Interpreting rare missense variants in ABCB4 using ACMG recommended in silico tools, bespoke prediction algorithm Vasor and AlphaMissense.

Intrahepatic Cholestasis of Pregnancy (ICP) is the most common pregnancy-related liver disorder. ABCB4 heterozygous variants are implicated in ICP, but interpretation of rare variants remains challenging, leading to many variants of uncertain significance. This study compares predictions from multiple in silico tools on nine heterozygous missense variants identified in women with ICP. Using the Genomics England Research Environment, 253 women with ICP and whole-genome sequencing data were analysed. Variants with minor allele frequency < 0.05 were filtered, and nine rare missense variants were

AI-Powered Resting 12-Lead Electrocardiogram Algorithm for Predicting Low Peak Oxygen Consumption: Development and Validation Study.

BACKGROUND: Low peak oxygen consumption (V̇O2) is associated with higher cardiovascular and all-cause mortality, while improvements in peak V̇O2 reduce this risk. Although early detection allows timely intervention, practical screening tools remain lacking. As electrocardiograms (ECGs) reflect both cardiac and age-related changes, they may offer a viable screening approach. OBJECTIVE: This study aimed to detect low peak V̇O2 using resting 12-lead ECGs analyzed by a trained neural network. METHODS: The low peak V̇O2 estimation model was developed using data from 965 individuals (n=540, 56% wi

Multioutput Machine Learning Model for Predicting Postoperative Outcomes After Liposuction: Algorithm Development and Validation Study in a Multicenter Cohort.

BACKGROUND: Liposuction is widely performed to remove localized fat deposits and improve body contour, yet individualized prediction of postoperative outcomes remains challenging. Existing machine learning (ML) studies have largely focused on single-outcome prediction, with limited attention to the interdependence between postoperative body weight and circumferential size. OBJECTIVE: This study aimed to develop and validate a chained multioutput ML framework to jointly predict postoperative body weight and circumferential size after liposuction using a large multicenter cohort from the 365mc

Laboratory Medicine in Immune Checkpoint Inhibitor-Induced Autoimmune Disorders: Baseline Assessment, Diagnostic Algorithms, Monitoring, and Predictive Biomarkers.

Immune checkpoint inhibitors (ICIs) have substantially improved clinical outcomes across multiple malignancies, but they can disrupt self-tolerance and induce immune-related adverse events resembling autoimmune disorders. In many cases, laboratory abnormalities precede overt clinical manifestations, placing laboratory medicine at the center of baseline evaluation, early recognition, diagnostic confirmation, longitudinal monitoring, and treatment follow-up. This review provides a laboratory-focused overview of ICI-induced autoimmune disorders, focusing on baseline laboratory assessment, monitor

OBJECTIVE: The aim of this study was to screen the biomarkers of ginger against gastric cancer (GC) by network pharmacology, WGCNA and machine algorithms. To find the upstream transcription factors and downstream signaling proteins constituting the signaling axis, so as to predict the possible mechanism of action of ginger against GC. METHODS: Ginger was screened for active ingredients and targets through public databases. GC genes were screened using disease database, GEO database and WGCNA. The intersection of the four was taken to obtain the potential core genes. Machine algorithms was use

Developing predictive models for COVID-19 positive tests based on the XGBoost and random forest algorithms with internet search data.

BACKGROUND: Although strategies for COVID-19 have shifted towards normalized measures globally, establishing predictive models based on Internet search data remains crucial for swiftly controlling and preventing future outbreaks. This study aims to utilize Internet search data for early epidemic surveillance and warning. METHODS: We collected the daily number of COVID-19 positive tests and the daily Baidu Search Index (BSI) of COVID-19 related keywords. First, we screened keywords with a maximum correlation coefficient exceeding 0.9 by time-lagged correlation analysis. Then, we used the origi

Development of deep learning auto-encoder algorithms for predicting alcohol use in Korean adolescents based on cross-sectional data.

Alcohol is a highly addictive substance, presenting significant global public health concerns, particularly among adolescents. Previous studies have been limited by traditional research methods, making it challenging to encompass diverse risk factors and automate screening or prediction of adolescents' alcohol use. This study aimed to develop prediction algorithms for adolescent alcohol use in South Korea using machine learning (ML) and deep learning (DL) models, and to identify important features. The study utilized a combination of DL (i.e., Auto-encoder) and ML (i.e., Logistic regression, R…

A machine learning approach to predict postoperative sleep disturbance after total knee arthroplasty: a comparative study of multiple algorithms.

BACKGROUND: Postoperative sleep disturbance (PSD) is a common complication following total knee arthroplasty (TKA), which negatively impacts patient recovery. Despite the critical need for early detection and management, there is limited research on predictive models for early PSD, particularly those integrating machine learning (ML) techniques. OBJECTIVE: This study aimed to develop a predictive model for early PSD following TKA using ML algorithms, identify key predictive factors, and provide an interpretable model to guide clinical decision-making. METHODS: The study included 505 patients

Assessing accuracy of BiliPredics algorithm in predicting individual bilirubin progression in neonates-results from a prospective multi-center study.

BACKGROUND: Neonatal jaundice affects more than half of neonates. As bilirubin values usually peak few days after hospital discharge, jaundice remains a leading cause of rehospitalization. The recently developed BiliPredics algorithm, integrated in the first CE-approved bilirubin prediction tool, predicts individual bilirubin progression for up to 60 h into the future. Goal of the prospective study was to assess accuracy of this algorithm in predicting individual bilirubin prior to hospital discharge in neonates. METHODS: A prospective multi-center study was conducted in 2021 at the Universit

The best machine learning algorithm for building surgical site infection predictive models: A systematic review and network meta-analysis.

BACKGROUND: Many machine learning (ML) algorithms have been used to develop surgical site infection (SSI) prediction models, but little is known about their predicting performance. We conducted a network meta-analysis to compare the performance of different ML algorithms and to explore which one may perform best. METHODS: MEDLINE, EMBASE, CINAHL, Web of Science, and Cochrane Library were systematically searched from inception to November 25, 2023. We included diagnostic accuracy trials constructing SSI predictive model by ML. Two reviewers selected relevant studies and extracted data. The cer
