Serologic and Molecular Evidence of Arboviruses in Nonhuman Primates in Northeast Brazil.

Arbovirus surveillance in marmosets (Callithrix spp.) that live close to humans helps identify viral circulation in the environment and contributes to public health. We investigated the exposure to arboviral infections in 47 captive and free-living Callithrix from urban and peri-urban areas in the semiarid region of northeastern Brazil (SNB) in 2018. The samples were tested for antibodies against 23 arbovirus antigens and the presence of Orthoflavivirus zikaense (ZIKV), Orthoflavivirus denguei, Alphavirus chikungunya, and Orthoflavivirus flavi using hemagglutination inhibition (HI) and RT-qPCR

Mandibular shape and diet in extant primates: a 3D geometric morphometric analysis.

Establishing a link between mandibular morphology and diet in extant primates has long been a goal in biological anthropology because it should provide important insight into the diets of extinct primates, including fossil hominins. To date, efforts to explore this question have produced mixed results, largely perhaps due to a reliance on the use of 2D morphological data. Here, we report a study where we investigated whether 3D shape data would provide a clearer picture. We used geometric morphometrics to analyse 3D mandibular shape variation in a sample of > 200 primate specimens, representin…

Onset and Progression of Disease in Nonhuman Primates With PDE6C Cone Disorder.

PURPOSE: The California National Primate Research Center contains a colony of rhesus macaques with a homozygous missense mutation in PDE6C ( … SIONS: These data suggest that, like humans, macaque PDE6C heterozygotes are normal, and homozygote primates have absent cone function and reduced foveal ONL thickness from infancy. Cone photoreceptors probably degenerate over time and macular atrophy can occur. Rod photoreceptor function may wane in late stages.

Quantitative Analysis of the Carpal Tunnel and Its Inner Structures in Primates.

To explore the anatomical factors potentially involved in the high incidence of carpal tunnel syndrome in humans, we have quantified the anatomical variations of the carpal tunnel and its inner structures in humans, non-hominoid primates (monkeys), and hominoid primates (apes). In specimens of six humans, eight monkeys, and three apes, we assessed the size of the carpal tunnel, the tendons of the digit flexor muscles, and the median nerve. We compared the size of the carpal tunnel normalized by the wrist size, and the size of the median nerve and the tendons of the digit flexors normalized by …

Cognitive and kinematic markers of ketamine effects in behaving non-human primates.

Ketamine is widely used to probe cognitive functions relying on the properties of methyl-D-aspartate receptor (NMDAR) synaptic transmission. Numerous works have proved that cognitive performance and adjustments in the decision or perceptual domains are affected after ketamine injection in general circulation of primates. Here, we take advantage of that in the brain stem; horizontal saccade deceleration is controlled by glycine-NMDAR-gated current, while gamma-aminobutyric acid (GABA) current controls vertical deceleration to demonstrate that despite general circulation level manipulation of NM…

Monoclonal antibody therapy protects nonhuman primates against mucosal exposure to Lassa virus.

Lassa fever (LF) is an acute viral illness that causes thousands of deaths annually in West Africa. There are currently no Lassa virus (LASV) vaccines or antivirals approved for human use. Recently, we showed that combinations of broadly neutralizing human monoclonal antibodies (BNhuMAbs) known as Arevirumab-2 or Arevirumab-3 protected up to 100% of cynomolgus macaques against challenge with diverse lineages of LASV when treatment was initiated at advanced stages of disease. This previous work assessed efficacy against parenteral exposure. However, transmission of LASV to humans occurs primari

Zygomatic arch root position in relation to dietary type in haplorhine primates.

The zygomatic root, along with other key craniofacial features, is hypothesized to play a crucial role in strengthening the face in response to stresses and strains related to feeding. As such, it has been cited as indicative of dietary specialization among fossil taxa, although it remains unknown how variable zygomatic arch root position is among living primates, and whether its positioning predicts differences in diet. We test whether primates that consume more mechanically challenging foods possess more anteriorly positioned zygomatic roots compared to those consuming less challenging foods…

The neural substrate and underlying mechanisms of executive control fluctuations in primates.

Trial-by-trial alterations in response time have been linked to fluctuations of executive control and transient lapses of attention. Here, w … the ongoing task. These findings bring insights to the neural architecture of executive control in primates and suggest that DLPFC and ACC support sustained executive control, but OFC is more involved in setting and restoring the control.

Negative seroprevalence for Toxoplasma gondii in free-living primates from Central Amazonia.

This study presents 35 negative serologies for antibodies anti-T. gondii in free-living primates from Central Amazonia. Our results suggest that these populations have not had contact with the parasite and, therefore, do not have antibodies. This was the first study surveying T. gondii in Cacajao, Callicebus, Pithecia, and Saguinus monkeys.

INDY-From Flies to Worms, Mice, Rats, Non-Human Primates, and Humans.

I'm Not Dead Yet (Indy) is a fly homologue of the mammalian SLC13A5 (mSLC13A5) plasma membrane citrate transporter, a key metabolic regulator and energy sensor involved in health, longevity, and disease. Reduction of Indy gene activity in flies, and its homologs in worms, modulates metabolism and extends longevity. The metabolic changes are similar to what is obtained with caloric restriction (dietary restriction). Similar effects on metabolism have been observed in mice and rats. As a citrate transporter, INDY regulates cytoplasmic citrate levels. Indy flies heterozygous for a P-element inser

A diastolic dysfunction model in non-human primates with transverse aortic constriction.

Transverse aortic constriction (TAC) has been widely used to study cardiac hypertrophy, fibrosis, diastolic dysfunction, and heart failure in rodents. Few studies have been reported in preclinical animal models. The similar physiology and anatomy between non-human primates (NHPs) and humans make NHPs valuable models for disease modeling and testing of drugs and devices. In the current study, we aimed to establish a TAC model in NHPs and characterize the structural and functional profiles of the heart after TAC. A non-absorbable suture was placed around the aorta between the brachiocephalic art…

In vivo CRISPR base editing of PCSK9 durably lowers cholesterol in primates.

Gene-editing technologies, which include the CRISPR-Cas nucleases1-3 and CRISPR base editors4,5, have the potential to permanently modify disease-causing genes in patients6. The demonstration of durable editing in target organs of nonhuman primates is a key step before in vivo administration of gene editors to patients in clinical trials. Here we demonstrate that CRISPR base editors that are delivered in vivo using lipid nanoparticles can efficiently and precisely modify disease-related genes in living cynomolgus monkeys (Macaca fascicularis). We observed a near-complete knockdown of PCSK9 in …

Clinicopathologic and Transcriptomic Analysis of Radiation-Induced Lung Injury in Nonhuman Primates.

PURPOSE: Radiation-induced lung injury (RILI) is a progressive condition with an early phase (radiation pneumonitis) and a late phase (lung fibrosis). RILI may occur after partial-body ionizing radiation exposures or internal radioisotope exposure, with wide individual variability in timing and extent of lung injury. This study aimed to provide new insights into the pathogenesis and progression of RILI in the nonhuman primate (NHP) rhesus macaque model. METHODS AND MATERIALS: We used an integrative approach to understand RILI and its evolution at clinical and molecular levels in 17 NHPs expos

Recombinant Human HPS Protects Mice and Nonhuman Primates from Acute Liver Injury.

Acute liver injury shares a common feature of hepatocytes death, immune system disorders, and cellular stress. Hepassocin (HPS) is a hepatokine that has ability to promote hepatocytes proliferation and to protect rats from D-galactose (D-Gal)- or carbon tetrachloride (CCl4)-induced liver injury by stimulating hepatocytes proliferation and preventing the high mortality rate, hepatocyte death, and hepatic inflammation. In this paper, we generated a pharmaceutical-grade recombinant human HPS using mammalian cells expression system and evaluated the effects of HPS administration on the pathogenesi
