream: Unraveling hematological chaos and clinical implications.

Parasitic infections pose a significant global health burden, particularly in regions with limited medical resources and poor sanitation. These infections often have detrimental effects on the human body, including disruptions to the hematopoietic system, the intricate network responsible for blood cell production. Parasites such as Plasmodium, Schistosoma, and Leishmania are notorious for causing blood disorders such as anemia, thrombocytopenia, and leukopenia. Understanding the intricate interactions between parasites and the hematopoietic system is crucial for developing effective diagnosti…

Versatile Imidazole Scaffold with Potent Activity against Multiple Apicomplexan Parasites.

Malaria, toxoplasmosis, and cryptosporidiosis are caused by apicomplexan parasites Plasmodium spp., Toxoplasma gondii, and Cryptosporidium parvum, respectively, and pose major health challenges. Their therapies are inadequate, ineffective or threatened by drug resistance. The development of novel drugs against them requires innovative and resource-efficient strategies. We exploited the kinome conservation of these parasites to determine the cellular targets and effects of two Plasmodium falciparum inhibitors in T. gondii and C. parvum. The imidazoles, (R)-RY-1-165 and (R)-RY-1-185, were develo…

Novel isothermal nucleic acid amplification method for detecting malaria parasites.

Malaria, a parasitic disease caused by Plasmodium spp. and transmitted by Anopheles mosquitoes, remains a major global health issue, with an estimated 249 million cases and 608,000 deaths in 2022. Rapid and accurate diagnosis and treatment are crucial for malaria control and elimination. However, limited access to sensitive molecular tests means that microscopic examination and rapid diagnostic tests (RDT) are the most used methods in endemic areas, despite their lower diagnostic accuracy. Therefore, there is a need for developing sensitive, simple, accurate, and rapid diagnostic tools suitabl

Zoonotic Lagochilascaris minor and nine other parasites in a cat.

A case of polyparasitism in an 8-year-old female cat from Southern Brazil is reported. Among the described clinical signs, both gurltiosis and lagochilascariasis are of rare occurrence, being categorized as neglected diseases. The diagnosis of feline crural parasitic paraplegia was established through clinical signs, necropsy lesion observations, and the presence of Gurltia paralysans in histological sections of the spinal cord. Lagochilascaris minor was diagnosed with an ulcerated wound in the cervical region and further confirmed by coprology. In addition, other gastrointestinal parasites (i…

Gastrointestinal Parasites in Asian and African Elephants: A Systematic Review.

Gastrointestinal parasites (GIPs) in elephants have been reported in several studies over the last decades. Nonetheless, comprehensive data on clinicopathology of elephant GIPs, parasite burden threshold value, and the effectiveness of conventional anthelmintic drugs are still lacking. Herein, we have systematically reviewed the available knowledge on elephant GIPs identified among different parts of the world based on their prevalence, epidemiology, pathology, diagnosis, treatment, and control. Two electronic databases were searched for publications that met the inclusion criteria. About19 En…

Propensity of selecting mutant parasites for the antimalarial drug cabamiquine.

We report an analysis of the propensity of the antimalarial agent cabamiquine, a Plasmodium-specific eukaryotic elongation factor 2 inhibitor, to select for resistant Plasmodium falciparum parasites. Through in vitro studies of laboratory strains and clinical isolates, a humanized mouse model, and volunteer infection studies, we identified resistance-associated mutations at 11 amino acid positions. Of these, six (55%) were present in more than one infection model, indicating translatability across models. Mathematical modelling suggested that resistant mutants were likely pre-existent at the t…
