Shogaol triggers apoptosis and ferroptosis in Ewing sarcoma by targeting the polo-like kinase 1/nucleophosmin 1 axis.

Ewing sarcoma is an aggressive pediatric bone cancer with poor prognosis, highlighting the urgent need for novel therapeutics. This study demonstrates the potent anti-tumor activity of shogaol, a bioactive compound from ginger, against Ewing sarcoma, using both in vitro and in vivo model systems. In vitro, shogaol significantly inhibited cell proliferation and induced both apoptosis and ferroptosis in Ewing sarcoma cells, as evidenced by increased lipid reactive oxygen species, Malondialdehyde, and Fe2+ levels, as well as altered expression of BAX, BCL2, GPX4, and SLC3A2. In vivo, shogaol sign

Olfactory Groove Meningioma Detected 10 Months after Cranial Radiotherapy in a Metastatic Ewing Sarcoma Survivor: A Case Report.

INTRODUCTION: Secondary intracranial neoplasms are a well-recognized late complication of cranial radiotherapy. Among these, meningiomas are the most frequently reported radiation-associated tumors. However, olfactory groove meningiomas (OGMs) are uncommon, and their development after cranial irradiation with a very short latency remains exceptionally rare. CASE PRESENTATION: A 38-year-old woman with Ewing sarcoma of the right thigh and brain metastases underwent multimodal treatment of surgical resection, VDC-IE chemotherapy, and subsequent cranial radiotherapy (54 Gy in 27 fractions). Retro

Ewing sarcoma of the first metacarpal: a rare case report with thumb-sparing resection and fibular graft reconstruction.

BACKGROUND: Ewing sarcoma is a malignant primary bone tumor that predominantly affects the long bones and pelvis of children and adolescents. Involvement of the hand is exceptionally rare, particularly at the level of the first metacarpal. When the dominant thumb is affected, treatment becomes especially challenging due to the critical functional role of this structure. CASE PRESENTATION: We report the case of an 11-year-old right-handed boy who presented with a painful swelling of the right thumb. Imaging revealed an aggressive osteolytic lesion of the first metacarpal with soft tissue exten

A Rare Case of Mid-clavicular Ewing Sarcoma Treated With Total Claviculectomy: A Case Report and Literature Review.

Ewing sarcoma of the clavicle is an extremely rare tumor. The currently available evidence is limited to a small number of case reports. We present a case of a 13-year-old female patient referred to our tertiary care center with a progressively growing lump over the mid-clavicular, mystically diagnosed as a benign lipoma in a rural hospital. Radiological investigations and histopathological examination confirmed the diagnosis of metastatic Ewing sarcoma of the mid-clavicle. We outlined the management plan done for this patient followed by a description of the surgical technique of the total cl

Strategies of the management of pulmonary metastases in children with Ewing sarcoma- single institution experience of 137 patients.

BACKGROUND: The role of pulmonary metastasectomy in pediatric Ewing sarcoma remains controversial, with significant treatment selection bias complicating the interpretation of retrospective series. OBJECTIVE: To evaluate treatment outcomes in isolated pulmonary metastases (PM) using propensity score analysis to control for selection bias. METHODS: Retrospective analysis of 137 patients with pulmonary metastases (85 isolated PM, 52 PM with extrapulmonary metastases) treated between 1998 and 2024. Primary comparison focused on thoracotomy alone versus radiotherapy alone in isolated PM patients

N'-(1-phenylethylidene)-benzohydrazide cytotoxicity is LSD1 independent and linked to Fe-S cluster disruption in Ewing sarcoma.

The noncompetitive LSD1 inhibitors SP-2509 and SP-2577 are N'-(1-phenylethylidene)-benzohydrazides that display potent activity in Ewing sarcoma. They block transcriptional regulation of the causative oncogenic fusion protein, EWSR1::FLI1, and cause cell death. However, SP-2509 and SP-2577 are the only LSD1 inhibitors active in Ewing sarcoma; other LSD1 inhibitors have little effect. Studies from our group and others suggest SP-2509 activity may result from off-target activity affecting the mitochondria. Here we identified potential off-target mechanisms of N'-(1-phenylethylidene)-benzohydrazi

High-dose chemotherapy with autologous stem cell rescue in children and young adults with high-risk Ewing sarcoma.

BACKGROUND: A combination treatment of surgery, chemotherapy, and radiotherapy can improve the survivals of pediatric patients with Ewing sarcoma (ES). However, prognosis remains poor for patients with metastatic disease at diagnosis or recurrence. Other high-risk (HR) features include large tumor burden, tumors of the axial skeleton, and poor histologic response. Several studies have documented high-dose chemotherapy with autologous stem cell rescue (HDC-ASCR) to be effective in such patients. In this retrospective study, we present the results of HDC-ASCR for high-risk ES (HRES) in children

Chronic abdominal aortic occlusion in a patient with an underdeveloped, irradiated pelvis after childhood treatment of Ewing Sarcoma.

Chronic radiation-induced arterial injury is generally predictable by known tumor types and anatomic location. We present the first case of radiation-induced chronic aortic occlusion associated with a small pelvis secondary to the treatment of childhood Ewing sarcoma. The patient presented with profound claudication and accelerated atherosclerosis obliterans of the aortoiliac system and failed endovascular treatment. Successful aortic reconstruction was performed. This case highlights the long-term effects of chemoradiation to the aortoiliac segment and pelvic bones in a child, and the technic

Two-Phase Deep Learning Algorithm for Detection and Differentiation of Ewing Sarcoma and Acute Osteomyelitis in Paediatric Radiographs.

BACKGROUND/AIM: Ewing sarcoma is a highly malignant tumour predominantly found in children. The radiological signs of this malignancy can be mistaken for acute osteomyelitis. These entities require profoundly different treatments and result in completely different prognoses. The purpose of this study was to develop an artificial intelligence algorithm, which can determine imaging features in a common radiograph to distinguish osteomyelitis from Ewing sarcoma. MATERIALS AND METHODS: A total of 182 radiographs from our Sarcoma Centre (118 healthy, 44 Ewing, 20 osteomyelitis) from 58 different p

Clinicopathologic and Molecular Cytogenetic Analysis of 8 Cases With Uterine Cervical Ewing Sarcoma: Case Series With Literature Review.

Ewing sarcoma (ES) is a highly malignant tumor that rarely occurs in the uterine cervix. Herein, we report 8 cases with ES arising primarily in the uterine cervix by focusing on clinicopathologic and molecular cytogenetic features and differential diagnoses. Eight cases of cervical ES were diagnosed between February, 2012, and September, 2018. The age of patients ranged from 13 to 47 years. Abnormal vaginal bleeding and lower abdominal pain were the most common symptoms. Histologically, the tumor was composed of uniform, round, and oval cells with a narrow rim of eosinophilic cytoplasm. Primary, Dural-Based, Ewing Sarcoma Manifesting with Seizure Activity: Presentation of a Rare Tumor Entity with Literature Review.

BACKGROUND: Primary Ewing sarcoma arising from the calvarial bone and/or underlying dura matter are relatively rarely reported in the literature. Even rarer are considered to be those that originate from the dura and proliferate in both directions, toward the brain parenchyma and through the dura invading the overlying bone. CASE DESCRIPTION: We report the case of a girl aged 11 years who presented with an ictus of generalized tonic-clonic seizures, without known focal neurologic deficit. Neuroradiologic work-up verified the existence of a tumor in the right fronto-parietal area of the brain,

SRC Family Kinase Inhibition in Ewing Sarcoma Cells Induces p38 MAP Kinase-Mediated Cytotoxicity and Reduces Cell Migration.

Ewing sarcoma (ES) is a highly aggressive bone and soft tissue cancer, representing the second most common primary malignant bone tumor in children and adolescents. Although the development of a multimodal therapy, including both local control (surgery and/or radiation) and systemic multidrug chemotherapy, has determined a significant improvement in survival, patients with metastatic and recurrent disease still face a poor prognosis. Moreover, considering that ES primarily affects young patients, there are concerns about long-term adverse effects of the therapy. Therefore, more rational strate

Unusual presentation of Ewing sarcoma in the adrenal gland: a secondary malignancy from a survivor of Burkitt lymphoma.

The occurrence of Ewing sarcoma as a secondary malignancy is an extremely rare event in long-term cancer survivors. In addition, the occurrence of Ewing sarcoma in the adrenal gland is highly unusual. In this case report, we treated a 20-year-old male patient with cyclophosphamide, doxorubicin, vincristine, dexamethasone, and methotrexate and cytarabine chemotherapy following a diagnosis of Stage IV Burkitt lymphoma. Following complete remission, he had been maintained for 2 years without evidence of disease. However, a regular follow-up computed tomography scan found a left adrenal gland mass

Untargeted Metabolite Screening of Ewing Sarcoma Cells and Tissues with Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS).

Altered metabolism is a hallmark of cancer, where rewiring of cellular metabolic pathways is essential for tumor initiation, support of rapid proliferation, and adaptation to new metastatic sites. Metabolomics has emerged as a powerful tool to comprehensively characterize the metabolome of cancers and holds promise to transform understanding of how oncogenes can drive tumorigenesis and metastasis, identify cancer-specific metabolic dependencies, and provide novel therapeutic opportunities. This chapter describes methods to extract metabolites from Ewing sarcoma (EwS) cells grown in 2D or 3D cu

Combined inhibition of CDK4/6 and PI3K pathways exhibit highly synergistic activity and translational potential in Ewing sarcoma.

Ewing sarcoma is a highly aggressive solid malignancy affecting children and young adults. Ewing sarcoma is driven primarily by EWSR1::FLI1, a fusion oncoprotein that has been notoriously difficult to target with traditional pharmacologic agents. There are numerous examples of promising preclinical combinations of small molecules that are never tested in pediatric clinical trials because agents fail to reach the market due to limited efficacy for common adult cancers. Moreover, the effectiveness of single-agent therapies for cancer treatment is often limited. To address these limitations, we s

Genome-wide association study meta-analysis identifies susceptibility loci informing Ewing sarcoma etiology and potential mechanisms of risk.

Ewing sarcoma (EwS) is a rare, aggressive pediatric malignancy driven by FET::ETS family fusions (EWSR1::FLI1 in >85% of cases) with no established environmental risk factors. To investigate germline predisposition, we analyzed 2,014 EwS cases and 10,525 cancer-free controls in a two-stage analysis that combined an international genome-wide association study and a case parent trio study. The combined meta-analysis identified 18 variants at 14 susceptibility loci (9 novel, 5 replicated) with moderate effect sizes (odds ratios≥1.25). Integrative analyses of the EwS loci revealed enrichment of ex

Preoperative Radiotherapy in Patients With Localized Ewing Sarcoma Enrolled on AEWS1031: A Report From the Children's Oncology Group.

Patients with resectable Ewing sarcoma (ES) in select sites, such as the pelvis or chest wall, have a higher risk of positive microscopic margins following surgery. AEWS1031 was the first North American/Australasia ES cooperative group trial allowing standardized preoperative radiotherapy, intended to minimize the morbidity of combined modality local tumor control as well as improve the likelihood of clear surgical margins. In this study, we found that the use of low-dose preoperative radiation to a smaller target volume resulted in a high rate of R0 resection. The robust pathologic response r
