Parafibromin is a tumor suppressor protein encoded by HRPT2, a gene recently implicated in the hereditary hyperparathyroidism-jaw tumor syndrome, parathyroid cancer, and a subset of kindreds with familial isolated hyperparathyroidism. Human parafibromin binds to RNA polymerase II as part of a PAF1 transcriptional regulatory complex. The mechanism by which loss of parafibromin function can lead to neoplastic transformation is poorly understood. Because the subcellular localization of parafibromin is likely to be critical for its function with the nuclear PAF1 complex, we sought to experimentall…

The mushroom poisoning of phalloid type is caused by amatoxins, mainly by alpha-amanitine. It is a cytotoxin which acts through an indirect mechanism. It binds irreversibly the nucleolar enzyme, RNA-polymerase II in cells with intensive proteosynthesis (enterocytes in the digestive tract, hepatocytes in the liver and epithelial cells of proximal renal tubules). A tight binding of the toxin with the enzyme blocks the transcription from DNA to m-RNA and thus makes the proteosynthesis impossible. This results in the cell necrosis. Pathophysiology of the phalloid type poisoning is rather complicat

DNA polymerase epsilon (Pol ε), a component of the core replisome, is involved in DNA replication. Although genetic defects of Pol ε have been reported to cause immunodeficiency syndromes, its role in haematopoiesis remains unknown. Here, we identified compound heterozygous variants (p.[Asp1131fs];[Thr1891del]) in POLE, encoding Pol ε catalytic subunit A (POLE1), in siblings with a syndromic form of severe congenital transfusion-dependent anaemia. In contrast to Diamond-Blackfan anaemia, marked reticulocytopenia or marked erythroid hypoplasia was not found. Their bone marrow aspirates during i

Human cytomegalovirus (HCMV) primary infection, re-infection, and reactivation from latency cause morbidity in immune-compromised patients. Consequently, potential therapeutic strategies remain of interest for the treatment of infection. Naturally occurring triterpenoids derived from plants have been demonstrated to have anti-viral activity, although their precise mechanisms of action are not always fully understood. Here, we investigate the activity of Mormordin Ic (Mc) and demonstrate that it is potently anti-viral against HCMV. Through investigation of the mechanistic basis of this anti-vir

Human transcription factor IIH (TFIIH) is part of the general transcriptional machinery required by RNA polymerase II for the initiation of eukaryotic gene transcription. Composed of ten subunits that add up to a molecular mass of about 500 kDa, TFIIH is also essential for nucleotide excision repair. The seven-subunit TFIIH core complex formed by XPB, XPD, p62, p52, p44, p34, and p8 is competent for DNA repair, while the CDK-activating kinase subcomplex, which includes the kinase activity of CDK7 as well as the cyclin H and MAT1 subunits, is additionally required for transcription initiation. …

Mucopolysaccharidosis type II (Hunter syndrome) is an X-linked lysosomal storage disorder caused by a deficiency of the enzyme iduronate-2-sulfatase. We sequenced genomic DNA and RT-PCR products in the iduronate sulfatase (IDS) gene in 6 unrelated patients with Hunter syndrome to assess genotype/phenotype relationships and offer carrier testing where required. Six novel mutations were identified: four missense mutations, one four-base pair deletion (596-599delAACA) and a cryptic splice site mutation. Three of the missense mutations were significant amino acid substitutions (S143F, S491F, E341K

We report a clinically heterogeneous, multigenerational pedigree with the syndrome of mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) associated with a mutation at nucleotide 3243 in the mitochondrial DNA tRNA(Leu)(UUR) gene. Our findings suggest that the mutation at nucleotide 3243 is not always associated with the classic MELAS phenotype and that other symptoms (notably cardiac and gastrointestinal abnormalities) should raise the suspicion of a mitochondrial disorder.

By means of specific human relaxin primers that originated from relaxin A and B chains, a monoclonal antibody, and 125I-relaxin, the expression of mRNA and immunoreactive protein and the presence of binding sites for relaxin were investigated in human fallopian tubes. Reverse transcription polymerase chain reaction (RT-PCR) analysis of total RNA isolated from tubal tissues revealed the predicted 434-bp fragments originating from both the H1 and H2 relaxin genes. Restriction enzyme digestion of the RT-PCR products with Msp I (Hpa II), present only in the relaxin H1 sequence, resulted in the ant

Originally identified as a factor crucial for RNA polymerase (Pol) II transcriptional elongation of cellular genes, the P-TEFb kinase was subsequently shown to also serve as a specific host co-factor required for HIV-1 transcription. Recruited by either the bromodomain protein Brd4 to cellular promoters for general transcription or the HIV-1 Tat protein to the viral LTR for activated HIV-1 transcription, P-TEFb stimulates the processivity of Pol II through phosphorylating the C-terminal domain of Pol II and a pair of negative elongation factors, leading to the synthesis of full-length transcri

BACKGROUND: Hepatocellular carcinoma (HCC) develops in a chronically hypoxic microenvironment; however, the contribution of hypoxia-regulated long non-coding RNAs (lncRNAs) remains poorly defined.
AIM: We aimed to identify MIR100HG as a hypoxia-responsive regulator in hepatocellular carcinoma (HCC) and to define its transcriptional control by hypoxia-inducible factor 1 alpha.
METHODS: Basal MIR100HG expression was analyzed in four hepatocellular carcinoma cell lines (Hep3B, Huh-7, SNU-398, and SNU-475) and non-tumorigenic Clone-9 hepatocytes. Hypoxia was induced by cobalt (II) chloride. Prol

BACKGROUND: Cyclin-dependent kinase 9 (CDK9) is a central regulator of RNA polymerase II elongation and has emerged as a therapeutic target in tumors characterized by transcriptional addiction. Growing interest in selective inhibitors and targeted degraders has renewed attention to the translational potential of CDK9-directed therapy.
AREAS COVERED: This review summarizes the molecular functions of the CDK9/positive transcription elongation factor b (P-TEFb) axis, its role in super-enhancer-driven oncogenic programs, and the mechanisms by which CDK9 inhibition promotes apoptosis, epigenetic d…

Transcription-replication conflicts (TRCs) drive genomic instability and are linked to tumorigenesis and treatment response. Replication stress exacerbates TRCs, but how cells coordinate transcription and replication to limit these conflicts remains unclear. Here, we identify NONO as a key regulator that maintains transcription-replication balance via BRD4 under replication stress. Replication stress triggers NONO acetylation at K243, enhancing BRD4 binding. On single-stranded DNA, NONO-BRD4 form condensates that block paused RNA polymerase II (RNAP2) release, fine-tuning elongation. NONO loss…

The tandem BRCT domains in the SMC5/6 complex localization factor 1 (SLF1, SLF1tBRCT) mediate interactions with a phosphorylated region in Rad18 and interaction targets the SMC5/6 complex to DNA damage sites for repair. Whether SLF1tBRCT mediate interactions with additional proteins is unclear. In this study, we searched for novel SLF1tBRCT binding partners and found that RNA polymerase II subunit 2 (RPB2) may contain a SLF1tBRCT binding site. The crystal structure of SLF1tBRCT in complex with a RPB2 peptide containing the predicted binding site revealed a SLF1tBRCT-RPB2 interface that bares s…

Activation-induced deaminase (AID) mutates immunoglobulin genes to initiate antibody diversification by class switch recombination and somat … gh DOT1L activity. In the absence of DOT1L, nascent transcription largely increases despite reduced RNA Polymerase II (RNAPII) occupancy. Integrative genomic analyses reveal that DOT1L locally restricts transcription elongation velocity proportionally to H3K79me2/3 levels and extends RNAPII pausing. These transcriptional conditions enhance AID occupancy and thereby …

This study investigates the transcriptional impact of Q901, a highly selective cyclin-dependent kinase 7 (CDK7) inhibitor in clinical development. Q901 primarily disrupted MYC and E2F-dependent transcription programs, down-regulating cell cycle control and DNA damage repair pathways. CDK7 binding at the promoter-proximal regions was markedly stabilized by Q901, leading to reduced occupancy of MYC, E2F, and RNA polymerase II (RNAPII). These findings offered a novel therapeutic strategy to enhance cancer susceptibility to topoisomerase I (TOP1) DNA-protein cross-links (TOP1-DPCs) induced by TOP1…

Oncolytic virotherapy has shown promise for various cancers, but its application in osteosarcoma (OS) remains underexplored. This study provides evidence that M1, a natural Getah-like alphavirus, exerts oncolytic activity against OS. We demonstrate that OS cells exhibit heightened sensitivity to M1 infection, with its anti-tumor effects not solely dependent on the canonical ER stress-induced apoptosis. Proteomic and ChIP-seq analyses show the DNA-directed RNA polymerase II subunit RPB1 contributes to oncogenic super-enhancer activity and serves as a direct target of M1-mediated regulation. The…
