An In Silico Investigation of the Pathogenic G151R G Protein-Gated Inwardly Rectifying K+ Channel 4 Variant to Identify Small Molecule Modulators.

Primary aldosteronism is characterised by the excessive production of aldosterone, which is a key regulator of salt metabolism, and is the most common cause of secondary hypertension. Studies have investigated the association between primary aldosteronism and genetic alterations, with pathogenic mutations being identified. This includes a glycine-to-arginine substitution at position 151 (G151R) of the G protein-activated inward rectifier potassium (K+) channel 4 (GIRK4), which is encoded by the KCNJ5 gene. Mutations in GIRK4 have been found to reduce the selectivity for K+ ions, resulting in m

Effects of Small Conductance Ca2+-Activated Potassium Channel Agonists on SLEs in Varied Epilepsy Models, From Animal Slices to Pharmacoresistant Human Tissue.

To assess the impact of SK channel agonists on seizure-like events (SLEs) in various seizure models in slices of the temporal cortex obtained from pharmacoresistant patients. SLEs were triggered by applying 4-aminopyridine (100 μM) to slices of the entorhinal cortex taken from both normal and pilocarpine-treated rats. Additionally, SLEs were induced in slices of the temporal cortex obtained from individuals who had undergone epilepsy surgery. In the case of human slices, SLEs were also provoked by increasing potassium levels along with the administration of either 4-AP (100 μM) or bicuculline

Phosphorus-potassium fertilizer exposure induces oxidative stress and riboflavin metabolism disruption in juvenile turbot (Scophthalmus maximus).

Intensive agriculture's excessive fertilizer use significantly contributes to coastal nutrient pollution. While phosphorus's ecological impacts are recognized, the toxicity of combined phosphorus nutrients on marine life remains unclear. This study examined phosphorus-potassium fertilizer's (P-K fertilizer) effects on juvenile turbot (Scophthalmus maximus) via a 96-hour acute exposure experiment. Exposure solutions were prepared at environmentally relevant concentrations, resulting in elevated seawater phosphorus and potassium levels by 117.25 mg/L and 152.94 mg/L, respectively. Compound ferti

Identification of compound mutations of SLC12A3 gene in a Chinese pedigree with Gitelman syndrome exhibiting Bartter syndrome-liked phenotypes.

BACKGROUND: Gitelman syndrome is a rare salt-losing renal tubular disorder associated with mutation of SLC12A3 gene, which encodes the Na-Cl co-transporter (NCCT). Gitelman syndrome is characterized by hypokalemia, metabolic alkalosis, hypomagnesemia, hypocalciuria, and renin-angiotensin-aldosterone system (RAAS) activation. Different SLC12A3 variants may lead to phenotypic variability and severity. METHODS: In this study, we reported the clinical features and genetic analysis of a Chinese pedigree diagnosed with Gitelman syndrome. RESULTS: The proband exhibited hypokalaemia, hypomagnesemia,

Iodine stability in salt double-fortified with iron and iodine.

Deficiencies in small quantities of micronutrients, especially iodine and iron, severely affect more than a third of the world's population, resulting in serious public health consequences, especially for women and young children. Salt is an ideal carrier of micronutrients. The double fortification of salt with both iodine and iron is an attractive approach to the reduction of both anemia and iodine-deficiency disorders. Because iodine is unstable under the storage conditions found during the manufacturing, distribution, and sale of salt in most developing countries, the effects of packaging m

Enhancing the Cycling Stability by Tuning the Chemical Bonding between Phosphorus and Carbon Nanotubes for Potassium-Ion Battery Anodes.

Phosphorus/carbon (P/C) composites as promising potassium-ion storage materials have been extensively investigated for its compound superiorities of high specific capacity and favorable electronic conductivity. However, the effects of different chemical bonding states between P and the carbon matrix for potassium-ion storage and cycling performance still need to be investigated. Herein, three P/C composites with different chemical bonding states were successfully fabricated through simply ball-milling red P with carboxylic group carbon nanotubes (CGCNTs), carbon nanotubes (CNTs), and reduced c

Acute activation of endothelial AMPK surprisingly inhibits endothelium-dependent hyperpolarization-like relaxations in rat mesenteric arteries.

BACKGROUND AND PURPOSE: Endothelium-dependent hyperpolarizations (EDHs) contribute to the regulation of peripheral resistance. They are initiated through opening of endothelial calcium-activated potassium channels (KCa ); the potassium ions released then diffuse to the underlying smooth muscle cells, causing hyperpolarization and thus relaxation. The present study aimed to examine whether or not AMPK modulates EDH-like relaxations in rat mesenteric arteries. EXPERIMENTAL APPROACH: Arterial rings were isolated for isometric tension recording. AMPK activity and protein level were measured by EL

Selective and state-dependent activation of TRESK (K2P 18.1) background potassium channel by cloxyquin.

BACKGROUND AND PURPOSE: Cloxyquin (5-cloroquinolin-8-ol) has been described as an activator of TRESK (K2P 18.1, TWIK-related spinal cord K+ channel) background potassium channel. We have examined the specificity of the drug by testing several K2P channels. We have investigated the mechanism of cloxyquin-mediated TRESK activation, focusing on the differences between the physiologically relevant regulatory states of the channel. EXPERIMENTAL APPROACH: Potassium currents were measured by two-electrode voltage clamp in Xenopus oocytes and by whole-cell patch clamp in mouse dorsal root ganglion (D

Influence of Humic Acids on the Removal of Arsenic and Antimony by Potassium Ferrate.

Although the removal ability of potassium ferrate (K2FeO4) on aqueous heavy metals has been confirmed by many researchers, little information focuses on the difference between the individual and simultaneous treatment of elements from the same family of the periodic table. In this project, two heavy metals, arsenic (As) and antimony (Sb) were chosen as the target pollutants to investigate the removal ability of K2FeO4 and the influence of humic acid (HA) in simulated water and spiked lake water samples. The results showed that the removal efficiencies of both pollutants gradually increased alo

Kv1.5 blockers preferentially inhibit TASK-1 channels: TASK-1 as a target against atrial fibrillation and obstructive sleep apnea?

Atrial fibrillation and obstructive sleep apnea are responsible for significant morbidity and mortality in the industrialized world. There is a high medical need for novel drugs against both diseases, and here, Kv1.5 channels have emerged as promising drug targets. In humans, TASK-1 has an atrium-specific expression and TASK-1 is also abundantly expressed in the hypoglossal motor nucleus. We asked whether known Kv1.5 channel blockers, effective against atrial fibrillation and/or obstructive sleep apnea, modulate TASK-1 channels. Therefore, we tested Kv1.5 blockers with different chemical struc

Properties of ionic currents from isolated adult rat carotid body chemoreceptor cells: effect of hypoxia.

1. The electrical properties of chemoreceptor cells from neonatal rat and adult rabbit carotid bodies (CBs) are strikingly different. These differences have been suggested to be developmental and/or species related. To distinguish between the two possibilities, the whole-cell configuration of the patch-clamp technique was used to characterize the ionic currents present in isolated chemoreceptor cells from adult rat CBs. Since hypoxia-induced inhibition of O2-sensitive K+ currents is considered a crucial step in O2 chemoreception, the effect of hypoxia on the adult rat chemoreceptor cell curren

Functional role of Ca(2+)-activated K+ channels in resting state of carotid arteries from SHR.

Carotid arteries from spontaneously hypertensive rats (SHR) exhibited an active tone when exposed to a physiological salt solution; that is, the tension decreased when nifedipine was added. To determine the possible role of Ca(2+)-activated K+ (KCa) channels in the resting state of these arteries, the effects of agents that interact with these channels on tension and 86Rb efflux were compared in endothelium-denuded strips of carotid arteries from SHR and normotensive Wistar-Kyoto rats (WKY). The addition of charybdotoxin, a blocker of high-conductance KCa channels, to the resting strips produc

Cardiac tamponade: an alternative procedure for late feticide.

OBJECTIVE: Improvements in equipment and diagnostic skills mean that more abnormalities can now be detected antenatally, thus increasing the demand for pregnancy termination at later stages of gestation. Potassium chloride injected into the fetal circulation is the most frequently used procedure. In this study, we propose a new method of feticide using injection of normal saline into the fetal pericardial space to induce cardiac tamponade, resulting in late fetal reduction with minimal maternal risk. CASE REPORT: A 32-year-old, gravida 6, woman was a carrier of a balanced translocation 45,XX,

Effects of KCNQ1 channel blocker, 293B, on the acetylcholine-induced Cl- secretion of rat pancreatic acini.

In rat pancreatic acini (RPAs), acetylcholine (ACh) typically induces a tonic depolarization of membrane potential (Vm) via increasing cytoplasmic Ca2+ concentration and subsequent activation of Cl- channels. In this study, to investigate the role of K+ channels during the ACh-induced Cl- secretion, the intracellular Cl- concentration ([Cl-]i) of RPAs was monitored using SPQ, a fluorescent dye quenchable by Cl-, and the effects of K+ channel blockers were examined. Also, the secretion of fluid and enzyme from the whole pancreas of rat was measured. The fluorescence of RPAs loaded with SPQ (FSP

Improving the analysis of small precipitates in HSLA steels using a plasma cleaner and ELNES.

The change from producing high strength low alloy (HSLA) steel sheet by conventional thick slab casting to producing it by direct charged thin slab casting causes a major change in the evolution of the precipitation. A key area of interest is the composition of the sub-10nm precipitates used to produce dispersion hardening. Carbon extraction replicas are frequently used to study precipitates in steels and other metals. When used with annular dark field imaging, this technique gives high contrast images of the precipitates while the thin carbon film adds little background or additional characte

Enabling Noninvasive Systemic Delivery of the Kv1.3-Blocking Peptide HsTX1[R14A] via the Buccal Mucosa.

The peptide HsTX1[R14A] is a potent and selective blocker of the voltage-gated potassium channel Kv1.3, a well-recognized therapeutic target for autoimmune diseases. To overcome the poor oral absorption and consequent need for regular injections, the potential of the buccal mucosa for systemic delivery of HsTX1[R14A] was investigated. For in vitro studies, FITC-HsTX1[R14A] and HsTX1[R14A], in solution or formulated in a mucoadhesive chitosan-based gel (3%, w/v) with or without cetrimide (5%, w/w), were applied to porcine buccal epithelium mounted between Ussing chambers and buccal mucosal perm

Tin oxide coating of aluminous porcelain by reactive ion plating.

Alumina reinforced dental porcelain has been coated directly with tin oxide by reactive ion plating. Samples were prepared at different distances from the tin source in the ion plating rig. Tensile bond strengths of treated and untreated porcelain discs to a commercially available phosphate-methacrylate based dental cement were determined. Bond strengths of certain coated samples were found to be in excess of the cohesive strength of the porcelain substrates (greater than 7.8 MPa), whereas untreated porcelain achieved an average bond strength of only 3.4 MPa. The microstructures of coatings pr

Foliar spraying of salicylic acid and melatonin enhances the nutritional value of Arugula (Eruca vesicaria subsp. sativa mill.).

BACKGROUND: Arugula (Eruca vesicaria subsp. Sativa Mill.) is a leafy vegetable rich in nutritional value, vitamins, minerals, and antioxidants. While previous studies showed that foliar applications of melatonin and salicylic acid influence plant nutrient levels, research on the individual and combined effects of these two compounds in Arugula is Limited. Therefore, this study aims to assess both the individual and combined effects of salicylic acid and melatonin on the concentrations of essential elements, such as nitrogen, phosphorus, potassium, and various micronutrients in Arugula. Yeast strain Saccharomyces cerevisiae BYT45 lacking the cation extrusion systems ENA1-5 and NHA1 is suitable for the characterization of TASK-3 potassium channel antagonists.

Finding new potential antagonists of potassium channels is a continuing task. TASK potassium channels operate over a large physiological range of membrane voltages, why they are thought to contribute to the excitability and resting potential of mammalian membrane potentials. Additionally, they are regulated by extracellular stimuli like changes in pH and K+ concentrations. TASK malfunctions are associated with diseases, which makes them popular targets for the search of new antagonists. Identification of channel inhibitors can be a time-consuming and expensive project. Here, we present an easy
