Due to their advantages in structural stability and versatility, cysteine-rich peptides, which are secreted from the venom glands of venomous animals, constitute a naturally occurring pharmaceutical arsenal. However, the correct folding of disulfide bonds is a challenging task in the prokaryotic expression system like Escherichia coli due to the reducing environment. Here, a secretory expression plasmid pSE-G1M5-SUMO-HWTX-I for the spider neurotoxin huwentoxin-I (HWTX-I) with three disulfides as a model of cysteine-rich peptides was constructed. By utilizing the signal peptide G1M5, the fusion

Amino acids undergo many covalent modifications, but only few amino acid repair enzymes have been identified. Protein-L-isoaspartate (D-aspartate) O-methyltransferase (PIMT), also known as L-isoaspartyl/D-aspartyl protein carboxyl methyltransferase (PCMT), methylates covalently modified isoaspartate (isoAsp) residues accumulated in proteins via Asn deamidation and Asp hydrolysis. This cytoplasmic reaction occurs through the formation of succinimide cyclical intermediate and generates either isoAsp or Asp from succinimide. Succinimide conversion into Asp is spontaneous, while isoAsp is restored

The variable domain of the heavy chain antibodies (VHHs) is the smallest (15 kDa) intact single domain antigen-binding fragment. VHHs often exhibit sub-nanomolar affinity for their designated targets and therefore are receiving increasing attention in molecular targeting and nanomedicine engineering. We cloned and expressed four non-overlapping anti-HER2 VHHs in a prokaryotic expression system that yielded disulfide-bonded VHHs. Purified VHHs, before and after thiolation, were characterized by Western blot and their functionality against the ecto-domain of HER2 receptor was confirmed by ELISA

Trypanosoma cruzi, the agent of the American Trypanosomiasis, Chagas disease, contains cysteine, serine, threonine, aspartyl and metallo peptidases. The most abundant among these enzymes is cruzipain, a cysteine proteinase expressed as a mixture of isoforms, some of them membrane-bound. The enzyme is an immunodominant antigen in human chronic Chagas disease and seems to be important in the host/parasite relationship. Inhibitors of cruzipain kill the parasite and cure infected mice, thus validating the enzyme as a very promising target for the development of new drugs against the disease. In ad

Close reciprocal interactions in symbiotic systems have suggested the holobiont concept, in which the host and its microbiota are considered as a single entity. Ciliates are known for their ability to form symbiotic associations with prokaryotes. Relationships between the partners in such systems vary from mutualism to parasitism and differ significantly in their robustness. We assessed the viability of the ciliate Paramecium multimicronucleatum and its ability to maintain its intranuclear endosymbiont Ca. Trichorickettsia mobilis (Rickettsiaceae) after treatment with antibiotics characterized

Eimeria tenella is a highly pathogenic protozoan parasite that causes severe hemorrhagic cecitis in chickens, leading to substantial economic losses to the global poultry industry. Interleukin-1β (IL-1β), a master regulator of inflammation, is strongly upregulated during E. tenella infection; however, its precise functional role in cecal inflammation remains unclear. In this study, recombinant chicken IL-1β and IL-6 proteins were expressed and purified using a prokaryotic expression system. Hyperimmune sera against IL-1β and IL-6 were generated in specific pathogen-free (SPF) chickens and admi

Iron is considered an essential component of all life forms, but is maintained optimally by fine-tuning of iron homeostasis mechanisms. Iron storage proteins (ISPs), which are ferritin superfamily proteins, play a pivotal role in maintaining iron homeostasis by storing excess cellular iron, often serving as ROS scavengers for protecting cells from oxidative stress. Xanthomonas represents a large and ubiquitous group of phytopathogenic bacteria that causes severe crop loss worldwide. Xanthomonas survives host-imposed iron stress by governing iron uptake, transport, and metabolism in utmost coor

Glutathione peroxidases are conserved enzymes found in prokaryotic and eukaryotic organisms that reduce H2O2 to protect cells from oxidative stress damage. Here, we identified and characterized the GpoA glutathione peroxidase of Streptococcus pneumoniae, one of the most important human bacterial pathogens. We demonstrated that recombinant GpoA exhibits glutathione peroxidase activity and that the Cys36 residue is essential for this function. We conducted a comparative analysis of the phenotypic characteristics linked to GpoA and those linked to other peroxidases, including the TpxD thiol perox

Lawsonia intracellularis, the etiological agent for proliferative enteropathy, remains a major cause of productivity loss and economic burden in the global swine industry. Currently available vaccines provide suboptimal protection and raise safety concerns, highlighting the need for improved immunization strategies. In this study, we developed Salmonella Typhimurium-based vaccine candidates against L. intracellularis in a murine model, incorporating immunoprotective epitopes from the surface autotransporter protein LatA, the flagellar protein FliC, and the heat shock protein 60 (HSP60). Extracellular vesicles (EVs) have gained increasing attention in recent years due to their pivotal role in both health and disease. Emerging from both eukaryotic and prokaryotic cells, EVs serve as essential mediators of intercellular communication, exceeding the simplistic interactions observed with individual molecules. In this comprehensive review, we will focus on both Bacterial Extracellular Vesicles (BEV) and on Host derived Extracellular Vesicles (HEV) and highlight mechanistic principles, as well as their transformation into diagnostic and therapeutic tools. We will start with a short …

Pyocyanin is the primary phenazine produced by Pseudomonas aeruginosa, which possesses redox activity. This compound contributes to its virulence by generating reactive oxygen species and inducing oxidative stress in prokaryotic and eukaryotic cells. P. aeruginosa is able to resist high concentrations of pyocyanin despite its toxicity. In this work, we employed P. aeruginosa ID4365 and PAO1 rsmA mutants, which produce 276.2 μM and 6.9 μM of pyocyanin, respectively, to conduct a proteomic analysis in order to identify proteins involved in the protective response against this phenazine. We found

Tuberculosis (TB) is a chronic and persistent infectious disease caused by Mycobacterium tuberculosis (Mtb), posing a serious threat to global public health. The only widely available vaccine for TB is Bacillus Calmette-Guérin (BCG), and its limitations have become increasingly apparent in recent years, underscoring the urgent need for new TB vaccines. Moreover, subunit vaccines undergoing clinical trials face challenges such as complex production processes and weak immunogenicity. Efforts must be made to develop a safe, effective, easily mass-produced TB vaccine. Here, early secreted antigeni

Fibroblast growth factor 21 (FGF21) plays important roles in the regulation of glucose and lipid metabolism and energy balance in mammals. In this study, the full-length cDNA of swamp eel fgf21 was cloned. Sequence analysis showed that swamp eel FGF21 displayed high similarity with FGF21 of other vertebrates. Subsequently, a prokaryotic expression vector for swamp eel fgf21 was constructed, and recombinant FGF21 (rFGF21) was successfully induced and purified. To investigate the potential roles of swamp eel FGF21 in glucose and lipid metabolism, we examined the effects of rFGF21 on regulation o

Copper homeostasis is tightly regulated in both prokaryotic and eukaryotic cells to ensure sufficient amounts for cuproprotein biosynthesis, while limiting oxidative stress production and toxicity. Over the last century, copper complexes have been developed as antimicrobials and for treating diseases involving copper dyshomeostasis (e.g., Wilson's disease). There now exists a repertoire of copper complexes that can regulate bodily copper through a myriad of mechanisms. Furthermore, many copper complexes are now being appraised for a variety of therapeutic indications (e.g., Alzheimer's disease

Leptospirosis caused by Leptospira is a zoonotic disease of global importance but it is considered as an emerging or re-emerging infectious disease in many areas in the world. Until now, the mechanisms about pathogenesis and transmission of Leptospira remains poorly understood. As eukaryotic and prokaryotic proteins can be denatured in adverse environments and chaperone-protease/peptidase complexes degrade these harmful proteins, we speculate that infection may also cause leptospiral protein denaturation, and the HslU and HslV proteins of L. interrogans may compose a complex to degrade denatur

Voltage-gated sodium channels are key to the initiation and propagation of action potentials in electrically excitable cells. Molecular characterization has shown there to be nine functional members of the family, with a high degree of sequence homology between the channels. This homology translates into similar biophysical and pharmacological properties. Confidence in some of the channels as drug targets has been boosted by the discovery of human mutations in the genes encoding a number of them, which give rise to clinical conditions commensurate with the changes predicted from the altered ch

Coxiella burnetii is an obligate intracellular bacterium that directs biogenesis of a parasitophorous vacuole (PV) for replication. Effectors of PV maturation are likely translocated into the host cytosol by a type IV secretion system (T4SS) with homology to the Dot/Icm apparatus of Legionella pneumophila. Since secreted bacterial virulence factors often functionally mimic the activities of host proteins, prokaryotic proteins with eukaryotic features are considered candidate T4SS substrates. Genes encoding proteins with eukaryotic-type ankyrin repeat domains (Anks) were identified upon genome

YhhP, a small protein of 81 amino acid residues encoded by the yhhP gene in the Escherichia coli database, is implicated in cell division although the precise biological function of this protein has not been yet identified. A variety of microorganisms have similar proteins, all of which contain a common CPxP sequence motif in the N-terminal region. We have determined the three-dimensional solution structure of YhhP by NMR spectroscopy in order to obtain insight into its biological function. It folds into a two-layered alpha/beta-sandwich structure with a betaalphabetaalphabetabeta fold, compri
