Urine concentration and avian aquaporin water channels.

Although birds and mammals have evolved from primitive tetrapods and advanced divergently, both can conserve water by producing hyperosmotic … counterparts, but their distribution and function may not be the same. AQP2 expression in Japanese quail (q) evolves in the collecting duct of early metanephric kidneys and continues to increase in intensity and distribution during nephrogenesis and maturation. qAQP2 mRNA and protein are increased by arginine vasotocin (avian ADH), but vasotocin-induce …

Genotypic characterization and phylogenetic analysis of Cryptosporidium sp. from domestic animals in Brazil.

The purpose of the present study was the genetic characterization, sequencing and phylogenetic analysis of 18S rDNA sequences of Cryptosporidium isolates obtained from different animal hosts in Brazil. Fecal samples containing Cryptosporidium oocysts were obtained from chickens, ducks, quails, guinea pigs, dairy calves, dogs and cats. For amplification of 18S rDNA sequences the Secondary-PCR product of the extracted DNA from fecal suspension of each studied animal was utilized. The primary genetic characterization of Cryptosporidium sp. was performed using RFLP with the enzymes SspI and VspI. …

Looking inside an invasion wave of cells using continuum models: proliferation is the key.

Recently, a suite of cell migration assays were conducted to investigate the migration of neural crest (NC) cells along the gut during the development of the enteric nervous system (ENS). The NC cells colonise the gastro-intestinal tract as a rostro-caudal wave. Local behaviour was shown to be controlled by position relative to the leading edge of the wavefront. The assays involved chick-quail grafting techniques allowing the total invading population to be considered as a two-species system. A two-species continuum model with logistic proliferation and a migration mechanism is developed here …

The control of morphogen signalling: regulation of the synthesis and catabolism of retinoic acid in the developing embryo.

We consider here how morphogenetic signals involving retinoic acid (RA) are switched on and off in the light of positive and negative feedback controls which operate in other embryonic signalling systems. Switching on the RA signal involves the synthetic retinaldehyde dehydrogenase (RALDH) enzymes and it is currently thought that switching off the RA signal involves the CYP26 enzymes which catabolise RA. We have tested whether these enzymes are regulated by the presence or absence of all-trans-RA using the vitamin A-deficient quail model system and the application of excess retinoids on beads …

The avian embryo in fetal alcohol research.

The avian embryo has proven utility for studying ethanol's damaging effects upon the embryo. Chicken and quail are long-established models for developmental biology research; much of what we know regarding limb, craniofacial, neural crest, hindbrain, and cardiac morphogenesis was first established with avian models. These models also are for popular mechanistic studies of teratogens, including ethanol. Avian models have been used to explore ethanol's effects on neurogenesis, cardiogenesis, intracellular signaling, neurobehavior, and apoptosis. Presented here are several of these methodologies …

Cryo-SEM documentation of intact frozen-fractured avian eggs reveals species-dependent albumen freezing morphologies.

Cryo-scanning electron microscopy (cryo-SEM) was used to examine intact frozen-fractured eggs from five avian species-quail, hen, duck, turkey, and goose-spanning two taxonomic orders (Galliformes and Anseriformes) and a wide range of egg sizes. Whole eggs were frozen in liquid nitrogen within 24 h of laying, fractured at cryogenic temperature, and observed in the frozen-hydrated state after vacuum sublimation and gold coating. Multiple structural regions were examined, including yolk spheres, the albumen body, the shell-shell membrane-albumen interface, and the yolk-vitelline membrane-albumen…

Direct interaction and N-terminal phosphorylation of c-Jun by c-Mil/Raf.

c-Mil is the avian homologue of the mammalian serine/threonine kinase c-Raf-1. c-Mil/Raf is a mediator of signal transduction leading to gene expression via the c-Jun DNA-binding site, AP-1. Here we show that c-Mil immunopurified from MC29-virus-transformed quail fibroblasts phosphorylates c-Jun in vitro near its N terminus (Ser-63 and -73). Furthermore, the viral oncogene product Gag-Mil of the avian wild-type retrovirus MH2 phosphorylates c-Jun in vitro. A contribution by other known kinases phosphorylating c-Jun, such as the mitogen-activated protein kinases (MAPKs) and the c-Jun N-terminal…

An Early-Life Stage Alternative Testing Strategy for Assessing the Impacts of Environmental Chemicals in Birds.

Early-life stage (ELS) toxicity tests are recognized as an advancement over current testing methodologies in terms of cost, animal use, and biological relevance. However, standardized ELS tests are not presently available for some vertebrate taxa, including birds. The present study describes a Japanese quail (Coturnix japonica) ELS test that is a promising candidate for standardization and applies it to test 8 environmental chemicals (ethinylestradiol, benzo[a]pyrene, chlorpyrifos, fluoxetine, lead(II)nitrate, trenbolone, seleno-L-methionine, hexabromocyclododecane). Individual chemicals were …

[The epizootology and epidemiology of echinococcosis and paecilomycosis].

Fungi of the genus Paecilomyces, a causative agent of the fungal disease paecilomycosis, have been found in the blood of productive animals (sheep, cattle, pigs, goats) that are carriers of Echinococcus larvocysts. In the Republic of Uzbekistan, the echinococcosis affection of sheep, cattle, pigs, and goats continues to remain high. Domestic fowls (chickens, turkeys, and gooses) have been established to be blood carriers of Paecilomyces fungi, which is also true for wild birds, such as my-lady's-belts, quails, chukars, and crows. The degree of fungal affection has been also established for bir…

β-Lactam resistance genes present in UK pheasants and red-legged partridges.

BACKGROUND: Despite considerable recent reductions in antimicrobial use, the UK gamebird industry continues to struggle with production diseases during the rearing season, necessitating significant antibiotic use. This observational study investigated the presence of genes conferring resistance to β-lactam antibiotics within industry-reared pheasants and red-legged partridges in the UK. METHODS: DNA was extracted from 60 pooled caecal samples collected from gamebirds at routine postmortem examinations during the rearing season. Genes encoding extended-spectrum β-lactamases (ESBL) were detecte

Diencephalic origin of the pineal gland of the chicken embryo.

In the present paper, the diencephalic origin of the chick pineal gland was analyzed by a series of experiments: prosencephalic substitution; in vitro culture of isolated diencephalons; and total or partial excission of the diencephalic roof. The results indicate that the differentiation of the chick pineal gland in the rooof of the third ventricle is not influenced by the neighbouring brain vesicles and is of diencephalic origin. Moreover, in order to obtain chick embryos with pineal agenesia, the whole diencephalic roof has to be removed.

Biological activity of the three mono-beta-D-glucopyranosides of 1,25-dihydroxycholecalciferol.

The biological activity of the three mono-beta-D-glucopyranosides (mono-glucosides) of 1,25-dihydroxycholecalciferol [1,25(OH)2D3] synthesized chemically has been studied in chicks and Japanese quails. While the 1- and the 3-glucoside showed no or only little effect on serum calcium, bone weight, calcium binding protein (CaBP) or calcium deposition in the egg shell, the 25-glucoside was found to be more than half as active as the aglycone 1,25(OH)2D3. The bioactivity of this glucoside parallels a higher binding constant to the intestinal 1,25(OH)2D3 receptor compared to those of the two other …

Cellular interactions in the development of the olfactory system: an ablation and homotypic transplantation analysis.

In the current study, we addressed two questions: First, is the olfactory placode necessary for the development of the olfactory bulb and the entire telencephalon? Second, does the olfactory placode contribute cells to the olfactory bulb? We addressed these questions by unilaterally ablating the olfactory placode in chick embryos before an olfactory nerve was produced and, in a second series of experiments, by replacing the ablated chick olfactory placode with a quail olfactory placode. Our results indicate that the olfactory placode is critical for olfactory bulb development, but is not neces…

Development and validation of a quantitative method for 15 antiviral drugs in poultry muscle using liquid chromatography coupled to tandem mass spectrometry.

The objective of this work was to develop a quantitative multi-residue method for analysing antiviral drug residues and their metabolites in poultry meat samples. Antiviral drugs are not licensed for the treatment of influenza in food producing animals. However, there have been some reports indicating their illegal use in poultry. In this study, a method was developed for the analysis of 15 antiviral drug residues in poultry muscle (chicken, duck, quail and turkey) using liquid chromatography coupled to tandem mass spectrometry. This included 13 drugs against influenza and associated metabolit…

Hormone treatment of the adult transsexual patient.

Annual cycles in daylength provide an initial predictive environmental cue that plants and animals use to time seasonal biology. Seasonal changes in photoperiodic information acts to entrain endogenous programs in physiology to optimize an animal's fitness. Attempts to identify the neural and molecular substrates of photoperiodic time measurement in birds have, to date, focused on blunt changes in light exposure during a restricted period of photoinducibility. The objectives of these studies were first to characterize a molecular seasonal clock in Japanese quail and second, to identify the key…
